    Notes: Abstract A method for rapidly determining the vertical and horizontal distribution of insect eggs in fragile soil is described. Liquefied agar is allowed to permeate intact soil samples from below; after cooling, the resulting solid is cut into thin sections, from which eggs can be recovered by washes with hot water. This technique revealed that in organic (muck) soil in the laboratory, undisturbed onion flies, Delia antiqua (Meigen) (Diptera: Anthomyiidae), laid 95% of their eggs within a 10 mm diameter zone around the base of a surrogate onion ‘stem’ arising vertically from the soil. Ninety % of all eggs were found in the top 12 mm of soil, with an apparent maximum at depth of 2–4 mm. Increasing fly density from 30 to 200 flies per 30×30×42 cm cage flattened the horizontal distribution of eggs and extended the ovipositional range from c. 15 mm to beyond 60 mm, suggesting there was competition for the preferred ovipositional sites. Surface treatment of muck soil in the field by granular formulations of the insecticides Lorsban™ 15 G (active ingredient chlorpyrifos) and Dyfonate™ 15 G (fonofos) three weeks prior to bioassaying reduced egg-laying at depths greater than 8 mm. The relation between the measured egg distribution and mortality factors in soil (low moisture and high temperature) is discussed.

    Notes: Abstract In laboratory choice experiments, the spices dill, paprika, black pepper, chili powder, ginger, and red pepper deterredDelia antiqua oviposition by 88–100%. Dose-response choice tests demonstrated that 1 mg of ground cayenne pepper (GCP) placed within 1 cm of artificial onion foliage reduced oviposition by 78%. A synthetic analog of capsaicin, the principal flavor ingredient of red peppers, deterred oviposition by 95% when present at 320 ppm in the top centimeter of sand (the ovipositional substrate). However, in no-choice conditions 10 mg GCP was not an effective deterrent. Sevana Bird Repellent and Agrigard Insect Repellent both use red pepper as a principal ingredient; at recommended field rates, neither of these materials was an effective ovipositional deterrent either in laboratory or field. Capsaicin-based materials do not appear to be candidates for onion maggot control via behavioral modification.

    Notes: Abstract The chromosomal locations of genes for resistance to stem rust (Puccinia graminis Pers.: Pers. f. sp. tritici Eriks. & E. Henn.) in the wheat (Triticum aestivum L.) cultivar ‘Waldron’ (WDR) were determined by monosomic analyses. Wheat lines WDR-B1, -C2, -E4, and -F1,which have single genes for resistance to stem rust derived previously from WDR sel. ‘Little Club’, were crossed onto a complete set of 21 ‘Chinese Spring’ monosomics. The F2 and backcross-F1 (BC1F1) seedlings from each of the 84 crosses were tested for reaction to culture 111-SS2 (CRL-LCBB) of stem rust, and a few selected segregants were analyzed cytologically for chromosome number. The F2 from 2 crosses of WDR-C2, -E4 and -F1 and the BC1F1 from 2 crosses of WDR-F1 were tested also with culture Or11c (CRL-QBCN). Significant deviations from disomic ratios towards monosomic ratios in the F2 and BC1F1 were used to determine which chromosomes carried the genes for resistance. Cytological analyses of certain BC1F1 and susceptible F2 plants were used to help identify the location of the genes for rust resistance. WDR-B1 has a gene, herein designated Sr41, for resistance on chromosome 4D. WDR-C2 has a gene on chromosome 7 A that may be the same as one previously designated SrWld2. WDR-E4 has a gene on chromosome 2A, possibly SrWld1, which is effective against most or all North American stem rust cultures. WDR-F1 has a gene on chromosome 6B that is the same as or similar to Sr11.

    Notes: Abstract The Triticum aestivum L. cultivar ‘Waldron’ has long lasting resistance to most North American stem rust (Puccinia graminis Pers.:Pers. f. sp. tritici Eriks. and E. Henn.) isolates. The objective of this research was to develop wheat lines monogenic for resistance to stem rust from ‘Waldron’ using allelism tests and tests for reaction to a series of ten stem rust cultures having a range of virulences. Twelve lines homozygous for single resistance genes were selected as parents of a diallel cross to test for allelism among genes for resistance. We identified 6 lines or groups of lines (WDR-A1, the WDR-B1 and WDR-B2 group, the WDR-C1 and WDR-C2 group, WDR-D1, the WDR-E1, WDR-E2, WDR-E3, and WDR-E4 group, and WDR-F1) that carried different single genes for resistance from ‘Waldron’. A seventh line (WDR-G1) probably has two genes for resistance, one in common with WDR-C1 and WDR-C2. The gene in the WDR-E group is probably the same as SrWld1, and the one in WDR-F1 the same as Sri11. ‘Waldron’ probably has two or more genes for resistance to stem rust that previous genetic studies did not detect.

    Notes: Summary Several chemicals were evaluated for their gametocidal properties to control pollination in hybrid wheat (Triticum aestivum L. em Thell.) seed production. In a preliminary study, four chemicals were applied to three cultivars of spring wheat at two application times. RH-532 was the most effective of the four gametocides, reducing fertility to a 0 10% level for all three cultivars. In a second experiment, RH-532 was applied to one cytoplasmic male-sterile and three normal spring wheats at four rates (0.56, 1.12, 2.24, and 4.48 kg/ha) and at two times of application. Alternate strips of pollinator were planted throughout the experiment. All treatments decreased fertility. plant height, and spike length of the three normal wheats. RH-532 did not inhibit spike emergence of two semidwarf cultivars or one of conventional height. Cultivars did not respond similarly to treatments. Yields of cross-pollinated seed were only 1 21% of the normal yield when fertility levels on treated plants reached 0 10%. Hybrid seed content of the harvested seed ranged from 2 55%. On the treated male-sterile line, yield was significantly reduced in comparison with the open-pollinated check. Inhibitory properties of this chemical suppressed the fertilization potential of the female as well as inducing male sterility. The differential response of genotypes to treatments indicates that cultivars should be screened for male and female sensitivity to this chemical. If differential sensitivity is found, breeding and selection to improve this trait may be possible. Different environmental conditions may alter the response of cultivars to this chemical, with less effect on the female structures.

    Notes: Abstract Onion fly females,Delia antiqua (Diptera: Anthomyiidae) laid the most eggs on ovipositional dishes havingn-dipropyl disulfide (Pr2S2) release rates of 1–6 ng/sec from polyethylene capsules placed beneath a sand substrate. When dipropyl disulfide was released from the wax coating of surrogate foliage rather than from the substrate, ovipositing females again responded differentially to various concentrations, laying more eggs around stems containing 0.075 and 0.089 mg/stem. Factorial combinations of several concentrations released from surrogate foliage and substrate showed that releases from surrogate foliage stimulated four times more egg-laying than releases from the substrate. Females tended to lay more eggs around surrogate stems having Pr2S2 at the base rather than on the upper half of foliage. Observations of individual females performing preovipositional examining behaviors on Pr2S2-treated surrogate stems indicated that females tended to land on the upper portions of the foliage, but after landing, spent most of their time examining areas of soil and surrogate within 1 cm of the soil-surrogate foliage interface. Surrogate stems provide a realistic context for investigating effects of plant chemicals on host-acceptance behaviors.

    Notes: Abstract Decomposing onions at certain microbial successional stages produce potent volatile attractants and ovipositional stimulants of the onion fly,Delia antiqua (Diptera: Anthomyiidae). A reproducible source of these compounds was obtained by culturingErwinia carotovora var.carotovora (EC) on sterile onion tissue. In laboratory choice tests, EC-inoculated onion was more attractive thanKlebsiella pneumoniae (KP) cultured on onion, EC cultured on potato (a nonhost of onion fly), or the chemical synthetic baits dipropyl disulfide and an aqueous solution of 2-phenylethanol and pentanoic acid. Onion flies were mildly attracted to potato after inoculation with EC, but females did not accept EC-inoculated potato for oviposition. This work emphasizes that sources of semiochemicals may need to be defined microbiologically as well as physically and chemically.

    Notes: B-lymphocyte colonies were grown in semi-solid agar from mouse spleen or lymph node cells in the presence of mercaptoethanol with or without added sheep red cells. High levels of colony-forming cells were present in the spleen or normal mice and nu/nu (athymic) mice but colony-forming cells were rare in the thymus and not detected in activated T-lymphocyte populations. Colony-forming cells were θ-negative and most exhibited Fc receptors. Most colony-forming cells had the sedimentation velocity of small lymphocytes, were non-adherent and had a buoyant density similar to B-lymphocytes. Colony-forming cells were radiosensitive (Do 60 rads) and sensitive to cortisone. Colony formation was potentiated by the addition of adherent spleen cells or peritoneal macrophages. It is concluded that most cells forming B-lymphocyte colonies are themselves characterisable as B-lymphocytes.

    Notes: The mouse egg is an ideal model for the kinetic study of neutral amino acid exchange transport in development in that it possesses strong exchange-mediated uptake/efflux with little other efflux. Exchange transport was examined by preloading eggs in unlabeled or labeled amino acid and then measuring subsequent amino acid fluxes. The existence of two L-like (leucine-preferring) systems, a low-affinity exchange system, and a high-affinity weakly exchanging system is suggested by the differences between preloaded and control eggs in uptake specificity and in the kinetic parameters of methionine uptake (respectively, Km = 220 and 69 μM, Vmax = 98 and 32 fmol/min/egg). The Vmax of exchange-mediated efflux of methionine is similar to that for uptake into preloaded eggs, but the Km is about an order of magnitude higher. Preloading in alanine and serine (preferred substrates, along with cysteine, of system ASC) decreased the subsequent uptake of radiolabeled methionine, suggesting some ASC activity in the unfertilized mouse egg. The plateau in accumulation of leucine and phenylalanine, characteristic of simple exchange (mediated by system L), is not shared by methionine, suggesting the existence of another system (possibly an A, or alanine-preferring, system) for the transport of methionine. Amino acid transport in the mouse egg is more complex than previously thought, the egg possessing four interacting systems for the transport of the neutral amino acids.
