    Notes: Abstract: The adenylyl cyclase-cyclic AMP (cAMP) second messenger pathway has been proposed to regulate myelin gene expression; however, a clear correlation between endogenous cAMP levels and myelin-specific mRNA levels has never been demonstrated during the induction or maintenance of differentiation by the myelinating Schwann cell. Endogenous cAMP levels decreased to 8–10% of normal nerve by 3 days after crush or permanent transection injury of adult rat sciatic nerve. Whereas levels remained low after transection injury, cAMP levels reached only 27% of the normal values by 35 days after crush injury. Because P0 mRNA levels were 60% of normal levels by 14 days and 100% by 21 days after crush injury, cAMP increased only well after P0 gene induction. cAMP, therefore, does not appear to trigger myelin gene induction but may be involved in myelin assembly or maintenance. Forskolin, an activator of adenylyl cyclase, increased endoneurial cAMP levels only in the normal nerve, and in the crushed nerve beginning at 16 days after injury, but at no time in the transected nerve. Only by treating transected nerve with 3-isobutyl-1-methylxanthine (IBMX), an inhibitor of cAMP phosphodiesterases, in combination with forskolin was it possible to increase cAMP levels. No induction of myelin genes, however, was observed with short- or long-term treatment with IBMX and forskolin in the transected nerve. A three-fold increase in phosphodiesterase activity was observed at 35 days after both injuries, and a nonmyelinated nerve was shown to have even higher activity. These experiments, therefore, suggest an important role for phosphodiesterase in the inactivation of this second messenger-dependent stimuli when Schwann cells are non-myelinating, such as after sciatic nerve injury or in the nonmyelinated nerve, which again implies that cAMP may be required for the maintenance of the myelin sheath.

    Notes: An efficient sparse-matrix-based numerical method is constructed to simulate two-dimensional nuclear magnetic resonance spectra of many-spin systems including the effects of chemical exchange and/or relaxation. The method employs efficient numerical time propagation requiring O(22n) operations in the case of an n spin 1/2 system. Pulses are treated with a fast implementation algorithm achieving O(n22n) scaling (case of n spins 1/2). The method is tested in simulations of double-quantum-filter correlation spectroscopy and exchange spectroscopy experiments on five- and seven-spin systems with two sites. Observed scaling is consistent with the analytic predictions. © 2000 American Institute of Physics.

    Notes: Objective To establish the lowest dose of cyclical dydrogesterone that protects against endometrial hyperplasia induced by continuous 2 mg 17β oestradiol, and to study the dose effect on vaginal bleeding and side effects.Design Double-blind, prospectively randomised dose-ranging study.Setting Menopause clinics in the UK and The Netherlands.Subjects Three hundred and seventy-one postmenopausal women with intact uteri, aged 40 to 60.Interventions Administration of six 28-day treatment cycles of continuous daily micronised 178 oestradiol with a randomly allocated dose of 5 to 20 mg of dydrogesterone added for the last 14 days of each.Main outcome measures Histological assessment of adequate progestational endometrial response, bleeding patterns and adverse effects.Results The study was completed by 320 subjects (86 %). Endometrial transformation occurred in over 94 % of those talung 5 mg of dydrogesterone, and in over 97 % of those on hgher doses, without significant differences between the 10, 15 and 20 mg groups. Acceptable bleeding patterns were found at all doses, with the incidence of withdrawal bleeding rising with increasing dose. The day of onset of bleeding was predictable from cycle to cycle, and occurred later in the 20 mg group than in the others. The incidence of noncyclic bleeding was about 6 % at all doses. Withdrawal occurred in 3-3% due to unacceptable bleeding and in 5-4% due to side effects. There was no relation with dose.Conclusions A dydrogesterone-17p oestradiol combination hormone replacement therapy confers endometrial protection with an acceptable bleeding pattern and few side effects. At least 10 mg of dydrogesterone for 14 days is required for acceptable endometrial protection.

    Notes: A semiclassical estimate of exponentially decaying tails of energy eigenstates is used to tailor grids in configuration space for specific quantum molecular dynamics computations. The estimate is demonstrated to accurately describe the eigenstate tails of constrained (T-shaped) rare gas triatoms, permitting construction of highly efficient basis sets for accurate numerical simulation. The algorithm is designed to demand minimal exploration of configuration space, and to provide considerable savings in computation time in the case of many degrees of freedom. © 1999 American Institute of Physics.

    Notes: Monte Carlo simulation of argon cluster evaporation is in reasonable accord with molecular dynamics computations. The new Monte Carlo algorithm is based on assumptions of ergodicity and mixing. Our results affirm these statistical assumptions for argon cluster dynamics. Observations of algebraic long time decay in evaporation lifetime distributions are also in support of the assumptions. The Monte Carlo algorithm provides an easily implemented statistical approximation to decay dynamics in many degree of freedom systems. It is formulated for application to any sequential unimolecular decay process. © 1995 American Institute of Physics.

    Notes: Expansion of argon clusters in a vacuum is simulated via molecular dynamics computations. The resulting evaporation dynamics is investigated with observations of temperature and pV energy loss curves. Observed cooling curves (T vs n) and collapse curves (pV/n vs n) are found to depend on final cluster size but not the initial cluster ensemble. The evaporation mechanism consists of an initial rapid cooling-and-collapse stage of a preliquid dense-gas-like cluster, followed by "equilibrium'' evaporation, and then another cooling stage of the resulting relatively incompressible liquidlike subcluster. Elements of this dynamics evaporation mechanism are tested by examination of finite n phase diagrams constructed using Metropolis Monte Carlo simulations of the fixed T and p ensemble. © 1995 American Institute of Physics.

    Notes: Several empirical studies have estimated the value of agricultural land as open space to local residents. An important goup of individuals that may be affected by the loss of agricultural land are visitors to a region. The value of ranchland to tourists visiting a resort town in the Rocky Mountains is estimated through a travel cost model that combines information on observed behavior data from actual trips with contingent behavior data on intended current visitation if the resource were converted to urban and resort uses. The value of ranch open space to tourists is the gain or loss in consumer surplus derived from a visit to the study area attributable to the resource. A random effects Poisson regression model is estimated because of the panel nature of the data, accounting for the correlation of the multiple responses from heterogeneous individuals. Twenty-five percent of the sample would reduce visitation and 23 percent of the sample would increase visitation if ranch open space were converted to urban and resort uses. The overall effect of converting ranch open space to resort and urban uses is no net change in average consumer surplus per trip for summer tourists in general.

    Notes: Individual differences in electrodermal lability have been related to performance in vigilance and reaction time tasks. The goal of the present study was to employ an “additive factors’ approach to identify the stages of information processing that might underlie these effects. Nineteen labile and 17 stabile subjects performed a choice reaction time task in which a visual imperative stimulus was presented under two conditions of intensity (presumed to affect the speed of pre-processing operations) X three conditions of degradation (which influences later encoding processes related to feature extraction). Measures of both reaction time and movement time were obtained. The major findings were: (a) labile subjects had faster reaction times than stabiles, and (b) lability interacted significantly with stimulus degradation. Labiles also tended (p〈.10) to have faster movement times. This pattern indicates that labiles and stabiles differ in the performance of later encoding operations, and possibly in the speed of motor processes as well. However, they do not appear to differ in the early pre-processing of the simple physical attributes of a stimulus.

    Notes: Summary. Fifty pregnancies occurred in 37 women with hyperprolactinaemia. Thirty women (81%) initially attended with infertility and 24 (65%) had secondary amenorrhoea. Twelve women (31%) had pituitary macroadenomas, of whom seven (19%) were treated with neurosurgery and/or external radiotherapy before conception. Fortyone conceptions (82%) followed treatment with bromocriptine. There were no unusual obstetric or endocrinological complications. Scrum prolactin measured at least 6 months post partum when under basal conditions, without bromocriptine and not breast feeding, showed no significant trends compared with initial values. Tumour growth was not stimulated by the high oestrogen levels of pregnancy.
