    Notes: Abstract The majority of patients with acute myeloid leukemia (AML) are elderly, and their response to chemotherapy is poorer than that of younger patients. The combination of mitoxantrone (MTN) and cytosine arabinoside (Ara-C) is a possible alternative to an anthracycline/Ara-C combination for the treatment of AML in these patients. Of 52 older patients (〉 59 years) referred over a 3.5-year period, 33 patients (age range 60–78 years, median 67 years) received MTN and Ara-C as therapy for newly diagnosed AML. MTN was administered at a dose of 12 mg/m2/day, intravenously, for 3 days (23 patients), or 10 mg/m2/day for 5 days (10 patients), and Ara-C at a dose of 100 mg/m2 twice daily, intravenously, for 7 days. Complete remission (CR) was achieved in 16/33 patients (48%). The median remission duration was 6 months (range 1–37 months). The median survival was 14 months for those who achieved CR compared with 9 months for those with resistant disease. Two patients remain in first CR after 13 and 37 months, but three patients died whilst receiving consolidation therapy. In selected elderly patients with AML, the combination of MTN and Ara-C provides an acceptable alternative to an anthracycline/ Ara-C regimen, with a higher CR rate than historical controls. However, the CR rate and remission duration remain low compared with those of younger patients, supporting the need to investigate new approaches to treatment in this population.

    Notes: Abstract MRI was performed on patients with miliary pulmonary tuberculosis to look for brain involvement and to study the features sequentially, during treatment. We studied seven patients with typical radiographic tuberculosis, and no symptoms or signs of central nervous system involvement. Conventional spin-echo (SE) imaging, including contrast enhanced images, was performed in all cases. All patients showed brain involvement: four patients showed lesions mainly less than 3 mm in diameter, better seen on contrast-enhanced images. These patients showed oedema around the lesions after 2 months of treatment, with subsequent regression on follow-up. The remaining three patients had multiple lesions, 3 mm or more in diameter, which showed a gradual decrease on follow-up. We conclude that the brain may commonly be involved in miliary pulmonary tuberculosis. The response to treatment depends on the stage of the granuloma and shows a definite pattern of healing on follow-up.

    Notes: The physical properties of a new class of ionomers comprising a semirigid polyphenylene ether backbone were investigated. The material was studied principally in the form of films of ca. 150 μm micrometers in thickness. The study focused on material of 600 equivalent weight. Torsion pendulum results showed a significant increase in the glass transition temperature of the ionomer relative to that of its ester precursor but gave no evidence of ionic clustering. Stress-strain studies performed both in water and in 20% KOH solution at 80°C indicated that the mechanical properties of the material remain stable under these conditions. Differential scanning calorimetry was used to investigate both the high temperature behavior and the water uptake of the ionomer films. The behavior of this ionomer at high temperature and in an alkaline solution is of particular interest in view of the possible use of these films as separators in an alkaline electrolysis cell.

    Notes: The effect of polymer morphology on the dielectric breakdown of isotactic polypropylene was investigated under the influence of a ramped ac voltage. Samples were prepared by melt-pressing polypropylene beads into plaques and subsequently quenching or isothermally cyrstallizing these plaques. Some of the quenched samples were annealed at different temperatures to induce further crystallinity. The plaques were then characterized by wide-angle X-ray diffraction, density measurements, and differential scanning calorimetry. The 60 Hz, ac breakdown voltages of the samples were determined at 7°C, 23°C, and 45°C in a specially designed test cell. There was a lack of any significant effect on the ac breakdown strength of isotactic polypropylene of either the overall crystallinity or the crystallite thickness. This result can be understood in light of previous studies relating the electrical breakdown of polymers to morphology. It is postulated that the degree of crystallinity would influence the breakdown characteristics only when the amorphous regions constitute a small fraction of the total volume, that is, at very high crystallinity values - values beyond the range of the present study.

    Description / Table of Contents: Deterioration of timber in cooling towers. Possible causesTimber in cooling towers is always exposed to exacting working conditions and shows many signs of deterioration. In order to clarify two specific cases of deterioration, the timber of cooling towers (redwood and fir) has been subjected to mechanical tests for loss of mechanical strength whilst the cooling water and circulating water have been analyzed. It was found that alkaline water with iron content causes a marked loss of mechanical strength and that the impregnation of the wood with tar oils has a very great protective effect as it inhibits the corrosion caused by iron nails.
    Notes: Holz, das durchgängig in Kühltürmen harten Betriebsbedingungen ausgesetzt ist, zeigt vielartige Schäden. Zur Aufklärung zweier Schadensfälle wurde das Holz von Kühltürmen (Redwood u. Fichte) mechanisch auf Wertminderung (Steifigkeits-Einbuße) sowie das Kühlwasser und das Umlaufwasser analytisch untersucht. Festgestellt wurde, daß alkalische, eisenhaltige Wässer zu starkem Steifigkeitsverlust (Schwächung) führen und, daß Imprägnieren des Holzes mit Teerölen in hohem Maße zerstörungsschü;tzend wirkt, da dies auf die Korrosion eiserner Nägel inhibierend wirkt.

    Notes: Light scattering has been used to investigate the structure of human tracheobronchial mucin glycoproteins (HTBM) from the sputum of cystic fibrosis patients. The specimen was extracted using 6M guanidinium hydrochloride solution and fractionated by gel exclusion chromatography on Sephacryl S-1000. The fractionated HTBM was purified by density gradient ultracentrifugation. Purity of the resulting material was confirmed by SDS polyacrylamide gel electrophoresis and uv spectroscopy. Light scattering measurements on the fractionated mucins yield weight-average molecular weights Mw, and z-average radii of gyration Rg, z. The native cystic fibrosis HTBM consisted of a high molecular weight fraction with Mw = 9.3 × 106 daltons and a lower molecular weight fraction contanining partly degraded mucins. After reduction and carboxymethylation of the high molecular weight native fraction, the resulting material was separated into three pools with Mw values of 5.1 × 106, 1.6 × 106, and 400,000. The derived molecular weights for the protein cores Mp,w, and the experimental radii of gyration are found to be consistent with the Mp,w - Rg relation established previously for submaxillary, cervical, and gastric mucins. These results imply that HTBM has the same extended-coil conformation reported for other mucins and has a molecular structure consisting of subunits, linked into linear chains via covalent (disulfide) bonds.

    Notes: Human tracheobronchial mucin isolated from cystic fibrosis patients (CF HTBM) was purified using a combination of gel filtration and density gradient centrifugation. The resulting mucin was fractionated to reduce polydispersity and to facilitate studies of the molecular weight dependence of mucin viscoelasticity in concentrated solution. The viscoelastic properties of CF HTBM were examined in distilled water, 0.1M salt solutions and chaotropic solvents. In controlled strain experiments (strain ≥ 5%) with increasing mucin concentration, a crossover from sol to gel behavior is observed. The gel strength, as measured by the magnitude of the storage modulus at comparable mucin concentrations, is greatest for distilled water, intermediate for 0.1M NaCl, and lowest far 6M GdnHCl. In distilled water, high molecular weight mucin undergoes a sol-gel transition at ∼ 12 mg/mL, and shows evidence of a plateau modulus at higher concentrations. The storage and loss moduli of concentrated high molecular weight fractions in 6M GdnHCl exhibit a power law dependence on frequency typical of weak gels near the sol-gel transition at 20 mg/mL. Similar rheology is observed in 0.1M NaCl and 0.091M NaCl/3 mM CaCl2, but with evidence for additional weak associations at low frequency. The power law exponent in these systems is 0.70 ± 0.02, in good agreement with prediction for networks formed by a percolation mechanism. Low molecular weight fractions in these solvents exhibit a fluid-like viscoelastic response. However, low molecular weight mucin in distilled water shows a strain-dependent increase in elasticity at low frequency indicative of weak intermolecular associations. Comparison of the rheological behavior of CF HTBM with our earlier studies of ovine submaxillary mucin lends support to the idea that carbohydrate side-chain interactions are important in the gelation mechanism of mucins. © 1995 John Wiley & Sons, Inc.
