    Notes: Summary Intercellular junctions within the villous stroma and the cytotrophoblastic layer of the human full term placenta were investigated using thin sectioning and freeze-fracturing. Numerous maculae adherentes (desmosomes) were found between the cytotrophoblast cells and the syncytiotrophoblast. This junction type was also seen connecting adjacent cytotrophoblast cells. Large gap junctions were frequently observed in contact areas of perikarya or at processes of adjacent fibroblasts. They often exhibited a peculiar pattern of their particles on the P-face of the membrane. Small rows of junctional particles were found on the P-faces of interconnected smooth muscle cells and gap junctions frequently bridged myoendothelial and interendothelial contact zones. The significance of the junctional complexes is discussed in relation to functional systems within the villous stroma of the human full term placenta.

    Notes: Summary The capillaries in the atrioventricular node and atrioventricular bundle of the heart (mammals) were investigated by thin sections and freezeetch replicas. Fenestrated endothelial cells were found to occur in all mammals examined in this study. These fenestrations (500±50 Å diameter) are bridged by a diaphragm and occur in relatively small sieve areas. In contrast to the AV-node and AV-bundle, the non-specialized myocardium is supplied by continuous capillaries. The presence of the fenestrated capillary in the conduction system of the heart may serve to increase both permeability and passive transport capacity.

    Notes: Summary The distribution of ionic lanthanum (La3+) was studied after perfusion of the isolated rat heart with a buffered salt solution containing lanthanum chloride. We have demonstrated that the localization of lanthanum differed from that shown in previous studies in which hearts were exposed to ionic or colloidal lanthanum during or after fixation. La3+ passed rapidly through the intercellular cleft between endothelial cells. There was relatively little transport by micropinocytosis and no evidence of transendothelial passage through the cytoplasm. Perfusion with a solution containing 1 to 15 mM LaCl3 resulted in localization of La3+ on the entire cytoplasmic membrane system of muscle cells, i.e.: the non-specialized parts of the muscle cell membranes were stained by La3+ as well as the membranes of micropinocytotic vesicles, of the T-system and of the intercalated disc. The cytoplasmic leaflets of gap junctions were especially stained. However, the cell membranes of non-muscle cells were free of lanthanum precipitates. Freeze-fracturing of muscle cells showed concomitant alteration in the distribution of membrane-associated particles. The gap junction membranes also showed changes in particle aggregation. Intracellular binding of La3+ was restricted to muscle cells. Lanthanum was found in the cell nucleus associated with heterochromatin, in the nucleolus, and along the nuclear membrane. The myofibrils were stained mainly over the I-band.

    Notes: Summary Isolated rat hearts were perfused with a balanced electrolyte solution containing 1000mM glycerol for 15min and then perfused with normal electrolyte solution for up to 32 min. The perfusion with hypertonic glycerol solution and subsequent washout is termed “glycerol treatment”. Initially, glycerol removal causes swelling and rupture of the T-system in ventricular myocardial cells which correlates temporally with a period of cardiac arrest. Contractility returns during further glycerol removal and concomitant recovery of the T-system is observed. Atomic absorption spectometry and neutron activation analysis were used to measure ventricular sodium, potassium and calcium ion content. There is no apparent correlation between changes in ion content and cardiac arrest or recovery. The water movements were calculated from wet weight, dry weight and inulin space, and confirmed by morphometric analysis of extracellular and intracellular space. It is suggested that the swelling and rupture of the T-system is due to the rapid water movements that were observed during the onset of glycerol removal. Ultrastructural analysis of glycerol-treated atrium from the same hearts shows damage of mitochondria and of the L-system and intracellular edema. The structural changes are correlated with a loss of atrial contraction. As in ventricular myocardium, resumption of contraction is associated with an almost complete recovery from ultrastructural damage.
