    Notes: Summary This report concerns ultrastructural and immunohistochemical studies on ballooned neurons of ten patients with Creutzfeldt-Jakob disease (CJD). While abundant ballooned neurons and severe white matter degeneration was seen in six Japanese cases, only occasional ballooned neurons and no white matter degeneration was observed in four cases from the files of Montefiore Medical Center. Ultrastructurally, the ballooned neurons contained granule-coated fibrils of 25 to 40 nm in width and 10-nm neurofilaments. The immunohistochemical studies revealed that most ballooned neurons expressed αB-crystallin, with deposits of reaction products observed in the cytoplasm. A similar intracellular staining pattern was also seen with the antibody to phosphorylated neurofilament proteins (pNFP). Although the proportion of stained ballooned neurons was less, a positive reaction was also observed with antibodies against ubiquitin, stress-response protein 27 (srp 27) and synptophysin, but not with an antibody to srp 72. Our findings suggest that expression of pNFP and synaptophysin by ballooned neurons may reflect axonal impairment and that the presence of αB-crystallin, srp 27 and ubiquitin may be related to the degenerative processes that neurons undergo in CJD.

    Notes: Summary The specific activity of alkaline phosphatase in cultured human meningioma cells varies over a relatively wide range. There is no correlation between the levels of activity and the histological type of meningioma from which the cultures were derived. The enzyme is heat-labile and is strongly inhibited byl-homoarginine, levamisole, and l-bromotetramisole, but unaffected byl-phenylalanine andl-phenylalanylglycylglycine. These findings indicate that meningioma cells synthesize the liver/bone/kidney form of alkaline phosphatase. In contrast to cultures derived from pituitary adenomas, glioblastomas, and astrocytomas in which prednisolone and/or sodium butyrate elicit a manifold increase of alkaline phosphatase activity, with meningioma cells the hormone causes only a slight augmentation in specific activity, and the fatty acid is ineffective. As with other cells producing the liver/bone/kidney enzyme form, no increase in activity occurs in meningioma cells growing in hyperosmolar medium.

    Notes: Summary Ultrastructural alterations in human meningioma cells grown in vitro are reported. In early passages the cells retain some of the characteristic features of the original tumors. These include interdigitations associated with intercellular junctional devices (e.g., desmosomes, gap junctions). However, with repeated subculture these features tend to be less frequent. Typical whorl formations are observed only in primary cultures. The nunber of cytoplasmic filaments, lipid inclusions and other dense bodies increases with time in culture. Cytoplasmic invaginations into nuclei and the appearance of very large cells become more frequent in repeatedly transferred cultures.

    Notes: Summary Flow cytometric techniques were used to compare the DNA content, size and viability of meningioma cells obtained directly from surgical specimens with the same cells after a period of culture. Cells isolated from the original meningiomas and cells in primary culture were similar with regard to size and DNA content, regardless of the histologic subclassification of tumor. The cell populations were essentially diploid with a small proportion of tetraploid cells. Viable cells were smaller and more uniform in size than the nonviable cells. An increase in the number of cells having an elevated DNA content was seen with cultures repeatedly transferred. The latter results suggest that any transfer of information from long-term cultured meningioma cells to the in vivo situation must be done with caution.
