Notes: Spectrophotometric studies of nickel-transfusion gelatin complex in presence of increasing amounts of various metal ions viz., Cu++, Co++, Zn++, Cd++ and Pb++ were undertaken to know the binding of these metal ions to the protein under biuret conditions. Studies carried out at pH 12.0 indicated that these metal ions combined with nitrogen of the peptide linkage replacing nickel from its complex. The amount with which a particular metal ions lower the extinction coefficient of the nickel complex may be taken as a measure of the extent of its combination to the protein, and the relative decrease in the extinction coefficient by equal amounts of various metal ions has been interpreted in terms of their order of affinities for the nitrogen of the peptide linkage.

Notes: Sol-gel transformation of mixed gels of silica, alumina and ferric oxide has been studied by determining the variations in pH with time of dialysis. For silica-alumina mixtures a pH of 2.9 is invariably reached irrespective of the alumina content of the mixtures. The amount of alumina present in the mixture influences the time of setting of the gel to a very great extent and it decreases with increase of the alumina content of the mixtures. These results have been explained in light of chemical interaction between the two gels to give aluminosilicates as well as in terms of controlled coagulation phenomenon with alumina behaving as a foreign electrolyte for bringing about the gelation of silicic acid. The results of silicic acid, ferric oxide gel forming mixtures are altogether different. In this case the time of gelation increases with the increase in the amount of ferric oxide in the gelation mixture, while a pH of 4.2 is reached irrespective of the amount of ferric oxide present. No evidence for the formation of ferric-silicate is obtained nor the gelation of these mixed sols can be explained in terms of controlled coagulation. The colloidal silica appears to act as protective colloid for the highly acedic ferric oxide sol. Viscometric and pH metric studies on the addition of alumina sol to mixtures of ferric oxide and silica sols indicate that alumina competes with ferric oxide in the formation of mixed gels. Here, too a pH of 2.9 is invariably reached irrespective of the alumina content in the mixed gels.

Notes: Spectrophotometric studies of mixtures containing Cr(iii) and bovine serum albumin were carried out to know the binding of chromic ion to the serum protein. Experiments performed at varying metal: protein ratio and at different pH values gave a maximum at 580 mμ. An inflexion was obtained in molar extinction coefficient-pH curves at a pH value corresponding to the pK values of carboxyl groups. All these go to prove that the carboxyl groups are likely to be the Prinzipal sites through which the fixation of chromic ions have been affected. Experiments carried out in the neutral pH region gave no evidence regarding the binding of metal ions to the imidazole groups of bovine serum albumin. The lesser reactivity of bovine serum albumin in comparison with that of transfusion gelatin (previously studied) has been attributed to the lack of effective multipoint sites, due to its folded structure, so as to fix several chromium atom intramicellarly.

Notes: Absorption measurements of mixtures of copper-transfusion gelatin and nickel-transfusion gelatin were carried out with changing metal: protein ratio and at different pH-values (10-12.4). The addition of minute quantities of protein to cupric chloride gave a violet colour, and gradually increasing amounts of protein brought a change in colour from violet to red. The violet complex absorbed maximum light at 550 mμ whereas red complex absorbed at 525 mμ. The shift in λ max. with a change in metal: protein ratio has been ascribed to the formation of various types of chelate structure during the copper-transfusion gelatin biuret reaction. On the other hand nickel-transfusion gelatin biuret reaction has been characterised by the formation of only one type of complex corresponding to the violet complex of copper. λ max. for nickel complex was found to lie in the vicinity of 430 mμ.

Notes: Spectrophotometric studies of mixtures containing Cr(iii) and transfusion gelatin were undertaken to know the binding of chromic ion to the protein. Experiments carried out between pH 3:7 and 5.8 had indicated the binding of the metal through the carboxyl groups of the protein. The extent of metal binding was found to be dependent upon metal-protein ratio, maximum binding taking place in the ratio metal to protein as 42:1.

Notes: The influence of dyes on the gelation of ferric hydroxide sol has been studied with the help of viscometric, pH metric and cataphoretic methods. From these studies it has been concluded that out of the four dyes it is only alizarin sulphonic acid which brings about appreciable change in the above physical properties. The results have been interpreted in terms of structural changes in the dye molecule. Positive adsorption is found to take place in all cases except malachite green which is negatively adsorbed on the gel. In the case of alizarin sulphonic acid the adsorption data do not fit in well either in Langmuir or Freundlich adsorption isotherms.
