    Notes: The single alpha-tubulin gene of Tetrahymena thermophila was isolated from a genomic library and shown to encode a single protein. Comparisons of the rates of evolution of this gene with other alpha-tubulin sequences revealed that it belongs to a group of more evolutionarily constrained alpha-tubulin proteins in animals, plants, and protozoans versus the group of more rapidly evolving fungal and variant animal alpha-tubulins. The single alpha-tubulin of Tetrahymena must be used in a variety of microtubule structures, and we suggest that equivalently conserved alpha-tubulins in other organisms are evolutionarily constrained because they, too, are multifunctional. Reduced constraints on fungal tubulins are consistent with their simpler microtubule systems. The animal variant alpha-tubulins may also have diverged because of fewer functional requirements or they could be examples of specialized tubulins. To analyze the role of tubulin gene expression in regulation of the complex microtubule system of Tetrahymena, alpha-tubulin mRNA amounts were examined in a number of cell states. Message levels increased in growing versus starved cells and also during early stages of conjugation. These changes were correlated with increases in transcription rates. Additionally, alpha-tubulin mRNA levels oscillate in a cell cycle dependent fashion caused by changes in both transcription and decay rates. Therefore, as in other organisms, Tetrahymena adjusts alpha-tubulin message amounts via message decay. However the complex control of alpha-tubulin mRNA during the Tetrahymena life cycle involves regulation of both decay and transcription rates. © 1994 Wiley-Liss, Inc.

    Notes: We have constructed a chimeric β-tubulin gene that places the structural gene for the tubC β-tubulin of Axpergillus nidulans under the control of the benA β-tubulin promoter. Introduction of cither this chimeric gene or a second wild-type ben.A gene into a benomyl-resistant benA22 strain causes it to become benomyl sensitive, indicating that the introduced genes are functional. Analysis of the tubulin proteins synthesized in benA22 strains into which a second wild-type benA β-tubulin gene was transformed showed that the total amount of β-tubulin protein was the same as in the parental strain with a single benA gene. Thus the level of β-tubulin must be regulated. This was also true of transformants carrying an extra copy of the chimeric β-tubulin gene. The total amount of β-tubulin was the same as in the parental strain. Two-dimensional gel analysis showed that the endoge-nous benA22 and the introduced chimeric tubC gene contributed equally to the total β-tubulin pool. Th; fact that one-half of the benA β-tubulin could be replaced by tubC β-tubulin with no effect on the growth of the cells suggests that the benA and tubC β-tubulins are functionally interchangeable.

    Notes: Activation of T-lymphocytes by antigen, mitogenic lectins, or antibodies against the T-cell receptor complex, particularly in the presence of IL1, induces the secretion of the T-cell growth factor IL2. IL2 then has a major role in regulating the duration and magnitude of the immune response. Interaction of antigen, antibodies against the T-cell receptor complex, or mitogenic lectins with T-lymphocytes also induces hydrolysis of membrane phospholipids, leading to the production of diacylglycerol, an activator of the Ca2+- and phospholipid-dependent phosphotransferase, protein kinase C (PKC). Phorbol esters, potent activators of PKC, augment secretion of the T-cell growth factor, interleukin 2 (IL2). Activation of PKC may therefore serve as an important early event in the production and secretion of IL2. We have determined whether IL2 secretion can be induced in the murine cell T-lymphocyte line LBRM 331A5, where PKC is inhibited by staurosporine or sphingosine or in cells where PKC is depleted by prolonged incubation with high concentrations of phorbol esters. In cells in which PKC was either inhibited or depleted, antibodies against the T3 portion of the T-cell receptor complex and the mitogenic lectin phytohemagglutinin (PHA) still triggered IL2 secretion. In addition, the monokine Ill augmented this IL2 secretion irrespective of whether PKC was inhibited or depleted. These data indicate that activation of PKC is not an obligatory step for IL2 secretion in LBRM 331A5 murine T-lymphocytes.

    Notes: NMR spectroscopy is able to detect subtle changes to the surface chemistry of cells. We have previously shown that high-resolution 1H NMR methods can identify tumor cells with the capacity to metastasize, and we now report that the long T2 relaxation value (500-800 ms) observed in metastatic rat mammary adenocarcinoma cells is removed by treatment with fucosidase. Two-dimensional scalar-correlated NMR (COSY) spectra of fucosidase-treated cells show that a cross peak, consistent with scalar coupling between the methyl and methinè groups on fucose and usually associated with malignancy and metastatic ability, is absent. Metastases were observed in only two out of ten rats injected subcutaneously with enzyme-treated cells compared to eight out of ten with untreated cells. NMR studies on isolated cellular lipids identified the long T2 relaxation value only in the ganglioside fraction. This fraction accounts for 51% of the total 14C-labelled fucose incorporated into the cells. We propose that fucogangliosides are an indicator of metastatic potential in rats. The observation that a cell surface metastasis marker has an NMR signal with a characteristically long relaxation value has important consequences for the future use of magnetic resonance imaging and spectroscopy in the cancer clinic.

    Notes: The retrograde tracer wheat germ agglutinin-conjugated horseradish peroxidase was used to establish the organization of the extraocular muscle motoneuron pools in a prosimian, Galago senegalensis, for comparison with the organization in monkeys and non-primates. Medial rectus motoneurons were distributed in three subgroups in the ipsilateral oculomotor nucleus, a pattern similar to that of the monkey. Furthermore, the other component of the near response system, the preganglionic parasympathetic motoneurons, were confined within the Edinger-Westphal nucleus, as in the monkey. In contrast, the distribution of the levator palpebrae and superior rectus motoneurons was similar to that of the cat. Specifically, the majority of levator palpebrae motoneurons were located contralaterally, in the caudal central subdivision of the oculomotor nucleus, and the superior rectus motoneurons had a dorsocaudal location in the contralateral oculomotor nucleus. The distributions of motoneurons supplying the superior oblique and lateral rectus muscles were similar to those of other mammals. Unlike previously studied species, the galago was found to have two accessory muscles, that lie beneath the medial and lateral rectus muscles. Motoneurons supplying the accessory rectus muscles were found ventrolateral to the main abducens nucleus, in a position similar to that occupied by the cat accessory abducens nucleus; although others may be present in the main nuclei. Taken together, these results suggest that the organization of extraocular and preganglionic motoneurons in the galago exhibits both monkey and non-primate features. These observations are consistent with the notion that the galago is a primate species whose oculomotor organization is more similar to the general mammalian scheme. © 1993 Wiley-Liss, Inc.

    Notes: In serum-containing medium, ascorbic acid induces maturation of prehypertrophic chick embryo sternal chondrocytes. Recently, cultured chondrocytes have also been reported to undergo maturation in the presence of bone morphogenetic proteins or in serum-free medium supplemented with thyroxine. In the present study, we have examined the combined effect of ascorbic acid, BMP-2, and serum-free conditions on the induction of alkaline phosphatase and type X collagen in chick sternal chondrocytes. Addition of either ascorbate or rhBMP-2 to nonconfluent cephalic sternal chondrocytes produced elevated alkaline phosphatase levels within 24-72 h, and simultaneous exposure to both ascorbate and BMP yielded enzyme levels at least threefold those of either inducer alone. The effects of ascorbate and BMP were markedly potentiated by culture in serum-free medium, and alkaline phosphatase levels of preconfluent serum-free cultures treated for 48 h with BMP + ascorbate were equivalent to those reached in serum-containing medium only after confluence. While ascorbate addition was required for maximal alkaline phosphatase activity, it did not induce a rapid increase in type X collagen mRNA. In contrast, BMP added to serum-free medium induced a three- to fourfold increase in type X collagen mRNA within 24 h even in the presence of cyclohexamide, indicating that new protein synthesis was not required. Addition of thyroid hormone to serum-free medium was required for maximal ascorbate effects but not for BMP stimulation. Neither ascorbate nor BMP induced alkaline phosphatase activity in caudal sternal chondrocytes, which do not undergo hypertrophy during embryonic development. These results indicate that ascorbate + BMP in serum-free culture induces rapid chondrocyte maturation of prehypertrophic chondrocytes. The mechanisms for ascorbate and BMP action appear to be distinct, while BMP and thyroid hormone may share a similar mechanism for induction. J. Cell.     Notes: Photophores in Porichthys notatus consist of a cellular lens, an underlying area of photogenic tissue and a deep ensheathing reflector. Lens cells exhibit a dense compacted filamentous cytoplasm. Their processes interdigitate and desmosomes exist along their borders. The photogenic tissue, richly supplied by blood vessels, consists of two main cell types: (1) a presumed “photogenic” cell; and (2) a supportive cell. Photogenic cells are characterized by a highly vesiculated cytoplasm and peripheral microvilli. Frequently, they exhibit lamellar membranous whorls. Some whorls display cytoplasmic cores with vesicles. Many of these vesicles communicate via pores with an extracellular channel that envelops the cells. Supportive cells contain cytoplasmic filaments and extend processes around the photogenic cells. Except for isolated desmosomal contact points, a wide extracellular channel intervenes between supportive and photogenic cells. A prominent basal lamina separates supportive cells from the surrounding connective tissue. The strongly birefringent reflector is composed primarily of cells containing guanine crystals. The crystals lie stacked in groups, each membrane-bounded crystal being separated from its neighbor by an intervening layer of cytoplasm. Such an arrangement produces constructive interference and accounts for the high reflectivity of this multilayered structure. Possible relationships of the above structural features and the production of light are discussed.

    Notes: Significant regional morphometric differences are reported for the normal human left ventricle. Epimyocardial capillary density (2,439 capillaries/mm2) is 21% greater than endomyocardial capillary density (2,014 capillaries/mm2). The minimum intercapillary distance is, therefore, increased in the inner region. Endocardial myocytes (617 μm2) have a 53% greater cross-sectional area than epicardial myocytes (403 μm2).
