Zusammenfassung An isolierten Glomerula und Nierenrindenschnitten von Ratten mit toxischem nephrotischem Syndrom nach Daunomycin wurden Sauerstoffverbrauch, Oxydation der Fettsäuren Oleat, Caprylat, Acetat, Ketonkörperbildung und Gehalte an Coenzym A und Acetyl-CoA nach Inkubation mit Ölsäure gemessen. Glomerula nephrotischer Ratten zeigen gegenüber gesunden Glomerula: a) einen erhöhten Sauerstoffverbrauch und eine gesteigerte14CO2-Bildung mit14C1-Ölsäure als Substrat, b) eine erhöhte14CO2-Bildung mit14C1-Caprylat als Substrat und eine nur geringfügig erhöhte Ketonkörperbildung, c) keine Differenzen der Sauerstoffaufnahme, des Substratumsatzes und der Ketonkörperbildung mit Essigsäure als Substrat, d) einen erhöhten stationären Gehalt an Coenzym A und Acetyl-CoA nach Inkubation mit Ölsäure. Nierenrindenschnitte gesunder und kranker Tiere zeigen hingegen keine Unterschiede im Sauerstoffverbrauch, der Substratutilisation, der Ketonkörperbildung und der Gehalte an Coenzym A und Acetyl-CoA nach Inkubation mit Ölsäure. Die Untersuchung weisen bei dieser vorwiegend glomerulären Erkrankung auf einen gesteigerten Abbau lang- und mittelkettiger Fettsäuren und eine ungestörte Endverbrennung im Tricarbonsäurecyclus hin. Die differenten Befunde gegenüber der nephrotoxischen Nephritis lassen einen unterschiedlichen pathogenetischen Mechanismus beim nephrotischen Syndrom nach Daunomycin gegenüber der immunologischen nephrotoxischen Nephritis vermuten.

Summary Uptake of oxygen, utilisation of fatty acids (oleate 0.84 mM, octanoate 3 mM, acetate 10 mM), production of ketone bodies and the content of free coenzyme A and actyl CoA after incubation with oleate of kidney cortex slices and isolated glomeruli were determined. In nephrotic glomeruli compared with glomeruli of healthy rats: a) the oxygen consumption and the production of14CO2 during incubation with14C1-oleate is increased, b) the production of14CO2 during incubation with14C1-labeled octanoate and the synthesis of ketone bodies is slightly elevated, c) oxygen uptake and production of14CO2 during incubation with14C1-labeled acetate and synthesis of ketone bodies is the same in both groups, d) the content of free coenzyme A and acetyl-coenzyme A after incubation with oleate is increased. Kidney cortex slices of controls and nephrotic animals showed no differences in oxygen uptake, substrate utilisation, synthesis of ketone bodies and content of coenzyme A and acetyl-coenzyme A after incubation with oleic acid. The results indicate an increased oxidation of long-chain and middle-chain fatty acids and an undisturbed endoxidation by isolated glomeruli of rats with nephrotic syndrome. The different results in nephrotoxic nephritis indicate different pathogenetic mechanisms for the daunomycin induced syndrome and for the immunological nephrotoxic nephritis.
