    Notes: Both ac and dc conduction processes through thin Langmuir–Blodgett (LB) films of the dodecyloxyphenylurethane of 2-bromo-5 (2'-hydroxyethoxy) tetracyanoquinodimethan (DDOP-C-TCNQ, a redox-active molecule) sandwiched between dissimilar metal (Pt and Mg) electrodes have been studied. The dc conduction changed from a linear I/V characteristic for the lowest applied voltages (±20 mV) to a symmetric nonlinear characteristic obeying a ln I∝V1/4 dependence for voltages up to ±1.5 V, in as-prepared samples. For larger positive voltages, a large increase in current was observed with the dependence changing to a ln I∝V3 law, a dependence not reported previously for metal/LB film/metal systems. For increased negative voltages, the ln I∝V1/4 was again observed. For low applied ac fields, the conductance was found to follow a ωn law with a value of n close to 0.8. The effect of heat annealing the samples was also studied with significant differences in the observed changes in conductance between bilayer and monolayer structures. Emphasis was placed on understanding the conduction process through the single DDOP-C-BHTCNQ layer film. For the monolayer device, hysteresis is observed in the positive bias, and both as-prepared and annealed samples exhibit ln I∝V3 behavior. Such behavior was not observed for the corresponding negative voltages, indicating rectification across a distance approximated by the length of an individual molecule. Possible models of the three-dimensional arrays for both monolayer and multilayer metal/LB film/Mg structures are presented. The weight of existing experimental results does not support the Aviram–Ratner "molecular rectifier'' concept for presently known Pt/LB monolayer/Mg devices.

    Notes: Abstract: Mitochondrial dysfunction and attendant bioenergetic defects are increasingly recognized as playing an important role in neurodegenerative disorders. The increased attention on mitochondrial involvement points to the need for developing cell lines that have neuron-like characteristics for the genetic analysis and modeling of these diseases. We describe the creation of respiratory-deficient SH-SY5Y neuroblastoma cell lines (ρ064/5) by selectively depleting mitochondrial DNA through prolonged exposure to ethidium bromide. Oxygen consumption in these cells and activities of the electron transport chain enzyme complexes I and IV that contain subunits encoded by the mitochondrial genome are eliminated. In contrast, the function of complex II, a nuclear-encoded electron transport chain component, is largely intact in these cells. The ρ064/5 cells retain the ability to differentiate into cells with neuron-like phenotypes following treatment with phorbol ester or retinoic acid. Normal respiratory function is recovered by repopulation of ρ064/5 cells with exogenous human platelet mitochondria. The ρ064/5 cell line serves as a valuable model for the study of neurologic diseases suspected of involving mitochondrial dysfunction.

    Notes: Abstract: Progressive supranuclear palsy (PSP) is a neurodegenerative movement disorder of unknown etiology. We hypothesized that mitochondrial DNA (mtDNA) aberration could occur in this disease and contribute to its pathogenesis. To address this we created transmitochondrial cytoplasmic hybrid (cybrid) cell lines expressing mitochondrial genes from persons with PSP. The presence of cybrid mtDNA aberration was screened for by biochemical assay of mitochondrial gene products. Relative to a control cybrid set, complex I activity was reduced in PSP cybrid lines (p 〈 0.005). Antioxidant enzyme activities were elevated in PSP cybrid lines. These data suggest that mtDNA aberration occurs in PSP, causes electron transport chain pathology, and can produce oxidative stress. Further study of mitochondrial dysfunction in PSP may yield insights into why neurodegeneration occurs in this disease.

    Notes: Abstract: Parkinson's disease may be linked to defects in mitochondrial function. Mitochondrially transformed cells (cybrids) were created from Parkinson's disease patients or disease-free controls. Parkinson's disease cybrids had 26% less complex I activity, but maintained comparable basal calcium and energy levels. Parkinson's disease cybrids recovered from a carbachol-induced increase in cytosolic calcium 53% more slowly than controls even with lanthanum and thapsigargin blockade. Inhibition of complex I with the Parkinson's disease-inducing metabolite 1-methyl-4-phenylpyridinium (MPP+) similarly reduced the rate of recovery after carbachol. This MPP+-induced reduction in recovery rates was much more pronounced in control cybrids than in Parkinson's disease cybrids. Parkinson's disease cybrids had less carbonyl cyanide m-chlorophenylhydrazone-releasable calcium. Bypassing complex I with succinate partially restored Parkinson's disease cybrid, and MPP+ suppressed control cybrid recovery rates. The subtle alteration in calcium homeostasis of Parkinson's disease cybrids may reflect an increased susceptibility to cell death under circumstances not ordinarily toxic.

    Notes: Abstract: Oxidative stress induced by acute complex I inhibition with 1-methyl-4-phenylpyridinium ion activated biphasically the stress-activated c-Jun N-terminal kinase (JNK) and the early transcription factor nuclear factor-κB (NF-κB) in SH-SY5Y neuroblastoma cells. Early JNK activation was dependent on mitochondrial adenine nucleotide translocator (ANT) activity, whereas late-phase JNK activation and the cleavage of signaling proteins Raf-1 and mitogen-activated protein kinase (MAPK) kinase (MEK) kinase (MEKK)-1 appeared to be ANT-independent. Early NF-κB activation depended on MEK, later activation required an intact electron transport chain (ETC), and Parkinson's disease (PD) cybrid (mitochondrial transgenic cytoplasmic hybrid) cells had increased basal NF-κB activation. Mitochondria appear capable of signaling ETC impairment through MAPK modules and inducing protective NF-κB responses, which are increased by PD mitochondrial genes amplified in cybrid cells. Irreversible commitment to apoptosis in this cell model may derive from loss of Raf-1 and cleavage/activation of MEKK-1, processes reported in other models to be caspase-mediated. Therapeutic strategies that reduce mitochondrial activation of proapoptotic MAPK modules, i.e., JNK, and enhance survival pathways, i.e., NF-κB, may offer neuroprotection in this debilitating disease.

    Notes: Beta amyloid (Aβ) peptides accumulate in Alzheimer's disease and are neurotoxic possibly through the production of oxygen free radicals. Using brain microdialysis we characterized the ability of Aβ to increase oxygen radical production in vivo. The 1–40 Aβ fragment increased 2,3-dehydroxybenzoic acid efflux more than the 1–28 fragment, in a manner dependent on nitric oxide synthase and NMDA receptor channels. We then examined the effects of Aβ peptides on mitochondrial function in vitro. Induction of the mitochondrial permeability transition in isolated rat liver mitochondria by Aβ(25–35) and Aβ(35–25) exhibited dose dependency and required calcium and phosphate. Cyclosporin A prevented the transition as did ruthenium red, chlorpromazine, or N-ethylmaleimide. ADP and magnesium delayed the onset of mitochondrial permeability transition. Electron microscopy confirmed the presence of Aβ aggregates and swollen mitochondria and preservation of mitochondrial structure by inhibitors of mitochondrial permeability transition. Cytochrome c oxidase (COX) activity was selectively inhibited by Aβ(25–35) but not by Aβ(35–25). Neurotoxic Aβ peptide can increase oxidative stress in vivo through mechanisms involving NMDA receptors and nitric oxide sythase. Increased intracellular Aβ levels can further exacerbate the genetically driven complex IV defect in sporadic Alzheimer's disease and may precipitate mitochondrial permeability transition opening. In combination, our results provide potential mechanisms to support the feed-forward hypothesis of Aβ neurotoxicity.

    Notes: Two experiments were conducted to examine the relationship between sward surface height, herbage intake and liveweight gain in beef cattle grazed on pasture. In Experiment 1, two ‘animal types’ (18 Charolais × Angus steers and 18 Friesian bulls) were continuously grazed for 22 days during the late autumn on replicated swards maintained at sward surface heights of 6, 10 and 15 cm. Herbage intakes, assessed from the faecal concentration of chromium delivered from an intraruminal controlled release capsule and the in vitro digestibility of hand-plucked herbage samples, were curvilinearly related to sward height (r = 0·76, p〈0·0·01). Average liveweight gains were 0·02, 0·61 and 1·31 kg d-1 (P 〈0·05) and increased linearly (r= 0·84, P 〈0·001) with sward height. The maintenance organic matter intakes of the steers and bulls, with initial mean (± s.e.) live weights of 225 ± 15 kg and 172 ± 15 kg respectively, were estimated to be 3·6 and 3·5 kg d-1 respectively. In Experiment 2 (spring) 36 cattle, including 35 of those used in Experiment 1, were reallocated to sward heights of 5, 10 and 15 cm using the same design as for Experiment 1. Average liveweight gains were 0·94, 1·57 and 1·68kg d-1 (p 0·05) and were curvilinearly related to sward height (r= 0·093, p 〈0·05). Maintenance intakes could not be reliably extimated for the cattle in Experiment 2 because few animals had liveweight gains close to zero. These trials confirm that liveweight gain in continuously grazed finishing steers and bulls increases with sward surface height to maximum of 8–10cm with spring ryegrass/white clover pastures while, in autumn, swards of 12–15cm height are required to achieve maximum performance.
