    Notizen: Presented is a comprehensive program designed to isolate human cytokine genes and investigate their relative induction, and to analyze cytokine activities in cell culture, animal tumor models, and human clinical trials. Human cytokine cDNAs have been isolated from a cDNA library made from normal human peripheral blood leukocytes (PBLs) treated with Sendai virus and the relative induction of tumor necrosis factor (TNF), alpha and gamma interferons (IFN-α, IFN-γ), and interleukin-1 beta IL-1β genes has been analyzed. In the Sendai virus-induced PBL system, IL-1β mRNA was shown to be approximately twofold higher than TNF or IFN-α mRNA whereas IFN-γ mRNA was 50-100-fold lower than TNF or IFN-α mRNA. The cytotoxic activity of TNF was analyzed on several cell lines and IFN-α and IFN-γ were shown to potentiate TNF cytotoxicity about 2-200-fold depending on cell lines. The LD50 for recombinant TNF in BALB/c mice was determined to be 6 × 107 U/kg and the therapeutic dose of recombinant TNF in sarcoma 180 bearing BALB/c mice was 3 × 105 U/kg, indicating a wide therapeutic index. Phase I clinical trials of recombinant TNF given I.V. indicated a tolerated dose of 150,000 U/kg with biphasic half-life (T-1/2) of 2 and 31 min following TNF injection. Phase II trials of TNF and trials of TNF combined with IFN-α are in progress. These studies indicate that cytokines such as TNF and IFN-α are subject to similar induction systems, potentiate each other's activities, and can be tolerated at specific doses for potential therapeutic use.

    Notizen: Summary Irides from 3–5 day old rats have been grown 1–3 mm from superior cervical or lumbar paravertebral sympathetic ganglia in modified Rose chambers. The two muscles of the iris received distinctly different innervation patternsin vitro, and these were similar to those seenin vivo. Varicose, adrenergic fibres were consistently associated with the dilator pupillae rather than with the sphincter pupillae while excitatory, cholinergic junctions developed between the nerve fibres and the muscle cells of the sphincter but not the dilator. There was a lack of specificity shown by the sympathetic neurons during this innervation. Fibres from lumbar ganglia formed plexuses within the dilator similar to those formed by superior cervical fibres, and sympathetic, cholinergic fibres were able to substitute for the normal parasympathetic, cholinergic fibres in the sphincter.

The distribution and morphology of chromaffin cells in the para-aortic region and in the ganglia of the paravertebral sympathetic chain was studied with fluorescence histochemistry and electron microscopy. 2. Four types of chromaffin cell were distinguished largely on the basis of their vesicular content: Type I cells contain large, electron-dense vesicles (600–7000 Å) and are comparable to noradrenaline-containing cells in the adrenal gland, Type II cells contain large, vesicles (600–7000 Å) that are filled with a less electron-dense material than that in Type I cells and are comparable to adrenaline-containing cells in the adrenal gland, Type III cells contain smaller vesicles (1000–3000 Å) that are incompletely filled with an electron-dense material and may represent cells that have been depleted of their catecholamines by stimulation, Type IV cells are clearly different from the other three cell types with respect to the size and appearance of the vesicles (1000–1500 Å), nuclei and rough endoplasmic reticulum and may represent immature sympathetic neurons. 3. Nerve profiles, identified as cholinergic, were found in close apposition with all four cell types. No examples of a close association between processes of chromaffin cells and sympathetic neurons were found.

Two types of chromaffin cells were distinguished: Type A cells contain large, electron dense vesicles (2000–6000 Å) and are equated to Type I chromaffin cells seen in vivo, i.e. they contain noradrenaline; Type B cells contain smaller vesicles (700–2000 Å) which are incompletely filled with an electron dense material and are equated to Type III chromaffin cells seen in vivo, i.e. cells depleted of their catecholamines by stimulation. No cells comparable to Types II and IV cells in vivo were seen. 3. Close associations between the cultured chromaffin cells and sympathetic neurons were observed with the light microscope, but no examples of synaptic structures were seen in the material examined with electron microscopy in this study.

    Notizen: Summary The distribution of the sarcoplasmic reticulum and sarcolemmic tubules in the radula protractor muscle of the whelk, Busycon canaliculatum, has been investigated. The sarcoplasmic reticulum consists of an interconnected system of cisternae and tubular channels. The cisternae are closely associated with the sarcolemma. The tubular channels project from the cisternae into the interior of the cell and run parallel to the long axis of the myofilaments. Parallel tubular channels are interconnected with one another by short branches. This finding of an elaborate sarcoplasmic reticulum supports previous physiological work on this smooth muscle which indicated the presence of an intracellular compartmentalization of calcium ions. There is also an extensive system of tubular invaginations of the sarcolemma which we have termed sarcolemmic tubules. These tubules are 600 Å in diameter and about 0.5 microns in length. There is a substructure associated with the leaflet of the tubular membrane bordering the extracellular space. The sarcolemmic tubules penetrate only half a micron from the surface of the cell and interdigitate with the sarcoplasmic reticulum associated with the sarcolemma. Calculations have shown that the surface area of this smooth muscle cell is more than doubled by the presence of sarcolemmic tubules.

    Notizen: Three pairs of thymus primordia are found at 6 to 6.5 mm. on the dorsal lateral ends of the second, third and fourth visceral pouches. Those on each side after fusing by growth and migratin come to lie above the third visceral pouch, whence the thymus migrates upward and backward; growing in size, it stretches above the ends of all the gill pouches. It pushes inward into the mesenchyme at 12 to 13 mm. and becomes perforated and surrounded by blood vessels and connective tissue which separate it almost completely from the epithelium. No septa are found; occasionally the third primordium fails to fuse and forms a separate lobe.The early thymus is a syncytium in which are found lymphoblasts, identified by structure of the cytosome and its behavior during mitosis. Evidence is presented that lymphoblasts migrate into the thymus where they increase in number with corresponding increase in length of cytoplasmic bridges and size of intercellular spaces. At 10 mm. begins a rapid increase in size of the thymus and in number of lymphoblasts and decrease in size of the latter, culminating at 12 to 13 mm. in their transformation into thymocytes. A medulla associated with blood vessels is unmistakable at 30 mm.

    Notizen: The thyroid primordium is a solid median outgrowth from the pharynx which is attached to the truncus arteriosus at its bifuracation. The thyroid is soon detached from the pharynx and migrates to its definitive position ventral to the aorta between the bases of the third visceral pouches. After detachment the primary follicle appears in the lower part of the primordium; during migration and early growth it is divided apparently by stress and pressure to form secondary follicles. Independent follicles are formed also by secretion of colloid between solid masses of thyroid cells. Other secondary follicles are formed by pinching off evaginations from large follicles. Colloid appears soon after the primary follicle is divided. The adult thyroid is a group of follicles scatterd in a venous plexus in the ventral pharyngeal region, around the aorta.

    Notizen: Triton X-100 insoluble neurofilament (NF) fractions were obtained from two parts of the stellate ganglion and the main giant axon. These were analyzed by one- and two-dimensional gradient polyacrylamide gel electrophoresis, cyclic assembly and disassembly, and electron microscopy. The NF fractions from the ganglion cell bodies (GCB) and from the part of the ganglion mainly consisting of axon initial segments (GIS) were of similar composition; neither contained detectable amounts of the 220 kda and high molecular weight ( 400 kda) NF subunits that were prominent in the axonal NF fraction. However, the GCB and GIS did contain large quantities of a set of 65 kda polypeptides that were minor constituents of the axonal NF fraction. The 65 kda-containing NF fraction from the ganglion could be cyclically disassembled and reassembled, but only under low salt conditions, in contrast to the high salt conditions used to cycle axonal NFs. A comparison of the peptide map of the 65 kda polypeptides with that of the 60 kda axonal NF subunit showed them to be different. These biochemical differences between the ganglionic and axonal NF fractions correlated with morphologic distinctions: ganglionic NFs were relatively smooth surfaced, whereas axonal NFs had long sidearms. Such observations support the hypothesis that the NF cytoskeleton of the neuron soma is different from that of the axon. Furthermore, the change from the somal form to the axonal form of NFs appears to occur in the region where the axon initial segment increases in diameter to become the axon proper.
