    Notes: Summary Six out of ten lactic acid bacteria strains tested displayed linamarase activity.Lactobacillus plantarum strain A6 displayed the greatest activity affecting 36U/g cells on MRS cellobiose. The strain also broke down in less than 2 hours the linamarin extracted from cassava juice. HPLC analysis of the products of the reaction showed that the bacteria converted the linamarin into lactic acid and acetone cyanohydrin.

    Notes: Summary The suitability of disc fermenter for efficient production of conidiospores by Trichoderma harzianum is limited to a working capacity of 0.61 agar medium due to large decrease in spore production per cm2 of the culture surface area with further increase in the capacity. In contrast, Zymotis, a large scale solid state fermenter designed at ORSTOM, France and use of inert solid support for absorbing nutrients offer many advantages for production of conidiospores in quantity sufficient enough to inoculate pilot and larger fermenters. Five times higher production of conidiospores in Zymotis, as compared to the agar medium in flask, constitutes a success in the development of large scale inoculum.

    Notes: Summary Recovery of cellulases from solid state cultures ofTrichoderma harzianum was efficiently achieved by hydraulic pressing. Pressing of fermented solids yielded carboxymethyl-cellulase (CMCase) extraction efficiency of 71% and a ratio of leachate to fermented solids of 0.58 (v/w). Addition of water to pressed solids and second pressing improved the efficiency (95%) with simultaneous increase in the ratio to 1.16 (v/w). The overall extraction of filter paper activity was lower (85%) than that of CMCase. This technique is simple and its extraction efficiency is similar to that obtained in multiple-contact counter-current systems. The hydraulic press in its individuality was not used earlier to leach the product from fermented solids.

    Notes: Summary Germination of condiospores fromA.niger in very concentrated suspension was required to inoculate solid fermentation media, but a germination self-inhibitor was detected in spores. It was found that the inhibitory effect depended on the composition of the medium and was reduced when glucose was used as a carbon source. The effect of the self-inhibitor was eliminated by washing the spores and using glucose and a protein nitrogen source in the germination medium. By this method it was possible to increase about 100 times (106 to 108) spore concentration, keeping more than 90% germination.

    Notes: Abstract A new technique for separation and determination of ATP, ADP, AMP, NAD+, NADP+, NADH and NADPH in Lacwbacillus plantarum has been developped. It involves acid and basic extraction of nucleotide pool, analysis by reverse-phase high performance liquid chromatography on a 5 μm Spherisorb ODS-1 column and UV detection. The method offers advantages in accurate estimations of adenylic and nicotinamide nucleotides concentrations in Lactobacillus plantarum during the growth phase. Such results indicate the potential of this technique as an important research tool.

    Notes: Abstract  Experiments were carried out to study the production of aroma compounds from coffee husk by a fungal culture of C. fimbriata in solid state fermentation. Hot water treated coffee husk seemed to be useful substrate for aroma production in comparison to whole coffee husk or its water extract. Raw data were integrated in order to calculate the total volatiles (TV) accumulated during the fermentation using the Gompertz model. Glucose addition between 20–35% increased significantly the yields of aroma compounds. Supplementation of leucine further improved the TV production significantly (about 58%), specially the esters. When soybean oil was added, TV production was similar to the control, showing that the fungus was not able to use soybean oil for its primary metabolism, nor it acted as precursor for the synthesis of methyl ketones as reported for other fungi. Addition of saline solution drastically decreased the volatile production. Under optimized conditions, a total of 13 compounds were produced which included alcohols (2), aldehyde (1), ketones (2) and esters (8). Ethyl acetate was the prominent compound, followed by ethanol.

    Notes: A mathematical model was developed and tested to simulate the generation and transfer of heat in solid substrate fermentation (SSF). The experimental studies were realized in a 1-L static bioreactor packed with cassava wet meal and inoculated with Aspergillus niger. A simplified pseudohomogeneous monodimensional dynamic model was used for the energy balance. Kinetic equations taking into account biomass formation (logistic), sugar consumption (with maintenance), and carbon dioxide formation were used. Model verification was achieved by comparison of calculated and experimental temperatures. Heat transfer was evaluated by the estimation of Biot and Peclet heat dimensionless numbers 5-10 and 2550-2750, respectively. It was shown that conduction through the fermentation fixed bed was the main heat transfer resistance. This model intends to reach a better understanding of transport phenomena in SSF, a fact which could be used to evaluate various alternatives for temperature control of SSF, i.e., changing air flow rates and increasing water content. Dimensionless numbers could be used as scale-up criteria of large fermentors, since in those ratios are described the operating conditions, geometry, and size of the bioreactor. It could lead to improved solid reactor systems. The model can be used as a basis for automatic control of SSF for the production of valuable metabolites in static fermentors.
