    Die Erythrozytenalterung als Modell für die pharmakologische Beeinflussung der Erythrozytenverformbarkeit (1979)

    Description / Table of Contents: Summary The methods to measure red cell flexibility are reviewed and own investigations reported based on red cell aging as a model for a physiological reduction in red cell flexibility. The measurement of this parameter was performed with whole-blood by means of filtration through filters with a pore diameter of 5 µm. The model of red cell aging and the method used for the measurement of red cell flexibility is suitable as a screening system to evaluate whether chemical compounds influence red cell flexibility. In two series of experiments pentoxifylline in concentrations of 10 µg and 20 µg/ml of blood and theophylline and prednisolone in concentrations of 96.6 µg or 20 µg/ml of blood, respectively, were investigated. For pentoxifylline a dose-dependent increase in red cell flexibility was observed, theophylline and prednisolone did not change this parameter in comparison to the control. The effect of pentoxifylline is probably related to an increase in red cell ATP content. Scanning electron microscopy revealed that this compound does not influence the outer membrane of the red cells.
    Notes: Zusammenfassung Es werden die Möglichkeiten der Messung der Erythrozytenverformbarkeit dargestellt und eigene Untersuchungen mit einer Methode beschrieben, welche die Erythrozytenalterung als Modell für eine physiologische Reduzierung der Erythrozytenverformbarkeit benutzt. Die Messung dieser Größe erfolgt in Vollblut mittels Filtration durch Filter mit Poren von 5 µm. Das Erythrozytenalterungsmodell und die Methode zur Messung der Erythrozytenverformbarkeit eignet sich für die routinemäßige Prüfung, ob chemische Substanzen die Erythrozytenverformbarkeit beeinflussen. In zwei Untersuchungsserien wurden Pentoxifyllin in Konzentrationen von 10 µg und 20 µg/ml Blut sowie Theophyllin in einer Konzentration von 96,6 µg/ml Blut und Prednisolon in einer Konzentration von 20 µg/ml Blut untersucht. Für Pentoxifyllin fand sich eine dosisabhängige Erhöhung der Erythrozytenverformbarkeit, unter Theophyllin und Prednisolon fanden sich keine signifikanten Änderungen gegenüber der Kontrolle. Der Effekt von Pentoxifyllin ist wahrscheinlich in einer Erhöhung des erythrozytären ATP-Gehaltes begründet. Rasterelektronenmikroskopische Untersuchungen zeigen, daß die morphologische Struktur der äußeren Erythrozytenmembran durch Pentoxifyllin nicht beeinflußt wird.

    Notes: Summary The capillary viscosimeter (Viscotimer) tested with different Ubbelohde-viscosimeters is suitable for measurements of plasmaviscosity; the limitting factor of the measuring method is the exact adjustment of the temperature. When testing the plate-cone-viscosimeter (Wells-Brookfield Microviscosimeter), we achieved scientifically adequate results only when using shear rates 230, 115 and 46 sec−1. An influence of different thrombocyte-concentrations on the viscosity of whole blood and plasma could not be detected. The flow capacity of whole blood and plasma is distinctly dependent on the temperature.

    Notes: Summary The measurement of red cell deformability (flexibility or fluidity) according to the method of filtration strongly depends on the suspension medium, the hematokrit, filtration pressure as well as osmolarity and is hard to differentiate over the influence of red cell aggregation.     Notes: Abstract Studies on smooth muscle cell differentiation and those on vascular development in mouse and humans have long been hampered by the lack of suitable markers. Here we describe a novel, large isoform of smoothelin, a structural protein of differentiated, contractile smooth muscle cells. The protein, which is highly conserved in mouse and humans, shows homology with other cytoskeleton-associated smooth muscle cell proteins and contains an actinin-type actin-binding domain. Northern blot analysis from various mouse organs identified short and long smoothelin mRNA forms, which exhibit distinct tissue expression patterns. The short form is highly expressed in visceral muscle tissues such as intestine and stomach and is not detectable in brain, while the long mRNA form is expressed in all vascularized organs. These results may provide new tools and approaches to study both smooth muscle cell differentiation and proliferative vascular disease.

It is shown that the sequences converge monotonously and the speed of convergence is better than linear.

    Description / Table of Contents: Summary 1. It was attempted to investigate the consequences of obesity and slimming on the adenohypophyse, the thyroid and the adrenal cortex in rats. 2. Young male rats were fattened and slimmed down again under the least traumatizing conditions. 3. By selection a strain of rats could be isolated more easy to be fattened and more difficult to be slimmed down. This was already obtained in three generations and still more enhanced in two further generations. 4. During fattening the adenohypophyse of rats of the original strain showed a histological picture suggestive of a resting condition. During slimming the thyreotropic and the corticotropic cells seemed stimulated. In animals of the selected strain degenerative changes of these cells were observed in consequence of a long-lasting fattening. 5. The thyroid and the adrenal cortex of rats of the selected strain appeared less active than those of animals of the original strain. Fattening is followed by a decreased activity of the thyroid and the adrenal cortex. This was more conspicuous in rats of the selected strain than in those of the original strain. 6. Possible predisposing influence of these changes of the endocrines on obesity are discussed.
    Notes: Zusammenfassung 1. Es wurde versucht, die Folgen der alimentär bedingten Verfettung und Abmagerung auf die Hypophyse, die Schilddrüse und die Nebenniere der Ratte zu ergründen. 2. Junge männliche Ratten wurden unter schonenden Bedingungen gemästet und wieder abgemagert. 3. Durch Zuchtwahl konnte in drei Generationen ein Rattenstamm isoliert werden, der schneller verfettet und langsamer abmagert als der ursprüngliche Stamm. In zwei folgenden Generationen wurde diese Eigentümlichkeit durch Zuchtwahl weiter verstärkt. 4. Die Hypophyse zeigt bei Tieren des Ausgangsstammes Ruhebilder während der Verfettung und Anregungsbilder während der Abmagerung. Bei Tieren des ausgewählten Stammes zeigen sich infolge langfristiger Mast Degenereszenzbilder in einzelnen Zellen der Hypophyse. 5. Die Schilddrüse und die Nebenniere scheinen bei Tieren des ausgewählten Stammes weniger aktiv als bei Tieren des Ausgangsstammes. Die Mast hat signifikante zusätzliche Verminderung der Aktivität der Schilddrüse und der Nebennieren zur Folge. 6. Eventuelle prädisponierende Einflüsse der endokrinen Drüsen auf die Verfettung werden erwogen.

    Notes: Zusammenfassung Mit dem Oszillographen nach Gesenius und Keller wurden bilateral Frakturen der oberen und unteren Gliedmaßen bezüglich der Durchblutung ober- und unterhalb der Bruchstelle untersucht. Bei frischen Frakturen zeigte sich oberhalb der Bruchstelle eine etwas bessere und unterhalb eine etwas schlechtere Durchblutung der Gliedmaße. Dieser Befund ist nach der völligen Bruchheilung nicht mehr eindeutig nachweisbar. Bei Anwendung der Kirschnerdrahtextension war auffallend, daß bei Oberschenkelbrüchen, besonders bei starkem Zug und älteren Leuten, die Durchblutung unterhalb der Bruchstelle abnimmt, so daß in diesen Fällen zur Vorsicht geraten werden muß. Brüche mit verzögerter Callusbildung und Pseudoarthrosen zeigten keinerlei Minderdurchblutung, ebenso nicht das Sudecksche Syndrom.

    Notes: Summary A method to filter whole blood through 5-µm filters at a pressure of 20 cm column of water and the use of aging red cells as a reproducible procedure to generate rigidified cells is described. The system is sensitive enough to evaluate in pharmacologic screening possible influences of chemical agents on red cell deformability. Investigations with the methylxanthine derivates pentoxifylline and theophylline, the beta-blocker penbutolol and prednisolone, revealed that only the first compound has a dose-dependent effect on red cell deformability.

    Notes: Summary I. The bibliography about corticosteroid influence on immune response is briefly reviewed. Generally, it is admitted that corticosteroids are immunosuppressive when administered in large amounts. Divergent opinions are recalled. II. The aims of the experiments are summarized: A) Investigate on the influence of corticoids (given in just substitutive amounts) on skin allograft rejection. B) Investigate on possible interactions between adrenal cortex and thymus with this test. III. It was observed, that A) Adrenalectomy resulted in a significant delay of allograft rejection. B) This could be suppressed partially by administering aldosterone, corticosterone, and desoxycorticosterone and completely by administering at least two of these hormones. C) Cortisol had shown a minor inhibitory influence. D) There was no obvious difference between adrenalectomized and thymectomized rats and those adrenalectomized only. IV. These observations are replaced in the context of bibliography and their significance is discussed.
