Notes: Intramolecular 1,3-Dipolar Cycloadditions of Diaryl-nitrile-imines Generated from 2,5-Diaryl-tetrazolesAlkenyl-substituted diaryl-nitrile-imines - generated by photolysis or thermolysis of alkenyl-substituted 2,5-diaryl-tetrazoles - undergo a regioselective intramolecular [2 + 3] cycloaddition to yield new heterocyclic compounds, e. g. fused 2-pyrazolines. With alkinyl derivatives, the corresponding pyrazoles have been formed. UV evidence is given for the intermediate nitrile-imine at - 190°. The latter can be trapped using an excess of carboxylic acid (UV evidence for a new intermediate at - 120°). In this case, the corresponding rearranged addition product N′-acyl-N′-aryl-benzohydrazide is isolated in good yield.

Notes: Synthesis of 4,4-Disubstituted 1,3-Thiazol-5(4H)-thionesAn easy synthesis for the 1,3-thiazol-5(4H)-thiones 5, a class of heterocycles which have hitherto only been available with difficulty, is described. Reaction of 3-amino-2H-azirines 25 with thiocarboxylic acids at 0° yields monothiodiamides of type 20 (Scheme 6) which, on treatment with Lawesson reagent at 100°, undergo thiation and cyclization to give 5 in good yield.

Notes: 1,4-Dithiafulvenes, Products of the Reaction of 4,4-Disubstituted 1,3-Thiazol-5(4H)-thiones and Acetylenic CompoundsOn heating in toluene, 4,4-disubstituted 1,3-thiazol-5(4H)-thiones 1 and acetylenecarboxylates or acetylenecarbonitriles 2 undergo a cyclosubstitution reaction to yield 2-methylidene-1,3-dithiol derivatives 3 (1,4-dithiafulvenes) and a nitrile. Further heating of 3a and 3b in the presence of excess 2a leads to the isomeric 2,3-dihydrothiophene-2-thiones 4a and 4b, respectively. The benzodithiafulvene 14 has been formed in a similar reaction from 1a and in situ generate benzyne.

Notes: Conformational Analysis of Tripeptide Models: The Influence of α,α-disubstituted α-Amino Acids on the Secondary Structure. X-Ray Analysis and Conformational Energy CalculationsThe X-ray analysis of tripeptide Z-Ile-Val(2-Me)-benzocaine (1f) reveals the presence of a type-III β-turn. Moreover, MMP2 calculations on tripeptides, e.g. Z-Ile-Aib-benzocaine (1c), Z-Ile-D-Val(2-Me)-benzocaine (1g), Z-Ile-Gly(2,2-Pr2)-benzocaine (1h), Z-Ile-Gly-benzocaine (1a), and 1f, fit well into the frame of NMR and CD investigations. They allow considerations on the relative stability of different types of β-turns depending on the peptide sequence, e.g. the kind of α,α-disubstituted amino-acid moieties.

Notes: Intramolecular 1,3-Dipolar Cycloaddition Reactions of Alkenyl-substituted 3,4-DiarylsydnonesThe 3,4-diarylsydnones 1 and 12a-e with an allyl or alkenyloxy substituent on the N(3)-phenyl group have been synthesized by classical methods starting from 2-allylaniline and the corresponding alkenyloxyanilines, respectively (Schemes 2 and 3). Whereas the allyl-substituted sydnone 1 undergoes an intramolecular 1,3-dipolar cycloaddition at room temperature in solution to yield 13 (Scheme 4), the alkenyloxy-substituted sydnones 12a-e are thermally stable. On irradiation of 1 and 12a-d, formation of the fused dihydropyrazoles 2 and 14a-d (Schemes 1 and 5) is observed. In the case of 12d, the yield of 14d is very low, and the symmetric 1,2,3-triazole 15a has been isolated as the main product. A very likely reaction mechanism for the formation of the photoproducts involves decarboxylation of the sydnones to give a nitrile-imine which undergoes an intramolecular 1,3-dipolar cycloaddition onto the C=C bond.

Notes: Carbophilic Additions of Organocuprates and 1,3-Thiazole-5(4H)-thionesOrganocuprates and 1,3-thiazole-5(4H)-thiones 1 react in THF at 0° via carbophilic addition onto the C—S bond to give 4,5-dihydro-1,3-thiazole-5-thiols 3 (Scheme 3). This observation is in marked contrast to the previously described reaction of organolithium compounds and 1, which undergo a thiophilic addition onto the exocyclic S-atom. As an exception, treatment of the 1,3-thiazole-5(4H)-thione 1a with tert-butyl cuprate leads to 7a (Scheme 3).

Notes: 3-(Dimethylamino)-2,2-dimethyl-2H,-azirine as an α-Aminoisobutyric-Acid (Aib) Equivalent: Cyclic Depsipeptides via Direct Amid CyclizationIn MeCN at room temperature, 3-(dimethylamino)-2,2-dimethyl-2H-azirine (1) and α-hydroxycarboxylic acids react to give diamides of type 8 (Scheme 3). Selective cleavage of the terminal N,N-dimethylcarboxamide group in MeCN/H2O leads to the corresponding carboxylic acids 13 (Scheme 4). In toluene/PhSH, phenyl thioesters of type 11 are formed (see also Scheme 5). Starting with diamides 8, the formation of morpholin-2,5- diones 10 has been achieved either by direct amide cyclization via intermediate 1,3-oxazol-5(4H)-ones 9 or via base-catalyzed cyclization of the phenyl thioesters 11 (Scheme 3). Reaction of carboxylic acids with 1, followed by selective amide hydrolysis, has been used for the construction of peptides from α-hydroxy carboxylic acids and repetitive α-aminoisobutyric-acid (Aib) units (Scheme 4). Cyclization of 14a, 17a, and 20a with HCI in toluene at 100° gave the 9-, 12-, and 15-membered cyclic depsipeptides 15, 18, and 21, respectively.

Notes: The allenecarboxylates 1a,b and allenic lactones 4a,b undergo thermally induced (1,3) H Shifts in the presence of Co2(CO)8. The non-isolated 1,3-dienes 2a,b react further affording the Diels-Alder Adducts 3a,b Scheme 1 in high yields. These adducts were not formed in the case of the 2-vinybutenolides 5a,b. On irradiationin the presence of Co2(CO)8 or Mn2(Co)10, the studied allenes reacted in a different manner, yielding either cyclization products 7 and 8 (Scheme 3) or products 9 and 10, formed via H abstracton and solvent addition (Schemes 4 and 5).

Notes: Thermal Generation and Reactions of (Benzylthio)-and (Arylthio)-Substituted Nitrile YlidesThermolysis of 4-(benzylthio)- and 4-(arylthio)-1,3-oxazol-5(2H)-ones 6, at 110-155° in the presence of dipolarophiles with activated C≡C, C=C, C=O, C=S, and N=N bonds, led to 5-membered cyclo-adducts and CO2 (cf. Schemes 3, 5-7). Heating 6a and 6c in the presence of ethyl propiolate yielded ethyl quinoline-3-carboxylate (19) and ethyl pyridine-3-carboxylate(22), respectively (cf. Scheme 8). These results are rationalized on the basis of the intermediate formation of thio-substituted nitrile ylides of type 7 (cf. Scheme 2), which undergo regioselective 1,3-dipolar cycloadditions with reactive dipolarophiles. In the absence of such a dipolarophile, the nitrile ylides isomerize via a [1,4]-H shift to give 2-aza-1,3-butadienes of type 20. The latter are trapped in a Diels-Alder reaction with ethyl propiolate (cf. Scheme 8).
