    Notizen: Summary The present investigation was conducted to examine the combination effect of N-butyl-N-(4-hydroxybutyl) nitrosamine (BBN) and N-methyl-N-nitrosourea(MNU) in rat urinary bladder carcinogenesis. Experiment 1 was performed in the groups treated by oral BBN administration in combination with MNU intraversical instillation and their control groups. In the groups given both BBN and MNU, the ratio of rats with the nonpapillary type to carcinoma-bearing rats was significantly higher than in the controls. Since most of the carcinomas were non-invasive, the observation period was prolonged in the groups given both BBN and MNU in experiment 2. However, even after a longer observation period, no invasive carcinoma was observed. These results suggest that high-grade and invasive carcinomas similar to those induced by BBN and MNU in the heterotopically transplanted rat bladder cannot be induced in the natural bladder of the rat.

    Notizen: Summary The expression of specific T-cell receptor gene segments by T lymphocytes appears to be critically important for the induction of several experimental autoimmune diseases mediated by these cells. We examined whether this situation also applied to non-obese diabetic mice by using various T-cell receptor VΒ-specific monoclonal antibodies. No significant age- or sex-related differences were observed in VΒ usage by peripheral and splenic T lymphocytes. CD8+ T lymphocytes among the islet-derived mononuclear cells isolated from 20-week-old female non-obese diabetic mice showed heterogeneity of their VΒ gene usage. In order to examine the role of T lymphocyte subsets expressing specific T-cell receptor VΒ gene segments in the development of diabetes mellitus, T-cell receptor VΒ-specific monoclonal antibodies were administered to 10-week-old male non-obese diabetic mice treated with cyclophosphamide. None of the antibodies used could significantly diminish the incidence of cyclophosphamide-induced diabetes and the severity of insulitis [anti-VΒ3 (11 of 22 mice became diabetic, 50%), anti-VΒ5 (9 of 14, 64%), anti-VΒ8 (9 of 21, 43%), anti-VΒ11 (12 of 23, 52%), anti-VΒ14 (7 of 12, 58%), and anti-VΒ5 + anti-VΒ11 (6 of 12, 50%)] when compared with control mice (12 of 21, 57%). In addition, there were no significant differences in T-cell receptor VΒ usage between diabetic and non-diabetic cyclophosphamide-treated mice. These results suggest that five T-lymphocyte subsets expressing different T-cell receptor VΒ gene segments, considered to be candidates involved in the pathogenesis of autoimmune diabetes, do not individually contribute to the development of cyclophosphamide-induced diabetes in non-obese diabetic mice.

    Notizen: To investigate which component of the plasma membrane of the hamster egg plays the central role in the sperm-egg fusion, the egg membrane was treated with a variety of proteolytic, carbohydrate-hydrolyzing, lipid-hydrolyzing, and other enzymes. The only enzyme that markedly effected the ability of the egg membrane to fuse with spermatozoa was phospholipase C. The lipid moieties of the egg plasma membrane (and possibly of the sperm membrane) must be of primary importance in sperm-egg fusion at fertilization.
