    Notizen: It is possible to replace the CaMV (cauliflower mosaic virus) ORF (open reading frame) II with foreign sequences without interfering with virus viability. Such recom-binants can induce the synthesis of substantial amounts of a foreign protein in infected plants and confer new properties to these plants. However, so far only three genes have been successfully cloned and expressed in this way. The expression mechanism of CaMV demands precise replacement of ORF II and probably certain structural features of the viral 35S RNA, which should not be disturbed by inserted sequences. Since these features are largely unknown, it cannot at present be pre-dicted whether an insert will be tolerated. It is more likely that larger inserts will disturb the viral gene expression mechanism than smaller ones.

    Notizen: Abstract  Fungal wheat (Triticum aestivum) diseases greatly affect crop productivity and require the economically and ecologically undesirable application of fungicides in wheat agriculture. We have generated transgenic wheat plants constitutively expressing an antifungal barley-seed class II chitinase. The transgene was stably expressed and the chitinase properly localized in the apoplast of the transgenic lines. The engineered wheat plants showed increased resistance to infection with the powdery mildew-causing fungus Erysiphe graminis.

    Notizen: Abstract  Phenotypically normal, transgenic wheat (Triticum aestivum, var. Frisal) plants expressing a barley seed ribosome-inactivating protein (RIP) were produced. Expression was controlled by an intron-enhanced cauliflower mosaic virus 35S promoter and has been completely stable over four generations so far, possibly due to matrix-associated regions (MARs) that flank the transgenes. An engineered fusion to a signal peptide derived from the barley seed ß-1,3-glucanase caused the transport of RIP to the apoplast. Activity of the accumulated protein could be shown by significant inhibition of a rabbit reticulocyte transcription/translation system. Plants expressing high levels of RIP were protected only moderately or not at all against infection by the fungal pathogen Erysiphe graminis.

    Notizen: Summary Indica-type rice provides the staple food for two billion people in Third World countries. Several problems involved in the stable and sustained production of high quality food cannot be solved by traditional breeding. Methods have been established for gene transfer to Indica rice breeding lines to study possible contributions from genetic engineering. Experiments are in progress on the development of transgenic resistance towards Yellow Stem Borer, resistance towards Rice Tungro Virus, accumulation of provitamin A in the endosperm, increase of essential amino acids in the endosperm such as lysine, cysteine and methionine and resistance towards fungal pests such as Rice Blast and Sheath Blight. Transgenic clones from Indica rice breeding lines have been recovered from several of the approaches mentioned, some of which have been regenerated to plants.
