    Notes: Summary Sulfonated shale oils (ammonium bituminosulfonate, ichthammol, Ichthyol), shown previously to induce the directed migration of human neutrophils in Boyden chambers and to inhibit the directed migration towards the chemotactic factors C5a, LTB4, and f-Met-Leu-Phe, were studied for their effect on other neutrophil functions, which are stimulated by chemotactic factors. Like other chemotactic factors ammonium bituminosulfonate increased the adherence of neutrophils to nylon fibers, but it did not induce the release of the primary granule enzyme glucosaminidase from cytochalasin B-treated cells and it did not stimulate the production of oxygen radicals as measured by lucigenin-dependent chemiluminescence if studied under nontoxic conditions. When added together with the chemotactic tripeptide f-Met-Leu-Phe, ammonium bituminosulfonate inhibited adherence augmentation, enzyme release, and oxygen-radical production induced by the chemotactic factor. The results indicate that ammonium bituminosulfonate not only inhibited chemotactic migration but the whole spectrum of neutrophil functions induced by a chemotactic factor.

    Description / Table of Contents: Zusammenfassung Bei Patienten mit Neurodermitis atopica leichten Schweregrades konnte im Rahmen einer kontrollierten Studie mit Hilfe von “matched pairs” eine gegenüber gesunden Kontrollpersonen signifikant (α=0,005) herabgesetzte in vitro 3H-Thymidin-Einbaurate in kultivierte Lymphozyten des peripheren Blutes, nach Stimulation mit dem T-Zellenmitogen Concanavalin A (Con A), festgestellt werden. Anhand von zehn differenten Konzentrationen von Con A wurden die Dosis-Wirkungsbeziehungen innerhalb der Paare bestimmt. Während höhere Con A-Konzentrationen in den Lymphozytenkulturen zu optimalen 3H-Thymidin-Einbauraten in neusynthetisierte DNS führten, waren die Unterschiede zwischen Neurodermitikern und Kontrollpersonen eindeutiger bei niedrigeren, suboptimalen Con A-Konzentrationen. Die spontane 3H-Thymidin-Einbaurate in kultivierte Lymphozyten ohne Con A-Stimulation (spontane DNS-Synthese) war bei Neurodermitikern im Vergleich zu Kontrollpersonen erhöht. Die Befunde weisen auf eine beeinträchtigte T-Zellenfunktion bei Neurodermitikern hin.
    Notes: Summary The Concanavalin A (Con A) stimulation response of peripheral lymphocytes in vitro was found to be decreased statistically significant (α=0.005) in patients with light atopic dermatitis, Strictly matched pairs were examined. Using ten graded concentrations of Con A, dose-response relations of the pairs were demonstrated, differences were more evident at lower suboptimal concentrations. Background DNA-synthesis of the patients was elevated.
