    Notes: Abstract The sensitivity of mast cells to H3-receptor modulation was studied in rat lung under various hormonal conditions. The heterogeneity of mast cell subpopulations in rat lung was assessed by the tissue content of rat mast cell protease I (RMCP I) and rat mast cell protease II (RMCP II). After 24 h fasting, concentrations of RMCP I were unchanged whereas the concentration of RMCP II was significantly reduced by 49%. The [3H]histamine (HA) synthesis was concomitantly decreased by 35%. In addition, the modulation of [3H]HA, synthesis by the H3 receptor agonist, (R)α-methylHA and by the antagonist, thioperamide, observed in control rats, was lost in fasted rats. Single and repeated, administrations of dexamethasone did not influence RMCPI concentrations, but decreased the concentrations of RMCP II with a parallel decrease in [3H]HA synthesis. The inhibitory effect of (R)α-methylHA on [3H]HA synthesis was also reduced. These results suggest that a subpopulation of RMCP II-containing mast cells, very sensitive to environmental factors, could be the mast cells synthesizing HA in an H3-receptor-dependant manner.

    Notes: Summary Intracorporeal ultrasonography was used as a new method to examine the bile ducts during laparoscopic cholecystectomy. A prototype rigid 7.5-MHz ultrasound probe, 10 mm in diameter and 50 cm in length, was introduced during 25 laparoscopic cholecystectomies. A dual scanning technique was developed for complete examination of the bile duct. This entailed transverse scanning via the subxyphoid trocar and longitudinal scanning via the umbilical trocar. The intrahepatic ducts were also visualized by placing the probe on the liver surface. Color Doppler imaging was useful to quickly distinguish the duct from vascular structures. Laparoscopic ultrasonography clearly delineated the bile ducts in all operations except one. The time required for imaging was significantly shorter for ultrasonography than for cholangiography. Our preliminary experience demonstrates that a complete examination of the bile ducts can be performed with intracorporeal ultrasonography in a relatively short period of time.

    Notes: Abstract Since the introduction of a recent laparoscopic ultrasound (LU), the value of this modality in examining the liver and pancreas has been reported. However, a precise scanning technique of LU has not previously been described. Based on our experience with intraoperative ultrasound during laparotomy, we have developed a technique for complete examination of the entire organs using a rigid LU probe. A 7.5-MHz rigid probe, 10 mm in diameter, was employed. The scanning was performed through three trocar ports: right subcostal, subxiphoid, and umbilical. For the liver, the subcostal scanning provided fundamental transverse views. The subxiphoid and umbilical scanning delineated the areas unable to be imaged by the subcostal scanning. For the pancreas, the subcostal and umbilical scanning demonstrated longitudinal and transverse views, respectively. The subxiphoid scanning enhanced examination of the pancreatic head. Three basic probe maneuvers (advancement-withdrawal, lateral movement, and rotation) and various scanning techniques (contact, probe-standoff, and compression scanning) should be utilized appropriately. With a rigid probe, complete LU examination of the liver and pancreas is possible using these techniques. We believe the present scanning method will help more surgeons learn LU.

    Notes: Abstract Aldosterone and dexamethasone stimulate sodium absorption in the rat colon in vivo. In vitro, increased amiloride inhibitable short-circuit current (ISC) has been demonstrated following aldosterone or dexamethasone treatment. Since ISC bears no relationship to sodium flux (JNa) in the untreated rat colon, we measured JNa in partially stripped voltage clamped segments of rat distal colon. Our results demonstrate directly that continuous infusion of aldosterone or dexamethasone for 4–7 days stimulated amiloride inhibitable JNa by stimulating JNaM-S. The amiloride inhibitable portion of JNaM-S was highly correlated with and approximately equal to the amiloride inhibitable ISC. Amiloride had no effect in controls. We conclude that JNaM-S in the rat distal colon is only sensitive to mucosal amiloride after treatment with aldosterone or dexamethasone. the amiloride sensitive ISC in these treated tissues was a good measure of the amiloride sensitive JNa. Small differences between aldosterone and dexamethasone treatment were noted in the effect on transepithelial resistance, potential difference, and the ISC after amiloride.

    Notes: A class of dynamic model approximation techniques based on a frequency domain Taylor series expansion is presented. These methods yield low-order models and are analytical rather than computational in nature; that is, one can retain important parameters (which may be time varying) from the original model in functional form in the low-order model. This feature is very attractive for on-line updating of the simplified model and subsequent control of the process. An example for a tenth-order original model has shown that the analytical low-order models provide excellent approximations to the step response of the original system.

    Notes: A new tracer technique is reported for the determination of liquid-solid contacting efficiency and liquid holdup in trickle beds packed with small alumina particles. Holdup was a function of liquid mass velocity for both porous and nonporous packing. Contacting efficiency remained uniform at a level of 65%.
