    Notes: Abstract Cuticular components of the green leaves of the Repandae section of theNicotiana species contain compounds that have been shown to be toxic to larvae of the tobacco hornworm larvae,Manduca sexta. The surface constituents of leaves of greenhouse-grownN. repanda, N. stocktonii, andN. nesophila were extracted with methylene chloride in order to isolate the active compounds. Solvent partitioning and gel chromatography was used to isolate a series of hydroxyacylnornicotines (HOAcylNN). The major component was identified asN′-(3-hydroxy-12-methyltridecanoyl)nornicotine. A number of minor 3-hydroxyacylnornicotines, with the acyl group containing C13-C15, were also identified. The HOAcylNN mixture in ethanol was topically applied to first-instar tobacco hornworm larvae at rates of 10, 50, 100, and 200 μg. Mortalities after 48 hr were 33, 44, 78, and 100% respectively.

    Notes: Two regular GC instruments were modified with injection port liners, split-mode injection systems, pressure regulators, effluent traps and needle valves to produce an all-glass GC-2 system. This simple and inexpensive system allows the quantitative analyses of phenols, hydrocarbons, acids, sterols, terpenoids, and alkaloids with excellent reproducibility and linear response. Even more impressive was its ability to analyze a C45-terpenoid alcohol, solanesol, as its TMS-derivative (M.W. 702).

    Notes: A new surface pretreatment for the preparation of wall-coated open tubular (WCOT) glass capillary columns has been evaluated. This technique involves the application of a non-extractable layer of Superox™-4, a 4,000,000 MW polyethylene glycol, to the glass surface as a pretreatment and deactivation agent. Unlike other polyethylene glycols, Superox-4 is stable at high temperatures (〉 300°C) in the absence of oxygen, coats smoothly onto a bare glass surface and resists droplet formation. WCOT columns (SE-54, Carbowax 20M, and SP-2250) prepared using this technique are compared to columns prepared using a modified Grob [2,3] BaCO3 procedure. The Superox-4 pretreated columns were equal or superior in quality to the BaCO3 pretreated columns, based on the appearance of an activity standard and the effective theoretical plates (Neff) per meter. Chromatograms showing practical application of the WCOT columns prepared using both methods is given.

    Notes: A cold on-column injection system for capillary gas chromatography (GC) applications was constructed. It was based upon a conventional split/splitless capillary GC inlet, which in turn was a modification of a conventional packed GC column inlet. The heart of the laboratory constructed cold on-column inlet design was a disposable pyrex micro-sampling pipet, which functioned as a needle guide for sample injection. The sample was injected through a traditional GC septum. Construction of the injection system is described and applications are illustrated by separations of a variety of complex mixtures.

    Notes: A new simplified method was developed for the preparation of immobilized stationary phase fused silica capillary columns coated with OV-1-vinyl, SE-54, and OV-1701-vinyl. This methodology includes the application of the moderately polar stationary phase OV-1701-vinyl as a surface deactivation agent that may also participate in the dicumyl peroxide initiated free radical immobilization process. Practical details of the procedure are presented and applications of the laboratory-produced columns are illustrated with capillary gas chromatographic separations of standard mixtures. Possible contributions of the OV-1701-vinyl deactivation layer to both crosslinking and surface bonding in the immobilization process are discussed.
