    Notes: This report describes the use of fluorescence energy transfer between an intrinsic energy donor (tryptophan 177) and two chemically added acceptors to study intermediates in the folding of the β2 subunit of E. coli tryptophan-synthase. Two early folding steps are thus identified and characterized. One is very rapid (its rate constant at 12°C is 0.02 sec-1) and corresponds to the folding of the N-terminal domain into a structure whose overall features approximate well those of the native domain. The second step is somewhat slower (its rate constant at 12°C is 0.008 sec-1) and involves a conformational rearrangement of the N-terminal domain brought about by the interactions between the N-and C-terminal domains within a monomeric β chain. This brings to five the number of intermediates which have been identified and ordered on the folding pathway of the dimeric β2 subunit.

    Notes: The β2 subunit of Escherichia coli tryptophan synthase can be either unfolded in 6 M guanidine, or extensively denatured at acidic pH. These two denatured form of β2 have different circular dichroism spectra and thus correspond to distinct physical states. Here we compare the folding pathways of these two different denatured forms of β chains. We describe the kinetics of regain of a variety of physical, functional, ad immunochemical signals characteristic of six successive steps previously identified on the folding pathway of guanidine unfolded β2. It is shown that whereas identical molecular events over with the same kinetics, the two folding pathways are different, and involve different structural intermediates.

    Notes: The effects of thermoforming on the instantaneous flexural properties and structure of thermoplastic fiber-reinforced composite (FRC) wires were studied to determine the preferred temperature range for clinical forming. Five different formulations of FRC wires were heated to clinically relevant thermoforming temperatures in a special mold that was designed to maintain their shape. In addition, one formulation was also heated without the restraining effect of the mold. Flexural properties were determined. A temperature above the Tg is necessary to allow sufficient softening and avoidance of distortion in shape during clinical forming. However, higher temperatures will result in significant structural disintegration of the wires with consequent decrease in flexural modulus. Thus, for every material, there is a heating range or “working range” where the material can be properly formed with minimal changes in the physical properties. This was primarily related to the Tg of the matrix used.

    Notes: The standard 90° peel test was modified in order to apply it to testing adhesion of thin polymer films to surfaces of silicon substrates, such as the silicon nitride passivation layer used in our solid-state chemical sensors. The goal of this task was to develop a procedure for testing adhesion of both dry and wet films, which would yield repeatable, quantitative results quickly, and with simple sample preparation. It is important to produce valid comparisons of adhesion both over variations of a given polymer composition, and between films made of dissimilar polymer matrices. The method described herein could be applied to adhesion testing of a wide variety of thin polymer layers to solid surfaces.

    Notes: With a view to permanently incorporating ultraviolet stabilizing moieties into polymer systems, monomers containing the phenyl salicylate structure have been prepared and polymerized. Phenyl 5-acryloxymethylsalicylate and phenyl 5-methacryloxymethyl-salicylate were prepared by the chloromethylation of phenyl salicylate and subsequent reaction with sodium acrylate or methacrylate. Homopolymers and copolymers of the methacryloxymethyl monomer were prepared and studied. The ultraviolet spectra of the chloromethyl derivative, the monomers, and the homopolymers were essentially similar to that of the starting material, phenyl salicylate. Copolymers of the ultraviolet-absorbing monomers (2%) with vinyl acetate, vinylidene chloride, and vinyl chloride were prepared. The ultraviolet stability of these copolymers was then compared to that of an unstabilized control series and a series containing phenyl salicylate. While there were some differences in the behavior of the copolymerized absorber it was at least as effective as phenyl salicylate in stabilizing the polymers.

    Notes: Collisional activation, charge reversal, and six different neutralization - reionization mass spectrometric experiments with [H2,O2]·+ radical cations and [H2,O2]·- radical anions were performed in order to probe the predicated existence of neutral water oxide, H2OO, the long sought after tautomer of hydrogen peroxide, HOOH. The experiments together with ab initio calculations indicate that H2OO is a local minimum on the [H2,O2] potential-energy surface, and the elusive molecule seems to be formed as a transient upon neutralization of the corresponding radical cation H2OO·+ in the gas phase.

    Notes: Composition depth profiles of the outer 50 Å of plasma-fluorinated poly(ethylene terephthalate) fibers were obtained by angle-dependent X-ray photoelectron spectroscopy (XPS). The effect of sample geometry on XPS sampling depth and the depth distribution function (DDF) was determined theoretically for cylindrical and hemispherical surfaces. The theoretical DDFs are nonexponential. For cylindrical surfaces, the effect is small, a 22% increase in surface sensitivity. The average XPS sampling depth for smooth, properly oriented fibers is shown to vary, as it does for a planar surface, as the sine of the nominal takeoff angle. The DDF appropriate for cylindrical surfaces was incorporated into a computer program for inversion of angle-dependent XPS data to obtain composition depth profiles of the fibers. Plasma-fluorinated PET fibers were used to demonstrate the use of angle-dependent XPS on fibers. XPS results indicate that most fluorination occurs within the top few “monolayers,” attack is preferentially at the phenyl ring, both —CHF— and — CF2 — moieties are formed, and fluorination causes partial loss of aromaticity. © 1994 John Wiley & Sons, Inc.

    Notes: In the addition of stabilizing agents to plastic materials, problems such as incompatibility, migration, volatility, and/or solvent extraction of the additive frequently arise. With a view toward overcoming such problems in the use of ultraviolet absorbing stabilizers, acrylate and methacrylate derivatives containing the 2-hydroxy-4-alkoxybenzophenone moiety have been synthesized. The ultraviolet absorbing monomers were synthesized by reaction of 2, 4-dihydroxybenzophenone with glycidyl acrylate and glycidyl methacrylate. They were homopolymerized and blended with poly(vinyl chloride), poly(vinylidene chloride), polystyrene, and poly(methyl methacrylate). They were also copolymerized with the corresponding monomers. The polymer blends and copolymers were investigated for ultraviolet stability versus corresponding controls. Effective ultraviolet stabilization was observed. Differences in the stability of some blends as compared to corresponding copolymers is thought to be due to incompatibility.

    Notes: Fungal α-amylase (E.C. 3.2.1.1) and glucoamylase (E.C. 3.2.1.3) were chemically attached to separate reactor modules made from Microporous Plastic Sheet (MPS). Immobilization of enzymes and subsequent chemical reactions were accomplished by pumping reactants through the sheet, i.e., perpendicular to the surface. The expressed activity of the reactor modules was ca. 800 U/g for both fungal α-amylase and glucoamylase. The kinetics and short-term effects of pH and temperature on the expressed activity of the immobilized enzymes were investigated. Using commercially available DE-42 corn syrup at 50% dissolved solids, half-lives of 2000 and 5000 h were achieved for glucoamylase and fungal α-amylase, respectively. The reactors were operated at 50°C and at pH 4.3 for glucoamylase and 5.5 for fungal α-amylase. A typical DE-62 corn syrup product was continuously produced in a two-stage reactor system by pumping the feedstock through the glycoamylase reactor and then through the fungal α-amylase reactor. Saccharide distributions at each stage were controllable to ±0.2%.
