Notes: Synthesis of New Morphine Partial Structure 15,16-SecomorphinanThe synthesis of a new morphine partial structure, 15,16-secomorphinan, is described. One of the series, (±)-15, 16-secocyclorphan (5), has the analgesic potency of morphine and exhibits good binding to the opiate receptor.

Notes: The diastereoselective synthesis of (±)-trans-transoid-7-bromo-8-hydroxy-1-methyl-1,2,3,4,4a,5,10,10a-octahydro-10-phenylbenzo[g]quinoline (8) is described, using an intramolecular Diels-Alder reaction and a reductive cyclisation for piperidine ring-formation as key steps. Compound 8 was prepared as a putative D-1 receptor antagonist which contains (2,2-diphenylethyl)amine as a partial structure.

Notes: Opiate antagonists stimulate the release of LH and might, therefore, contribute to an innovative therapy for the treatment of numerous clinical syndromes characterized by hypofunction of the HHG axis. The purpose of this work was to design and synthesize pure opiate antagonists useful for this therapy. Me, Et, Pr, and PhCH2 groups were introduced at the crucial 14β-position of morphines and morphinans via a hetero-Diels-Alder key step starting from thebaine derivative 1 and tested for opiate antagonism and LH-stimulating activity. Me-, Et-, and Pr-substituted compounds 11a-c were stronger antagonists than naltrexone, whereas Pr and PhCH2 substituents in 11c, 11d, 9d, 9d3, 9d4, and 9d5 led to orally active LH stimulators. Based on our finding that the μ-antagonists 12 and 11b5 showed no LH stimulation, we conclude that a combination of both μ-and χ-antagonism is necessary for potent LH stimulation.

Notes: 1,6-Additions to 3-Methyl-5-methylidene-2(5H)-furanone DerivativesThe anions of thiophenol, methyl malonate and malononitrile react with 3-methyl-5,6-dihydro-2 (4H)-benzofuranone (1c) by the formation of the corresponding 1,6-addition products cis-5c (63%), trans-6 (42%) and trans-7 (76%), respectively. Likewise, the reaction of 3-methyl-5-methylidene-2 (5H)-furanone (1b) with thiophenol yields the 1,6-addition product 5b (66%), and with the sodium salt of methyl aceto-acetate the 1,6-addition product 8 (11%) and the dispiro-dilactone 9 (39%).

Notes: The O 1s line of an ordinary, untreated glass surface and of a glass surface in its dealkalized state were studied by XPS. One single O 1s peak was detected on the dealkalized sample at a binding energy (BE) of 532.5 eV while on the alkali-containing sample, and also on the dealkalized sample after alkali solution treatment, a second peak at 531.1 eV BE also could be detected. Experimental data were backed by semi-empirical quantum chemical calculations. The observed low BE component of the O 1s line, according to the outlined experimental evidence, is connected with the presence of alkali ions. This clearly contradicts earlier publications, interpreting the O 1s line as originating from oxygen atoms in bridging and non-bridging positions.

Notes: We have dealt in detail with the decomposition of ammonium perchlorate in the presence of different amounts of pure, and doped with foreign ions, zinc oxide. The rate and extent of the reaction were markedly increased by the addition of zinc oxide, and the temperature of explosion of ammonium perchlorate was lowered with about 200°C. Doping of zinc oxide with lithium and aluminium oxide, resp., considerably influenced the decomposition, first of all the values for induction periods and rate constants. From the temperature dependence of the rate constants and the times required until explosion occurs, about 32 kcal activation energy was estimated, that is, both the decomposition catalyzed by zinc oxide and the explosion iniciated by zinc oxide take place according to an electron transfer mechanism. The role of zinc oxide is interpreted so that it promotes the melting of ammonium perchlorate.
