Notes: The morpholine ring of the title dione, C13H15NO3, shows a boat conformation that is distorted towards a twist-boat, with the boat ends being the two Csp3 atoms of the ring. The benzyl substituent is in the favoured `exo' position. In the monothione derivative, (±)-6-benzyl-3,3-dimethyl-5-thioxomorpholin-2-one, C13H15NO2S, this ring has a much flatter conformation that is midway between a boat and an envelope, with the dimethyl end being almost planar. The orientation of the benzyl group is `endo'. The dithione derivative, (±)-6-benzyl-3,3-dimethylmorpholine-2,5-dithione, C13H15NOS2, has two symmetry-independent molecules, which show different puckering of the morpholine ring. One molecule has a flattened envelope conformation distorted towards a screw-boat, while the conformation in the other molecule is similar to that in the monothione derivative. Intermolecular hydrogen bonds link the molecules in the three compounds, respectively, into centrosymmetric dimers, infinite chains, and dimers made up of one of each of the symmetry-independent molecules.

Notes: The tetrathiane ring of the title compound, C26H16S4, has a chair conformation and the molecule has approximate C2 symmetry. Each of the two fluorene ring systems is virtually planar, with the ring planes intersecting at an angle of 67.58 (5)°. This novel compound has been formed as a side product from the treatment of 9H-fluorene-9-thione with methyl N-[(benzylidene)phenyl]glycinate in the presence of LiBr and 1,6-diazabicyclo[5.4.0]undecane.

Notes: Addition Reaction of 3-Dimethylamino-2,2-dimethyl-2H-azirine with Phenylisocyanate and Diphenylketene3-Dimethylamino-2,2-dimethyl-2H-azirine (1a) reacts with carbon disulfide and isothiocyanates with splitting of the azirine N(1), C(3)-double bond to give dipolar, fivemembered heterocyclic 1:1 adducts. In some cases, these products can undergo secondary reactions to yield 1:2 and 1:3 adducts. In this paper it is shown that the reaction of 1a with phenylisocyanate also takes place by cleavage of the N(1), C(3)-bond, whereas with diphenylketene N(1), C(2)-splitting is observed. The reaction of 1a and phenylisocyanate in hexane at room temperature yields the 1:3 adduct 2 in addition to the trimeric isocyanate 3 (Scheme 1). A mechanism for the formation of 2 is given in Scheme 5. Hydrolysis experiments with the 1:3 adduct 2, yielding the hydantoins 4-6 and the ureas 7 and 8 (Schemes 3 and 5), show that the formation of this adduct via the intermediates d, e and f is a reversible reaction. The aminoazirines 1a and 1b undergo an addition reaction with diphenylketene to give the 3-oxazolines 14 (Scheme 8), the structure of which has been established by spectral data and oxidative degradation of 14a to the 3-oxazolin-2-one 15 (R1 — R2 — CH3, Scheme 9).

Notes: Reactions of valencepolaromeric ketenes of mesoionic heterocyles with 3-dimethylamino-2H-azirinesReactions of the 3-dimethylamino-2H-azirines 1a and 1b with the mesoionic oxazole 5 and the mesoionic dithiole 6 in acetonitrile at room temperature yield the 1:1 adducts 11, 12, 19 and 20, respectively (Schemes 5 and 8). These products can be formulated as adducts of the aminoazirines and the ketenes 5a and 6a, which are valence polaromeric forms of the mesoionic heterocycles 5 and 6 (Scheme 2).The structure of the adducts has been elucidated by spectral data and their comparison with the data of (Z)-11, the structure of which has been established by X-ray [19]. Oxidation of the 1:1 adducts with KMnO4 in a two-phase system yields 4-dimethylamino-3-oxazolin-2-ones (cf. Scheme 6) by clevage of the exocyclic C,C-double bond. A mechanism for the formation of the adducts is given in Scheme 9: Nucleophilic attack of 1 on the ketene leads to a primary adduct of type a, which undergoes clevage of the former N(1), C(2)-azirine bond to give adducts of type 11 or 19. The N(1), C(2)-ring opening of 1a in the reaction with ketenes contrasts with the N(1), C(3)-opening of 1a in the addition with, for instance, isothiocyanates. These different ring openings are explained by the difference in nucleophilicity of the heteroatoms X and Y in a′ (Scheme 10).

Notes: Synthesis of 3-Dimethylamino-3a,4,5, 7a-tetrahydro-1H-isoindol-1-ones by Intramolecular Diels-Alder ReactionThermolysis of N2-acylamidines, the acyl group of which derives from an α,β,γ,δ-unsaturated carboxylic acid (2, 5-7), yields 3-dimethylamino-3a,4,5,7a-tetrahydro-1H-isoindol-1-ones (3,8-10, Schemes 1 and 3) in 63-78%. Only the thermodynamically controlled cis-fused ring system is formed. The starting materials are readily available by the reaction of 3-dimethylamino2H-azirines (1 and 4) and carboxylic acid chlorides.

Notes: Selective Modification of the Terminal Amide Group of Linear Polyamides via 2-Oxazolin-5-ones as IntermediatesTreatment of aqueous or alcoholic solutions of diamides of type 2 with HCl leads to the formation of amide-acids and amide-esters of type 3 (Scheme 1 and Table). It has been shown, that 2-oxazolin-5-ones of type 4 are intermediates of this selective transformation of the disubstituted terminal amide group. The selectivity of the transformation is demonstrated by the reaction sequence shown in Scheme 3. No selectivity has been observed in the acid-catalyzed hydrolysis of triamide 9 with a monosubstituted terminal amide group (Scheme 4). Hydrolysis of the optically active dipeptide derivatives (+)-(L)-13 and (+)-(L)-15 with HBr in nitromethane at 60-80° yields the pure enantiomer (+)-(L)-14 and (+)-(L)-16, respectively (Scheme 5), i.e., no racemization takes place under the reaction conditions. These results show the usefulness of the dimethylamide group as a protecting group for carboxylic acids for example in the peptide synthesis.

Notes: Reactions of 3-Dimethylamino-2,2-dimethyl-2H-azirine with NH-Acidic Heterocycles; Synthesis of 4H-ImidazolesIn this paper, reactions of 3-dimethylamino-2,2-dimethyl-2H-azirine (1) with heterocyclic compounds containing the structure unit CO—NH—CO—NH are described. 5,5-Diethylbarbituric acid (5) reacts with 1 in refluxing 2-propanol to give the 4H-imidazole derivative 6 (Scheme 2) in 80% yield. The structure of 6 has been established by X-ray crystallography. Under similar conditions 1 and isopropyl uracil-6-carboxylate (7) yield the 4H-imidazole 8 (Scheme 3), the structure of which is deduced from spectral data and the degradation reactions shown in Scheme 3. Hydrolysis of 8 with 3N HCl at room temperature leads to the α-ketoester derivative 9, which in refluxing methanol gives dimethyl oxalate and 5-dimethyl-amino-2,4,4-trimethyl-4H-imidazole (10). On hydrolysis the latter is converted to the known 2,4,4-trimethyl-2-imidazolin-5-one (11) [6]. Quinazolin-2,4 (1H, 3H)-dione (12) and imidazolidinetrione (parabanic acid, 14) undergo with 1 a similar reaction to give the 4H-imidazoles 13 and 15, respectively (Schemes 4 and 5).In Scheme 6 two possible mechanisms for the formation of 4H-imidazoles from 1 and heterocycles of type 16 are formulated. The zwitterionic intermediate f corresponds to b in Scheme 1. Instead of dehydration as in the case of the reaction of 1 with phthalohydrazide [3], or ring expansion as with saccharin and cyclic imides [1] [2], f, undergoes ring opening (way A or B). Decarboxylation then leads to the 4H-imidazoles 17.

Notes: Reaction of 3-Dimethylamino-2,2-dimethyl-2H-azirine with 6-Methyluracil; Crystal Structure of the ProductsThe reaction of 3-dimethylamino-2,2-dimethyl-2H-azirine (1) with 6-methyl-uracil (4) in 2-propanol at 80° yields the 4H-imidazoje derivative 5 as the main product. This reaction is similar to the previously reported ones with heterocyclic compounds containing the sequence NH—CO—NH—CO. In the presence of water, 5 is easily hydrolyzed to 6. The structures of 5 and 6 have been established by X-ray crystallography.

Notes: Spiro 3-Oxazolines from the 1,3-Dipolar Cycloaddition of Benzonitrilio-2-propanide and 1,4-QuinonesOn irradiation with light of wavelength 290-350 nm, 2,2-dimethyl-3-phenyl-2H-azirine (1b) reacts with 1,4-naphthoquinone to give the 1H-benzo [f]isoindol-4,9-dione (11) (Scheme 3) via cycloaddition of the benzonitrilio-2-propanide (2b) onto the quinone C, C-double bond. With 2-methyl- and 2,3-dimethyl-1,4-naphthoquinone, the nitrile ylide 2b undergoes cycloaddition preferentially onto the C, O-double bond of the quinone, leading to spiro-oxazolines 12 and 14 (Scheme 4). Steric as well as electronic effects can be discussed to explain the observed site selectivity of the cycloaddition. With the 1,4-benzoquinones 15a, 15b, 15d and 15f, nitrile ylide 2b undergoes the 1,3-dipolar cycloaddition exclusively onto the C, O-double bond. The corresponding spiro-oxazolines have been isolated in 17-32% yield. This contrasts with the previously reported results with benzonitrilipo-phenylmethanide (2a), which undergoes cycloaddition to the C, C-double bond of 1,4-benzoquinones (cf. [1]). This difference in the site selectivity of the 1,3-dipolar cycloaddition can be explained with Houk's concept of LUMO-polarization, that is, the stronger nucleophilic dipol 2b polarizes the LUMO of a α,β-unsaturated carbonyl compound more efficient than the less nucleophilic 2a. This leads to a preference of the cycloaddition to the C, O-double bond in the case of 2b.With 2,3-dimethyl- (15c) and 2,3,5,6-tetramethyl-1,4-benzoquinone (15e), nitrile ylide 2b undergoes C, O- as well as C, C-cycloaddition (Schemes 7 and 8).

Notes: Synthesis of 3,3-Dimethylperhydro-1,4-diazepin-2,5,7-triones from 3-Dimethylamino-2,2-dimethyl-2H-azirine and Malonic Acid MonoamidesReaction of the aminoazirine 1 and malonic acid monoamides 5 in CH3CN yielded triamides of type 6 (Scheme 2), which were transformed to the corresponding phenylthioates 9 by treatment of a solution of 6 and thiophenol in CH3CN with HCl (Scheme 4). Cyclization of 9 to give the 1,4-diazepin-2,5,7-trione of type 10 was achieved with NaH in toluene at about 90°. It has been shown that 2-oxazolin-5-ones are intermediates in the selective cleavage of the therminal amide function of 6 (Scheme 3).
