Notes: A case of recombination between the putative class I ELA antigen series and the strudure(s) governing mixed lymphocyte reactivity in an informative horse family is described. The results of serological typing, ‘lysostripping’ and mixed lymphocyte culture tests strongly suggest that the recombination took place between two loci and is not intragenic. An alloantigenic membrane structure, provisionally called B1, which does not belong to the known ELA series, was also involved in the cross-over. The B1 antigen resembles the class II gene products of other species in two respects: it is not present on platelets, and doantiserum with specificity for B1 inhibits the stimulatory effect of B1-carrying cells in mixed lymphocyte cultures. The B1 antigen does not follow the classical distribution, however, being expressed on both B and T lymphocytes. The finding of separate loci for the first series of ELA antigens and the MLR governing structure(s) demonstrates the similarity of the genetic organization of the horse MHC to that in other species.

Notes: A randomised, double-blind investigation was undertaken into the value of metoclopramide and domperidone as anti-emetics in orthopaedic patients who received spinal anaesthesia. Seventy-seven patients were divided into three statistically comparable groups: Group I received 10 mg metoclopramide, Group II 10 mg domperidone, and Group III NaCl as placebo. These substances were given intravenously before the spinal puncture. There was a comparable decrease in mean arterial blood pressure (MAP), level of sensory block and dose of tetracaine in all three groups. The patients who were nauseated and/or vomited showed a greater decrease in MA P. The incidence of nausea and/or vomiting in the metoclopramide and the placebo group were similar, 20% and 15% respectively, whereas this complication was apparent in 35% of patients who received domperidone. We conclude that metoclopramide and domperidone in the dosages used are ineffective in alleviating nausea and vomiting after spinal anaesthesia.

Notes: Abstract By means of microsurgical lymph cannulation human skin lymph derived from the late phase of an elicitation reaction to diphenylcyclopropenone was sampled. Cells were isolated by centrifugation and then treated with mouse anti-CDl a mnonoclonal antibodies and sheep antimouse antibody-coated Dynabeads. Ultrastructural and immunocytochemical analyses revlaled anti-CDl a/Dynabead-rosetted CDl a- and protein S-100-positive cells which did not express monocyte surface markers, but surface antigens such as HLA-DR, ICAM-I and, in part, LFA-3. In comparison to freshly prepared human epidermal Langerhns cells (LC), a large fraction of these cells contained no or markedly fewer Birbeck granules and exhibited extensive ruffling of the surface. These data suggest that the phenotype of LC in skin lymph derived from the elicitation phase of allergic contact dermatitis is similar to LC cultured in vitro. In the functional concept or LC or our time, these cells correspond to the dendritic cells designated as “veiled”.

Notes: In a collaborative study by 12 laboratories the reproducibility of bioassay techniques was analysed by comparison and statistical treatment of the ED50-values (μg herbicide/g soil). Commonlyused bioassay techniques were investigated; two direct seeding methods, a transplanting method and a shoot extension method. The herbicides studied were two photosynthesis inhibitors, atrazine and metribuzin using Lepidium sativum and Brassica rapa as test plants; two germination inhibitors, tri-allate and trifluralin using Avena sativa as test plant. The mean ED50-value of atrazine in the direct seeding method was 0.18 with a variation from 0.12 to 0.29 and 0.12 (0.07–0.68) in the transplanting method. The values of metribuzin were 0.05 (0.02–0.11) and 0.03 (0.01–0.15) respectively. The values for triallate were 1.15 (0.50–2.60) in the direct seeding method and 1.10 (0.54–2.53) in the shoot extension method and for trifluralin 3.11 (0.72–18.31) and 1.33 (0.48–2.94) respectively. The reproducibility was best in the shoot extension method. No outliers (confidence intervals lying entirely outside the confidence interval of the overall mean) in the ED50-values were observed with atrazine and metribuzin when using the direct seeding method, whereas with the germination inhibitors a high number of results (four with tri-allate, six with trifluralin) could be considered as outliers. A comparison of the use of fresh weight and dry matter showed good agreement between the ED50-values they gave, with similar reproducibility.

Notes: A new, non-MHC linked alloantigenic membrane antigen on the equine lymphocytes is described. This antigen was characterized with alloantisera in the two-stage microcytotoxicity test and designated as ELy-1 antigen. The frequency of ELy-1 antigen positive animals in various populations is close to 50%. ELy-1 shows an autosomal, dominant inheritance. Since an allelic antigen (s) could not be demonstrated in family studies, it is assumed that only two alleles ELy-I+ and ELy-I- exist. The ELy-1 antigen in positive animals is expressed on both T and B lymphocytes but it is not present on erythrocytes and thrombocytes. The incidence of ELy-1 antigen positive animals is significantly higher in horse groups suffering from chronic bronchitis or from laminitis, compared to clinically healthy controls.

Notes: We report a 6-year-old boy suffering from acquired epidermolysis bullosa, who presented with extensive lesions of the mucous membranes and disseminated, herpetiform and‘cluster of jewels' like vesicles and bullae arising on erythematous plaques. Direct immunofluorescence showed linear deposits of IgG and C3 at the epidermal basement membrane zone. Indirect immunofluorescence demonstrated circulating autoantibodies(titre 1:128–1:256) directed at the blister floor of human NaCl-split skin, and reacting specifically with collagen VII of the anchoring fibrils, as demonstrated by immunoblotting. The disease was controlled with a combination of dapsone and prednisone. This case demonstrates the importance of modern immunological techniques in classifying childhood autoimmune bullous diseases, as precise diagnosis is important in determining an appropriate therapeutic regimen.
