Notes: Abstract Determination of alkanes in breath of laboratory animals and humans has become a standard-method for monitoring lipid peroxidation in vivo. Isothermal gas chromatography on Porasil C enables sensitive, rapid and repetitive determination of all C2-C5-hydrocarbons in breath. Volatile toxicants and metabolites, which would coelute with the alkanes of later injected samples, are deviated by using a precolumn. An automatic switching unit controls withdrawal and injection of samples and backflush of the precolumn in a repetitive manner at fixed intervals. This increases accuracy and sensitivity of analysis and enables virtually unattended operation. The system has been applied for a study on the oxygen-dependance of CCl4-metabolism in the rat.

Notes: An enantioselective gas chromatographic method has been developed and validated for the determination of the plasma concentration of the enantiomers of the anticancer drug ifosfamide (IFF). In this approach, the IFF enantiomers are separated from the plasma matrix by solid phase extraction, chromatographically resolved by gas chromatography on a chiral stationary phase, and detected by mass selective detection using selective ion monitoring. The assay has been validated for routine clinical and pharmacokinetic use and has a limit of detection in plasma of 250 ng/ml of each isomer.

Notes: Glass capillaries coated with Chirasil-Val, a chirally functionalised polysiloxane, are capable in principle of resolving all protein amino-acid enantiomers in a single run and within a short analysis time, thus allowing for example the quantitative amino acid determination by enantiomer labelling. The elution characteristics of the individual amino acids however are also dependent upon the chemical nature of the capillary wall surface, and a surface pretreatment is found to be necessary if all protein amino acids are to be analysed. Of the various methods of pretreatment tested, etching of borosilicate glass with gaseous HCl followed by deposition of colloidal silicic acid is considered to be the most suitable.

Notes: Trichloroacetic acid (TCA) is an atmospheric oxidation product of airborne C2-chlorocarbons and a widespread air pollutant. It was widely used as a herbicide in the fifties and sixties, in some countries, still today. It occurs in the foliage of conifers and deciduous trees of mountain forests in Germany, e. g. the Black Forest, the Northern Alps or the Erzgebirge. Quantitative determination in the ppb-range is achieved by gas chromatography using thick film capillary columns and electron-capture detection. The levels in spruce needles at two forest sites correlate with the extent of needle loss. The data are supportive of the hypothesis that TCA is causally involved in the induction of mountain forest decline.

Notes: Haloacetates in various environmental compartments can be determined by gas chromatography-negative-ion chemical-ionization mass spectrometry after derivatization with 1-pentafluorophenyl diazoethane. Detection limits in absolute amounts per injection are between 0.01 fg (chlorodifluoroacetate) and 80 fg (monofluoroacetate). Sampling of haloacetates in urban air was performed by means of cylindrical denuders coated with alkalized glycerol. The haloacetates detected are trifluoro-, monochloro-, dichloro-, trichloro-, monobromo-, and dibromoacetate. The concentrations in ambient air fluctuate strongly, e.g. between 20 and 3000 pg/m3 for TFA. Haloacetates are also found in river waters and tree foliage. A major problem is interference from contamination with trifluoro- and trichloroacetate.
