    Notizen: The slow Wallerian degeneration phenotype, WldS, which delays Wallerian degeneration and axon pathology for several weeks, has so far been studied only in mice. A rat model would have several advantages. First, rats model some human disorders better than mice. Second, the larger body size of rats facilitates more complex surgical manipulations. Third, rats provide a greater yield of tissue for primary culture and biochemical investigations. We generated transgenic WldS rats expressing the Ube4b/Nmnat1 chimeric gene in the central and peripheral nervous system. As in WldS mice, their axons survive up to 3 weeks after transection and remain functional for at least 1 week. Protection of axotomized nerve terminals is stronger than in mice, particularly in one line, where 95–100% of neuromuscular junctions remained intact and functional after 5 days. Furthermore, the loss of synaptic phenotype with age was much less in rats than in mice. Thus, the slow Wallerian degeneration phenotype can be transferred to another mammalian species and synapses may be more effectively preserved after axotomy in species with longer axons.

    Notizen: Abstract Nitric oxide (NO) is an important mediator in the relaxation of cavernosal smooth muscle. The present study examines the existence and location of the constitutive isoform eNOS (endothelial NO synthase) accompanying the already substantiated neurogenic NOS (nNOS) in the human corpus cavernosum of men with and without erectile dysfunction. Activities of NOS enzymes were examined in specimens of 11 potent and nine long-term impotent patients by means of light and electron microscopy using NADPH-diaphorase staining and immunohistochemical eNOS-specific, smooth muscle actin-specific and nNOS-specific markers. Cavernosal smooth muscle shows a distinct expression of eNOS. In contrast to the weaker expression of eNOS and nitrinergic innervation found in larger veins, the small intracavernosal helicine arteries express large quantities of eNOS and possess a dense nitrinergic innervation. Long-term impotent patients display a broad heterogeneity in eNOS expression and nitrinergic innervation while no overall correlation between NOS expression and erectile function was observed. The expression of eNOS indicates eNOS as a main source of NO alongside nNOS. The differentiated localization of eNOS supports at least a role of this isoform in vascular regulation.

    Notizen: Summary Perichromatin granules, nematosomes (threadlike bodies), or concentric lamellar bodies were rarely observed in profiles of principal ganglionic neurons in the untreated rat superior cervical ganglion. They were more frequenlty encountered in these neurons following long-term activation of the sympathetic nervous system by intermittent immobilization (6 to 18 12-h periods). The increased number of the described nuclear and cytoplasmic structures following immobilization is documented by morphometric data and discussed in relation to specific chronic neuronal hyperactivity.

    Notizen: Abstract Based on the idea of differentiation-related changes in the glycosylation pattern of neurons, the expression of two cell surface oligosaccharide epitopes, N-acetyl-lactosamine (NALA), and its sulpho-glucuronyl derivative (HNK-1), was studied, by immunohistochemistry and Western blot experiments, in the developing chick retina beginning on day 2 of incubation (E2) until day 18 post-hatching. NALA was detectable on neuroepithelial cells as soon as the primary optic vesicles formed, and this pattern continued until E3. During subsequent retinal development NALA expression became progressively restricted in concert with the appearance of postmitotic neurons as revealed by neurite outgrowth, and with the formation of synaptic contacts until it disappeared at the end of the incubation period. The pattern of NALA expression was the inverse of HNK-1 which was detected for the first time at E3 on postmitotic ganglion cells accumulating at the vitreal surface. The number of HNK-1+ cells steadily increased until around E10, when the entire neural epithelium was labelled. Synchronously to synaptogenesis, most neurons lost their HNK-1 immunoreactivity. At the time of hatching the adult-like pattern was found, characterised by subpopulations of labelled horizontal, bipolar, amacrine, and ganglion cells. Immunoblot experiments demonstrated transient NALA glycosylation of protein bands, partially identical in their apparent molecular weight to those proteins with HNK-1 glycosylation. The observed temporospatial changes in the glycosylation patterns of distinct proteins during retinal development suggest NALA as a suitable marker for neuronal proliferation, and HNK-1 for differentiation and establishment of final synaptic configuration.

    Notizen: Abstract Fibrosis, contraction and opacification of the posterior lens capsule after extracapsular cataract extraction, is a frequent complication following cataract surgery. In these cases, cellular proliferation occurs along an intact posterior capsule. We report a case of fibrosis adjacent to the anterior lens capsule, where cellular proliferations and collagen production completely sealed the anterior capsulotomy three months after a routine extracapsular cataract extraction with implantation of a posterior chamber lens. The fibrosis led to contraction of the remainder of the anterior capsule, significantly reducing vision. Examination of the excised material by light and electron microscopy, and immunohistochemistry, revealed cells with features compatible with fibrocytes that did not stain positive for cytokeratin. The cells were situated in a dense collagen matrix. An anterior capsulotomy that is too small prevents sufficient removal of lens epithelium, and may be a risk factor for this complication.

    Notizen: Abstract Prolonged postoperative hypotony is a severe complication frequently associated with trabeculectomies performed with a single perioperative application of Mitomycin C. We performed an animal experiment using 8 pigmented rabbits applying different concentrations of Mitomycin C, ranging from 0.05 to 1.0 mg/ml, under the conjunctiva on the intact sclera for 5 minutes. The eyes were examined by light and electron microscopy after 4 weeks. The nerves within the ciliary body in areas adjacent to the region of treatment showed toxic effects related to the concentrations of Mitomycin C applied. In cases where small concentrations were used, only the unmyelinated nerves were damaged with the myelinated remaining intact. In eyes treated with high concentrations, both groups of nerves exhibited severe signs of destruction. This effect is likely to compromise the production of aqueous humor in rabbits, and possibly also in humans.

    Notizen: Summary In the Ham-Wistar rat, a paraganglion was found within the vagus nerve at the site of branching of the recurrent laryngeal nerve. Due to its location the name “paraganglion supracardiale vagi” is suggested. Fluorescence microscopy of the paraganglionic cells displays an intense yellow-green fluorescence indicating the presence of biogenic amines. Ultrastructurally, chief cells containing dense-core vesicles form three kinds of synaptic contacts (afferent, efferent and reciprocal) with enlarged, mitochondria-rich nerve endings.
