    Notizen: Abstract  Rice sheath blight, caused by Rhizoctonia solani Kühn, is one of the three major diseases of rice. The present study was conducted with an F2 clonal population of Jasmine 85/Lemont. The F2 population, including 128 clonal families, was inoculated by short toothpicks incubated with a strain, RH-9 of the fungus. Based on field disease evaluations in 2 years and a genetic map with 118 evenly distributed molecular markers, we identified six quantitative trait loci (QTLs) contributing to sheath blight resistance. These QTLs, qSB-2, qSB-3, qSB-7, qSB-9-1, qSB-9-2 and qSB-11, were located on chromosomes 2, 3, 7, 9 and 11, respectively. The respective alleles of qSB-2, qSB-3, qSB-7, and qSB-9-2 from Jasmine 85 could explain 21.2%, 26.5%, 22.2% and 10.1% of the total phenotypic variation, respectively; while the alleles of qSB-9-1 and qSB-11 from Lemont could explain 9.8% and 31.2% of the total phenotypic variation. Of these qSB-2 and qSB-11 could be detected in both years, while remaining loci were detected only in a single year. Furthermore, four QTLs (qHD-2, qHD-3, qHD-5 and qHD-7) controlling heading date and three QTLs (qPH-3, qPH-4 and qPH-11) controlling plant height were also identified. Though rice sheath blight resistance may be influenced by morphological traits, such as heading date and plant height, in the present study most detected resistance loci were not linked to the loci for heading date or plant height.

    Notizen: Abstract  In order to understand the genetic basis of the paste viscosity characteristics (RVA profile, which is tested on the Rapid Visco Analyser) of the rice grain, we mapped QTLs for RVA profile parameters using a DH population derived from a cross between an indica variety, Zai-Ye-Qing 8 (ZYQ8), and a japonica variety, Jing-Xi 17 (JX17). Evidence of genotype-by-environment interaction was found by comparing the mapped QTLs between two locations, Hainan (HN) and Hangzhou (HZ). A total of 20 QTLs for six parameters of the RVA profiles were identified at least one location. Only the waxy locus (wx) located on chromosome 6 was detected significantly at both environments for five traits, i.e. hot paste viscosity (HPV), cool paste viscosity (CPV), breakdown viscosity (BDV), consistency viscosity (CSV) and setback viscosity (SBV). This locus explained 19.5%–63.7% of the total variations at both environments, suggesting that the RVA profiles were mainly controlled by the wx gene. HPV, CPV, BDV, CSV and SBV were also controlled by other QTLs whose effects on the respective parameter were detected only in one environment, while for the peak viscosity (PKV), only 2 QTLs, 1 at HN,the other at HZ, were identified. These results indicate that RVA profiles are obviously affected by environment.

    Notizen: Abstract  An awned rice plant was discovered among the progeny of the triploid Zhongxian 3037. Cytological investigation showed that this awned plant was a variation with extra chromosomes. Based on the properties of the extra chromosomes during both meiosis and mitosis – e.g., short stature, darkly staining, instability in chromosome number, and lack of synapsis with A chromosomes – they could be considered to be B chromosomes in rice. The B chromosome(s) in both asexual and sexual progenies exhibited a unique segregation that is different from that of telotrisomics and other aneuploids. Moreover molecular marker analysis detected no dosage effects between the B-chromosome plant and the normal diploid of Zhongxian 3037, indicating that the B chromosomes might not be directly derived from any A-chromosome fragments in rice.

    Notizen: Abstract  The apple rootstock M26 (Malus domestica) was infected with a binary vector system of Agrobacterium tumefaciens carrying the neomycin phosphotransferase II and Arabidopsis phyB genes. Thirteen transformed clones were obtained from 329 infected leaves. Five of the clones had a single copy integration, six clones had two copies, one clone had five copies and one of the clones had eight copies of the phyB gene integrated. No differences in rooting were found between transformed and untransformed plants. The stem length was reduced in nine of the 13 transgenic clones, and shoot, root and plant dry weights were reduced in all transformed clones compared with untransformed control plants. Northern analysis showed that the Arabidopsis phyB gene was expressed in the transformed clones.

    Notizen: Abstract A new rice semidwarfing gene which is not allelic tosd1, temporarily designated assdg, might be of use as a new source of semidwarfism in rice breeding programs. We report here the identification of a DNA marker closely linked to this gene. The DNA marker was identified by testing 120 mapped rice RFLP makers as hybridization probes for Southern analysis of a pair of nearly isogenic lines with or withoutsdg. Linkage association of the marker with the gene was verified using a F2 population segregating for semidwarfism. RFLP analysis showed thatsdg is closely linked to a single-copy DNA clone RZ182 on chromosome 5, with a distance of 4.3 centiMorgans between them. This marker may facilitate early selection for the semidwarfing gene in rice breeding programs

    Notizen: Abstract  The inheritance of grain quality is more complicated than that of other agronomic traits in cereals due to epistasis, maternal and cytoplasmic effects, and the triploid nature of endosperm. In the present study, an established rice DH population derived from anther culture of an indica/japonica hybrid was used for genetic analysis of rice grain quality. A total of five parameters, amylose content (AC), alkali-spreading score (ASS), gel consistency (GC), percentage of grain with a white core (PGWC) and the square of the white core (SWC), were estimated for the DH lines and the parent varieties. For each parent, the value of each parameter was relatively stable in three locations, Beijing, Hangzhou and Chengdu, while the differences between the parents were significant for all five parameters. AC showed a bimodal distribution, and the distribution of ASS was skewed toward the value of JX17, while the other three parameters displayed continuous distributions among the DH lines with partially transgressive segregations. For AC, a minor and a major gene were found on chromosomes 5 and 6 respectively. The major gene, which should be an allele of wx, explained 91.9% of the total variation. For GC, two QTLs were identified on chromosomes 2 and 7 respectively. For ASS, a minor and a major gene were both located on chromosome 6. The major gene should be the same locus as the alkali degeneration gene (alk). Genetic linkage between alk and wx was found in QTL mapping. For PGWC, two QTLs were located on chromosomes 8 and 12. Only a minor QTL was found for SWC on chromosome 3. The results and the molecular markers presented here may be useful in rice breeding for grain quality improvement.

DNA sequence analysis of the 1844-bp insert of RRS7 revealed that it contained six tandemly repeated units, of which five were 155 bp in length and one was 194 bp in length and contained an imperfect internal 39-bp duplication. Southern blot analysis showed that the boundary sequence contained in RRS7 is a single-copy sequence. A 155-bp consensus sequence derived from the six monomeric repeats contained no internal repeat and showed no significant homology to other currently known sequences. The results of Southern blot and sequence analysis revealed that there are at least two subfamilies present in the RRS7 family; these are represented by the DraI site and the MspI site, respectively. Restriction digestion with two pairs of isoschizomers MboI/Sau3A and MspI/HpaII demonstrated that most of the C residues in the GATC sites and the internal C in the CCGG sites of the RRS7 family in O. Alta were methylated. The usefulness of the RRS7 family in determining the evolutionary relationship of the genome DD and other Oryza genomes is discussed.

    Notizen: Abstract The level scheme of 143Gd has been extended to E = 8.7MeV (I = 53/2+) in an experiment with the GASP γ-ray spectrometer and the 100Mo(48Ti,5n) fusion reaction. Most of the level scheme shows single particle character. A strongly populated cascade consisting of ten stretched E2 transitions and another structure with weak M1 and strong E2 shows an indication of a collective character.
