    Notes: Abstract ECP31, an embryogenic-cell protein from carrot (Daucus carota L.), was purified by sequential column-chromatographic steps and digested by V8 protease on a nitrocellulose membrane. The resultant peptides were separated by reverse-phased column chromatography and sequenced. The sequences obtained were 70–80% homologous to those of a late-embryogenesis-abundant protein (D34) from cotton (Baker et al, 1988, Plant Mol. Biol. 11, 227–291). The level of ECP31 in somatic embryos of carrot was increased by treatment of the embryos with 3.7 · 10−6 M abscisic acid (ABA) for 48 h, and there was no change in this enhanced level for up to 192 h in the presence of ABA. No similar enhancing effect of ABA was observed on the level of ECP31 in embryogenic callus or segments of carrot hypocotyls. In an immunohistochemical analysis, ECP31 was found in epidermal tissue and in the vascular system of ABA-treated somatic embryos.

    Notes: Fluorescein isothiocyanate (FITC)-labelled asialotransferrin and pyridyl aminated oligosaccharides were prepared from asialotransferrin and human milk using affinity chromatography and high performance liquid chromatography (HPLC), respectively. These substances were incubated with galactosidase or sialyltransferase and then examined by lectin affinity HPLC. The elution patterns changed according to the period of incubation and amount of enzyme. This analytical method using lectin affinity HPLC with fluorescence labelled glycoprotein or oligosaccharides as the substrates has great value for detecting these enzyme under the same chromatographic conditions. In addition, differences were noted in the activity of β-galactosidase toward oligosaccharides having the Gal β(1 → 3)GlcNAc or Gal β(1 → 4)GlcNAc structure at reducing termini.

    Notes: We increased selectivity in the detection of glycoproteins on nitrocellulose membranes by introducing a washing step using sodium hydroxide solution. Glycoproteins on the nitrocellulose membrane were first oxidized by sodium periodate; biotin hydrazide was then coupled to the aldehyde groups generated in the sugar moiety of the glycoproteins. The membrane was washed twice using sodium hydroxide solution, and avidin-horseradish peroxidase was then coupled to the remaining biotin. This system allows the detection of nanograms of glycoproteins on nitrocellulose membranes, and its specificity allows the clear distinction of glycoproteins from the nonglycosylated protein of bovine serum albumin.

    Notes: Concanavalin A (Con A) was biotinylated to various degrees using N-biotinyl-ω-aminocaproic-acid-N-hydroxy succinimide ester as the biotinylation reagent, and then analyzed by isoelectric focusing using PhastGel IEF 3-9. The isoelectric points of biotinylated ConAs were found to decrease with increasing concentration of the biotinylation reagent. Analysis by isoelectric focusing followed by dot blotting clearly indicated that the biotinylated ConA with an isoelectric point lower than that of the original ConA by 2.2 ± 0.6 had the strongest binding activity for ovalbumin.

    Notes: A mixture of homopolymer and graft copolymer was obtained by adding the monomer at 0°C to the polylithiodiene solution. Styrene, methyl methacrylate, and acrylonitrile were used as the monomers. Polylithiodienes were prepared by the metalation of diene polymers, i.e., polybutadiene or polyisoprene, with the use of n-butyllithium in the presence of a tertiary amine (N,N,N′,N′-tetramethylethylenediamine) in n-heptane. The graft copolymers were separated by solvent extraction and were confirmed by turbidimetric titration and elementary analysis. Oxidation of the polybutadiene-styrene grafts revealed that the molecular weight of the side chains was the same as the molecular weight of the free polystyrene formed. The grafting efficiency and grafting percentage were studied for polybutadiene-styrene graft copolymers prepared under various conditions.

    Notes: It was found that the metallation of diene polymers with n-butyllithium in n-heptane takes place in the presence of a tert amine, i.e. N,N,N′,N′-tetramethylethylenediamine. Butyllithium was added to the diene polymer in n-heptane in the presence of an equimolar amount of the amine, and the mixture was allowed to react at 80°C. for 60 min. under nitrogen. As a result, 27% and 9% per monomer unit for polybutadiene and polyisoprene, respectively, were metallated. The effects of the concentrations of the diene polymer, butyllithium, and the amine on the reaction were studied at 80°C., and the reaction mechanism was discussed. The overall rate of metalation was proportional to the diene polymer concentration and to the square root of the butyllithium concentration. The overall activation energies of metalation were 6.6 and 8.4 kcal./mole for polybutadiene and polyisoprene, respectively. Many kinds of polymer derivative were obtained by treating the metalated polymers with various reagents, such as carbon dioxide, Michler's ketone, trimethylchlorosilane, triphenylchlorosilane, benzaldehyde, and pyridine.
