Notes: Summary Graded degrees of micrencephaly were produced in the progeny of rats given 0, 14, 22, or 30 mg methylazoxymethanol acetate (MAM Ac) per Kg b.wt. on gestation day (GD) 15. Brains of animals from five litters were examined at each dose level. Golgi-Cox-stained sections prepared from brains of progeny of rats given 25 mg MAM Ac/kg on GD 15 were also evaluated for four micrencephalic and four control animals. Each increase in dosage of MAM Ac resulted in significantly greater loss in brain weight in the progeny. The largest effect was on the telencephalon. In the low dose group the telencephalon, though noticeably smaller, was structurally normal. The two higher doses resulted in easily visualized neuropathologic lesions in both the cerebral cortex and hippocampus. The external layers of the cerebral cortex were most severely affected. Layers V and VI were much better preserved but measurements on pyramidal cells in layer VI in the Golgi-Cox-stained sections showed them to have significantly fewer dendrites and spines than the normal animals. In addition to an intrinsic disorganization of the neocortex, periventricular nodular heterotopias and hippocampal ectopias were common. All the structural anomalies were most severe at the highest dose, though they were also readily apparent at the intermediate dose. The findings suggest to us that MAM Ac treatment during gestation not only produces an acute lesion resulting in the destruction of many proliferating neuroblasts, but that many of the surviving neuroblasts may sustain a chronic lesion altering their subsequent development.

Notes: Summary Rats with bilateral anterior and posterior hippocampal lesions (APH), anterior hippocampal lesions (AH), and controls were trained on CRF for water, and then switched to FR-20. Both hippocampal groups adapted more readily than controls to the FR-20 schedule and maintained significantly higher rates for 45 days. There were no significant differences on FR-20 between the lesion groups. Following FR-20, the rates of all groups were equalized by training on FR-30. During subsequent extinction, the APH group made significantly more responses than both the AH and control groups, while AH and control groups were indistinguishable. The acquisition results were interpreted as indicating increased emotional reactivity to delay of anticipated reward in both hippocampally lesioned groups. The extinction results suggested that APH, but not AH, lesions also result in increased perseveration.

Notes: Abstract We employ a technique which has been used for in vitro studies of the vascular endothelium for obtaining flat preparations of different ocular structures such as corneal endothelium, epithelium of cornea, iris, ciliary body, lens and retinal pigment epithelium. The described method is relatively simple and allows evaluation of the overall state of these cellular sheets. After staining of the preparations with silver nitrate, we found intercellular lines to be present in all specimens studied except in the outer layers of the corneal epithelium. This method could be useful for in vitro studies of different pathological conditions of ocular tissues, both in animal models and in enucleated human eyes.

Notes: A mechanical and histological evaluation of LTI pyrolytic carbon implants was undertaken to determine the effect of various surface treatments on the retention characteristics of the implants. Five types of surfaces were evaluated, including as-deposited, fine grit-blasted, coarse grit-blasted, ground, and plasma oxygenated. The four surface treatments were chosen in an attempt to emulate the morphology of the as-deposited implants. The implants were evaluated in vivo by placement transcortically in the femora of adult mongrel dogs for periods of 12 to 24 weeks. Although the as-deposited implants exhibited the greatest interface strength at 12 weeks the results of mechanical testing after 24 weeks implantation indicated no statistically significant difference among the interface strength values or among the interface stiffness values of the implants. The histologic response of the implants was similar; while all implants exhibited areas of direct implant-bone apposition, the as-deposited implants exhibited this behavior to the greatest extent. Thus the ability to duplicate the biological response to the as-deposited LTI carbon surface appears possible by one or more of the treatments evaluated.
