    Notes: Abstract Presence of the dominant Tu gene in Lactuca sativa is sufficient to confer resistance to infection by turnip mosaic virus (TuMV). In order to obtain an immunological assay for the presence of TuMV in inoculated plants, the TuMV coat protein (CP) gene was cloned by amplification of a cDNA corresponding to the viral genome using degenerate primers designed from conserved potyvirus CP sequences. The TuMV CP was overexpressed in Escherichia coli, and polyclonal antibodies were produced. To locate Tu on the L. sativa genetic map, F3 families from a cross between cvs “Cobbham Green” (resistant to TuMV) and “Calmar” (susceptible) were genotyped for Tu. Families known to be recombinant in the region containing Tu were infected with TuMV and tested by the indirect enzyme-linked immunosorbent assay (ELISA) using the anti-CP serum. This assay placed Tu between two random amplified polymorphic DNA (RAPD) markers and 3.2 cM from Dm5/8 (which confers resistance to Bremia lactucae). Also, bulked segregant analysis was used to screen for additional RAPD markers tightly linked to the Tu locus. Five new markers linked to Tu were identified in this region, and their location on the genetic map was determined using informative recombinants in the region. Six markers were identified as being linked within 2.5 cM of Tu.

    Notes: Abstract The second largest cluster of resistance genes in lettuce contains at least two downy mildew resistance specificities, Dm5/8 and Dm10, as well as Tu, providing resistance against turnip mosaic virus, and plr, a recessive gene conferring resistance against Plasmopara lactucae-radicis, a root infecting downy mildew. In the present paper four additional genetic markers have been added to this cluster, three RAPD markers and one RFLP marker, CL1795. CL1795 is a member of a multigene family related to triose phosphate isomerase; other members of this family map to the other two major clusters of resistance genes in lettuce. Seven RAPD markers in the region were converted into sequence characterized amplified regions (SCARs) and used in the further analysis of the region and the mapping of Dm10. Three different segregating populations were used to map the four resistance genes relative to molecular markers. There were no significant differences in gene order or rate of recombination between the three crosses. This cluster of resistance genes spans 6.4 cM, with Dm10 1.2 cM from Dm8. Marker analysis of 20 cultivars confirmed multiple origins for Dm5/8 specificity. Two different Lactuca serriola origins for the Du5/8 specificity had previously been described and originally designated as either Dm5 or Dm8. Some ancient cultivars also had the same specificity. Previously, due to lack of recombination in genetic analyses and the same resistance specificities, it was assumed that Dm5 and Dm8 were determined by the same gene. However, molecular marker analysis clearly identified genotypes characteristic of each source. Therefore, Dm5/8 specificity is either ancient and widespread in L. serriola and some L. sativa, or else has arisen on multiple occasions as alleles at the same locus or at linked loci.

    Notes: It is shown in this paper that non-conforming finite elements on the triangle using second-degree polynomials can be easily built and used. Indeed they appear as an ‘enriched’ version of the standard piecewise quadratic six-node element. This work is divided into two parts. In the first we present the basic properties of the element, namely how it can be built and basic error estimates. We also show that this element exhibits a very peculiar regularity property. In the second part we apply our element to the approximation of viscous incompressible flows and more generally to the approximation of incompressible materials.

    Notes: A method to connect momentum Navier-Stokes equations with the universal law of the wall using the finite element method is developed for turbulent wall flows. This method is based on a domain decomposition of the fluid into subdomains near a solid boundary where the law of the wall is valid. A transmission formulation is introduced to match these regions and a new class of boundary finite element is used. This finite element takes into account the near-wall profile of the velocity and the transmission conditions. Computational results are presented for Poiseuille flow and flow over a backward-facing step.

    Notes: The present paper describes a directionally adaptive finite element method for high-speed flows, using an edge-based error estimate on quadrilateral grids. The error of the numerical solution is estimated through its second derivatives and the resulting Hessian tensor is used to define a Riemannian metric. An improved mesh movement strategy, based on a spring analogy, but with no orthogonality constraints, is introduced to equidistribute the lengths of the edges of the elements in the defined metric. The grid adaptation procedure is validated on an analytical test case and the efficiency of the overall methodology is investigated on supersonic and hypersonic benchmarks.

    Notes: Some finite element approximations of incompressible flows, such as those obtained with the bilinear velocity-constant pressure element (Q1-P0), are well known to be unstable in pressure while providing reasonable results for the velocity.We shall see that there exists a subspace of piecewise constant pressures that leads to a stable approximation. The main drawback associated with this subspace is the necessity of assembling groups of elements, the so-called ‘macro-elements’, which increases dramatically the bandwidth of the system.We study a variant of Uzawa's method which enables us to work in the desired subspace without increasing the bandwidth of the system. Numerical results show that this method is efficient and can be made to work at a low extra cost. The method can easily be generalized to other problems and is very attractive in three-dimensional cases.

    Notes: A finite element simulation of the dip coating process based on a discretization of the continuum with discontinuous pressure elements is presented. The algorithm computes the flow field from natural boundary conditions while an extra condition provided by the existence of free surface is employed to displace the meniscus location towards the actual position. The process is iterative and uses a pseudo-time stepping technique coupled to a cubic spline fitting of the free surface. Numerical predictions exhibit good agreement with experimental data for Newtonian fluids in the case of flat plate dip coating as well as in the case of wire dip coating.
