    Notizen: Abstract Records of 92 children were reviewed who, in the first 18 months of life had an intracranial tumour, and who were registered with the Manchester Children's Tumour Registry during the 34-year period from 1953 to 1987. The incidence of intracranial tumour was 1 in 25000 live births, and 85% of the tumours were malignant. The introduction of computed tomography (CT) in the region in 1976 improved the diagnostic accuracy and led to early diagnosis. This also coincided with refined radiotherapy and improved chemotherapy. The surgical attitude became more aggressive, and with early treatment, mortality was reduced, though the 1-month operative mortality still remains high at 15%. All survivors were available for follow-up. All children with axial tumours died within 9 years. Long-term follow-up did not suggest significant improvement in the overall survival or in the quality of life, though there was a significant improvement in the 5-year survival rate with modern management.

    Notizen: Summary The salt-tolerant cultivated tomato (Lycopersicon esculentum) accession, ‘PI174263’, and a sensitive cv, ‘UCT5’, were crossed to develop reciprocal F1, F2 and BC1 populations for genetic analysis of salt tolerance in tomatoes during seed germination. Variation was partitioned into embryo, endosperm and maternal (testa and cytoplasmic) components. Generation means analysis indicated that there were no significant embryo (additive, dominance or epistatic) effects on germination performance under salt stress. Highly significant endosperm additive and testa dominance effects were detected. The proportion of the total variance explained by the model containing these two components was R2=98.2%. Variance component analysis indicated a large genetic variance with additive gene action as the predominant component. Furhter inspection indicated that this variance was attributable to endosperm additive effects on germinability under salt stress. Narrow-sense heritability was estimated as moderately high. Implications for breeding procedures are discussed.

    Notizen: Abstract The germination responsiveness of an F2 population derived from the cross Lycopersicon esculentum (UCT5) x L. pennellii (LA716) was evaluated for salt tolerance at two stress levels, 150 mM NaCl + 15 mM CaCl2 and 200 mM NaCl + 20 mM CaCl2. Individuals were selected at both tails of the response distribution. The salt-tolerant and salt-sensitive individuals were genotyped at 16 isozyme loci located on 9 of the 12 tomato chromosomes. In addition, an unselected (control) F2 population was genotyped at the same marker loci, and gene frequencies were estimated in both selected and unselected populations. Trait-based marker analysis was effective in identifying genomic locations (quantitative trait loci, QTLs) affecting salt tolerance in the tomato. Three genomic locations marked by Est-3 on chromosome 1, Prx-7 on chromosome 3, and 6Pgdh-2 and Pgi-1 on chromosome 12 showed significant positive effects, while 2 locations associated with Got-2 on chromosome 7 and Aps-2 on chromosome 8 showed significant negative effects. The identification of genomic locations with both positive and negative effects on this trait suggests the likelihood of recovering transgressive segregants in progeny derived from these parental lines. Similar genomic locations were identified when selection was made either for salt tolerance or salt sensitivity and at both salt-stress treatments. Comparable results were obtained in uni- and bidirectional selection experiments. However, when marker allele gene frequencies in a control population are unknown, bidirectional selection may be more efficient than unidirectional selection in identifying marker-QTL associations. Results from this study are discussed in relationship to the use of molecular markers in developing salt-tolerant tomatoes.

    Notizen: Summary A technique is described by which families of potato seedlings can be rapidly screened for resistance to potato leaf roll virus (PLRV) in the greenhouse with a minimum expenditure of labour and bench space. Seed of different families was sown in rows in four replicated blocks and the seedlings produced were exposed to viruliferousMyzus persicae when very young. Symptoms resembling those typical of secondary infection with PLRV developed within a few weks and families with few affected seedlings compared with the susceptible control were classed as resistant. Tests done at different times with similar sets of families gave similar results. Applying infectiveM. persicae by shaking heavily colonised potato shoots above the seedlings was more effective and far less time consuming than transferring them individually by hand using a fine paint brush.

    Notizen: Abstract This study was conducted to identify genomic regions (quantitative trait loci, QTLs) affecting salt tolerance during germination in tomato. Germination response of an F2 population of a cross between UCT5 (Lycopersicon esculentum, salt-sensitive) and LA716 (L. pennellii, salt-tolerant) was evaluated at a salt-stress level of 175 mM NaCl + 17.5 mM CaCl2 (water potential ca. −950 kPa). Germination was scored visually as radicle protrusion at 6 h intervals for 30 consecutive days. Individuals at both extremes of the response distribution (i.e., salt-tolerant and salt-sensitive individuals) were selected. The selected individuals were genotyped at 84 genetic markers including 16 isozymes and 68 restriction fragment length polymorphisms (RFLPs). Trait-based marker analysis (TBA) which measures changes (differences) in marker allele frequencies in selected lines was used to identify marker-linked QTLs. Eight genomic regions were identified on seven tomato chromosomes bearing genes (QTLs) with significant effects on this trait. The results confirmed our previous suggestion that salt tolerance during germination in tomato is polygenically controlled. The salt-tolerant parent contributed favorable QTL alleles on chromosomes 1, 3, 9 and 12 whereas the salt sensitive parent contributed favorable QTL alleles on chromosomes 2, 7 and 8. The identification of favorable alleles in both parents suggests the likelihood of recovering transgressive segregants in progeny derived from these parental genotypes. The results can be used for marker-assisted selection and breeding of salt-tolerant tomatoes.

    Notizen: Summary The potential to improve seed germination responses to salinity was evaluated for 13 accessions representing six wild Lycopersicon species and 20 accessions of L. esculentum. Germination response times increased in all accessions at 100 mM NaCl. Analysis indicated that one accession of L. peruvianum (PI126435) germinated faster under high salinity than all other accessions and was closely followed by L. pennellii (LA716). The fastest germinating L. esculentum accession, PI174263, ranked third. Additional wild ecotypes exhibiting rapid germination at 100 mM NaCl were identified among L. pimpinellifolium and L. peruvianum.

    Notizen: Summary In studies to determine the inheritance of response-time traits, such as time to seed germination, some viable individuals may fail to respond during an experiment. If these right-censored observations are ignored, sample means and variances will be underestimated. This is illustrated using data from time to seed germination at 9°C for Lycopersicon esculentum (Mill.) fast germinating PI 120256, slow-germinating T3 and their reciprocal F1, F2 and backcross progeny. This paper presents methods to detect and to accommodate right-censored data in generation means analysis. Genetic interpretations derived from corrected and uncorrected estimates of generation means and variances are compared. Correction for right-censoring increased estimates of environmental and phenotypic variances, and decreased heritability estimates.

    Notizen: Summary Isozyme phenotypes at four known genetic loci were determined in parental inbreds and corresponding F1 hybrid seed lots of four commercial broccoli, two cauliflower, and two cabbage varieties to determine seed purity. Most inbred lines were completely homozygous at all four loci but differed with respect to alleles at one to three loci. Several parental inbreds of the cabbage hybrids were segregating at two to three of the loci. Models were developed to estimate seed purity in cases where parents were either fixed or segregating at diagnostic loci. Estimates of contamination ranged from 1.5 to 40.1%. These estimates were comparable with those from commercial grow-outs with a tendency for the former estimates to be higher. It was concluded that more stable SI alleles or genetic male sterility should be used to reduce contamination. Electrophoretic variation was further discussed as a tool for selecting homozygous plants and for strong self-incompatibility.
