    Description / Table of Contents: Summary 1. Pseudoisocyanin interacts with densly positioned electronegative groups of mucopolysaccharides in tissues and in solutions in the same way as it interacts with linear positioned electronegative groups of synthetic products in solution (for instance polyaethylensulfoacids). The metachromasia, which is due to this reaction of pseudoisocyanin with mucopolysaccharides shows a characteristic wave-band 5727 Å (Scheibe undSchauer 1958). The dye is bound electrostatically by the electronegative groups in form of a “reversible polymerisate”. 2. The metachromatic reaction with the “reversible polymerisate” has been demonstrated in tissue-sections. The polymerisate with the dyestuff is shown to adsorb light at the same wavelength in tissues as in solutions. This finding confirms the identity of the reaction in tissues and in solutions. 3. Pseudoisocyanin seems to be especially suited for the detection of mucopolysaccharides, for even a monomolecular layer of dyestuff allows the observation of the “reversible polymeric band” and therefore shows metachromasia. Further, after staining with pseudoisocyanin even small trans of mucopolysac charides which are not visible in the white light can be demonstrated by means of monochromatic light at the wave-length of the “polymer absorption”. 4. As shown by staining mastcells, cartilage-tissue, hyaliniced connectivetissue, pseudoisocyanin seems to be of use for appliance in histochemistry.
    Notes: Zusammenfassung 1. Pseudoisocyanin gibt mit den dicht gelagerten elektronegativen Gruppen von Mukopolysacchariden in Geweben und Lösungen, wie auch mit synthetischen Produkten mit linear angeordneten elektronegativen Gruppen in Lösung wie z. B. Polyäthylensulfosäuren eine metachromatische Reaktion mit der charakteristischen langwelligen Bande (vgl.Scheibe u.Schauer 1958). Die elektronegativen Gruppen binden die Farbstoffmoleküle elektrostatisch und bilden die Gruppierung des „reversiblen Polymerisates“. 2. Die metachromatische Reaktion mit der reversibel „polymeren“ Bande läßt sich in Gewebsschnitten deutlich demonstrieren. Das „Farbstoffpolymerisat“ absorbiert in Lösung bei der gleichen Wellenlänge wie im Gewebe, wodurch die Gleichheit der Vorgänge im Gewebe und in Lösung bewiesen ist. 3. Das Pseudoisocyanin erscheint für die Darstellung von Mukopolysacchariden besonders geeignet, da nach früheren Arbeiten (Scheibe 1938,Zimmermann u.Scheibe 1956) schon eine monomolekulare Schicht die reversibel polymere Bande und damit die Metachromasie beobachtbar macht. Ferner sind bei Betrachtung der mit Pseudoisocyanin gefärbten Schnitte im monochromatischen Licht bei der Wellenlänge der „polymeren Absorption“ Spuren von Mukopolysacchariden noch deutlich zu erkennen, die bei Betrachtung im weißen Licht unauffällig bleiben. 4. An Hand einiger Beispiele (Mastzellen, Knorpelgewebe, hyalinisiertes Bindegewebe) wird die Verwendungsmöglichkeit in der Histochemie gezeigt.

    Description / Table of Contents: Zusammenfassung Die von Chang entwickelte Methode, artefizielle Zellen — in Kollodium und Albumin verkapselte Aktivkohle — zur Hämoperfusion einzusetzen, wurde in vitro studiert. Geprüft wurde die Kapazität zur Adsorption von schlecht wie gut dialysablen Substanzen (Barbiturate, Diazepam, Parathion) und einem endogen toxischen Metaboliten (p-Hydroxyphenylessigsäure). — Mit der Adsorptions-Einheit lassen sich in 90 min 80–90% der Ausgangskonzentrationen von Barbiturat, Diazepam und p-Hydroxyphenylacetat eliminieren, von Parathion etwa 50%, Bromid und Ammoniak werden nicht adsorbiert. Versuche, durch den Einbau von Heparin in die Hülle der Kohlegranula die regionale Heparinisierung unnötig zu machen, waren erfolglos. Bei sachgemäßer Herstellung des Shunt-Systems, Einhaltung steriler Kautelen und regionaler Heparinisierung ist der Einsatz des „Entgiftungs-Shunts“ eine vielversprechende Möglichkeit zur Therapie schwerer exogener und endogener Intoxikationen, die einer konventionellen Therapie nicht zugänglich sind.
    Notes: Summary The method developed by Chang for using in collodion and albumin encapsuled charcoal as artificial cells for hemoperfusion has been studied in vitro. The capacity for the absorption of more or less dialysable exogenous substances (barbiturate, Diazepam, Parathion) and an endogenous toxic metabolite (p-hydroxyphenyl acetic acid) has been examined. With the absorption unit one can eliminate 80 to 90 per cent of the initial concentration of barbiturate, Diazepam and p-hydroxyphenyl acetic acid in about 90 min, of Parathion about 50 per cent in the same time, whereas bromide and ammonia are not absorbed. Attempts to avoid regional heparinisation by the incorporation of heparin into the capsule of charcoal granules were not successful. Assuming correct preparation of the hemoperfusion system, observance of steril precoutions and regional heparinisation, the use of the “detoxication shunt” is a promising means in the treatment of severe exogenous and endogenous intoxications which are not accessible by conventional therapeutic means.

    Notes: Summary A group of 27 patients with panic disorder with or without agoraphobia were compared with 10 control subjects before stress exposure. No statistically significant differences between patients and controls were found for the cardiovascular parameters. Skin conductance level and skin conductance reaction were significantly higher in the patient group. They also showed higher self-ratings in behavioural symptoms associated with anxiety. There were statistically significant higher venous plasma levels of norepinephrine in patients than in controls, although the epinephrine levels were similar. The number of binding sites of α2 and the affinity of3H-yohimbine to the α2-receptors on intact thrombocytes was statistically significantly lower in patients compared to controls. Significant differences between the gender groups of patients and controls were found for electrodermal activity and epinephrine levels. These data add further evidence to an overshooting activation of the noradrenergic pathway in patients with panic disorder, possibly based on a dysregulation of α2-receptor
