    Notes: Summary In this present study. 260 histologically conlirmed melanocytic skin tumours (188 benign nacvi and 72 malignant melanomas; from 1989 to 1990) were investigated with regard to valid surface microscopical criteria of malignancy. The tumours were analysed using a system which assessed eight components. Most melanomas were characterized by the following pattern: asymmetrical pigment distribution, more than three colours, black pigment, peripheral stripes, and asymmetrical depigmentation. The results were evaluated statistically by contingency tables and logistic regression procedures. On the basis of the classification derived, the sensitivity and specificity were determined for lesions from 1991. and were 0·9 and 0·85, respectively. Many melanocytic naevi were not identified by the above criteria, or were found only occasionally. Pigment network was often absent in naevi and melanomas, and was not decisive for the diagnosis. The present investigation demonstrates that in vivo diagnosis of pigmented melanocytic lesions can be improved by surface microscopy.

    Notes: Background/Aims: Different cytokine genotypes have been described in periodontal disease. The aim of the present study was to investigate the genetic association of two previously described interleukin-10 (IL-10) polymorphisms in patients with aggressive (AP) and chronic periodontitis (CP) and to investigate possible associations with clinical manifestations.Methods: Based on clinical parameters and radiographs, 23 patients with CP and 18 patients with AP were included in the study. Additionally, 21 age-matched healthy subjects served as a control group. Genomic DNA was isolated from whole blood samples and the IL-10 promoter sequences from positions − 597 to − 824 were amplified by polymerase chain reaction (PCR). Polymorphisms were detected by restriction-enzyme cleavage. The A and C alleles at the − 597 position were associated with the T and C alleles at the − 824 position, respectively. Fisher's exact test was used for the statistical analysis.Results: No significant differences were observed in the allele frequencies between controls and AP patients (p = 0.70) or CP patients (p = 0.43), although the previously reported association between allele A at position − 597 and allele T at position − 824 was observed in our population.Conclusion: We conclude that the investigated polymorphisms are not associated with periodontal disease.

    Notes: Background, aim: The aim of the present study was to measure interleukin-1β concentrations and neutrophil elastase activity in gingival crevicular fluid (GCF) during experimental gingivitis in humans.Material and Methods: 12 healthy young men participated. After prophylaxis, they performed optimal hygiene to reach plaque and gingivitis indices of or approaching zero. All oral hygiene measures were then ceased for a period of 18 days. The Quigley-Hein plaque index (PLI) and Saxer & Mühlemann papillary bleeding index (PBI) were assessed. GCF samples were taken from the mesiobuccal site of two contralateral teeth in the upper jaw by means of periopapers at baseline and on days 3, 7, 14 and 18. After measuring the gingival crevicular fluid volume (GCFV) with the Periotron 8000, the samples were analyzed in our laboratory for the detection of IL-1β concentration by ELISA.Results: PLI and PBI showed a reduction prior to baseline reaching almost zero, both increasing from day 0 to day 18 (PLI=from 0.1 to 2.9, PBI=from 0 to 2.0). IL-1β concentration increased from 229.25 ng/ml (day 0) to 526.13 ng/ml (day 18). Clinical data and IL-1β concentrations were correlated with elastase activity (EA). No significant correlation could be demonstrated between the clinical parameters assessed and IL-1β or EA (Spearman rank correlation coefficient). A correlation between GCFV and PBI from day 0 to day 18 could be demonstrated.Conclusion: Overall, both IL-1β and EA showed an increase from baseline throughout the whole study.

    Notes: Summary Guinea pigs supplied with 5 mg/100 g vitamin in the food seem to be at the marginal substitution for a survival and show no ability to adapt the ascorbic acid metabolism to this low support. Guinea pigs longlastingly (6–8 weeks) supplied with 20 mg/100g or 680 mg/100g vitamin C in the food show the typical symptoms of an evolving adaptation by an overshooting in the course of the ascorbic acid levels in several organs after a switch to an extremely different vitamin C supply.

    Notes: Summary “Haemostasis time” (HT), the occlusion time of a Butterfly 25 short cannula inserted into the cubital vein, is a bleeding time modification comparable to the skin bleeding times according to Duke and Ivy/Mielke. It also measures platelet function and is not influenced more than the latter tests by clotting factors [11]. In HT, subendothelium is replaced by a standard artificial surface. The technique was investigated in patients with haemophilia A and B, von Willebrand's disease (vWD), and defects of factors VII and XI. HT was prolonged in 9/16 patients with haemophilia A/B, but did not correlate with the factor VIII:C/IX:C values. However, it reflected the different bleeding tendencies in those patients as represented by early or late onset of bleeding symptoms and occurrence of spontaneous haemarthroses. Of the vWD patients, not classified by multimeric analysis, 15/31 had prolonged Simplate II bleeding times, 14/31 prolonged HTs. Only 20/31 patients had corresponding normal or prolonged bleeding times with both techniques. HT correlated significantly with the Duke bleeding time (p=0.011), ristocetin cofactor activity (p=0.003) and von Willebrand factor antigen (p=0.022), while no correlations were found between these parameters and the Simplate II method. Statistical evaluation shows, that in vWD, HT can replace the less precise Duke bleeding time but not the non-related Ivy/Simplate techniques.

    Notes: Summary In this comparative study of the antiproliferative effect of heparin (average mol. weight 16 kD) a different heparin sensitivity between cultivated smooth muscle cells derived from the media of the human aorta and the vena cava is observed. Heparin induces a partial proliferation inhibition of 54% in the aortic- and 17% in the vena cava-derived smooth muscle cells on day seven. The increase in cell volume suggests a selection of larger volume subpopulation of cells or an influence on the G2-phase of the cell cycle. Heparin and heparin-like compounds may develop into future therapeutic drugs for the prevention of increased smooth muscsle cell proliferation after vessel wall injury.

    Description / Table of Contents: Summary The specific amounts of the microsomal cytochromes P-450 (P-454) and b5 and the amounts of microsomal protein in liver and renal cortex of guinea pigs depend on the extent of the vitamin C supply to which the animals are adapted (at least 6 weeks). In the case of low supply — with 5 mg vitamin C in 100 g food — which still permits to survive, the amounts of the cytochromes are decreased in both organs (an accumulative supply — every 3.5 days via stomach tube — causes no evident decreases). The reduced amount of cytochrome P-450 gives rise to a corresponding elongation of the sleeping time after injection of evipane; this agrees with other reports upon restriction of metabolic activities by marginal supply. But the amount of the cytochrome is induced as well in these animals by several injections of evipane, to the same level as in animals supplied with more vitamin C. Our early report on a considerable decrease of the specific amount of cytochrome P-450 in the liver by omission of vitamin C for 14 days proves correct only in this organ and when the guinea pigs were previously abundantly supplied with vitamin C. There is no corresponding decrease after an adaptation to medium and lesser supplies of vitamin C. The decrease of the cytochrome after abundant supply seems to be due to a diminished stimulation of its synthesis in connection with modifications in the hormonal control of the metabolism and, thus, is only indirectly connected with the lack of vitamin C. The effects of omission of vitamin C supply probably depend generally on the previous adaptation, at least in the liver. The considerable decrease of the cytochrome P-450 and the simultaneous reduction of the mitochondrial compartment raise the question of if a maximum supply with vitamin C is favorable resp. the physiological optimum.
    Notes: Zusammenfassung Bei Meerschweinchen hängen die spezifischen Gehalte der mikrosomalen Cytochrome P-450 (P-454) und b5 und der Gesamtgehalt des mikrosomalen Proteins in Leber und Nierenrinde davon ab, an welches Ausmaß der Vitamin-C-Versorgung der Organismus adaptiert ist (mindestens 6 Wochen). Bei sehr niedriger Versorgung — mit 5 mg Vitamin C in 100 g Futter —, die gerade noch ein Überleben gestattet, sind die Gehalte der Cytochrome in beiden Organen erniedrigt (bei akkumulativer Zufuhr — alle 3 1/2 Tage via Schlundsonde — sind sie nicht deutlich vermindert). Der verringerte Gehalt des Cytochrom P-450 führt bei Injektion von Evipan zu einer entsprechend verlängerten Schlafzeit; dies steht mit anderen, bereits beschriebenen Einschränkungen von Stoffwechselaktivitäten bei marginaler Versorgung mit Vitamin C in Einklang. Durch mehrfache Injektionen von Evipan wird der Gehalt des Cytochroms aber auch bei diesen Tieren induziert, auf dasselbe Niveau wie bei besser mit Vitamin C versorgten Tieren. Unser alter Befund, daß bei 14tägiger Vitamin-C-freier Fütterung der spezifische Gehalt des Cytochrom P-450 in der Leber erheblich abnimmt, trifft nur für dieses Organ und für den Fall zu, daß die Meerschweinchen vorher reichlich mit Vitamin C versorgt waren. Nach Adaptation an mittlere und geringe Zufuhr von Vitamin C erfolgt kein entsprechender Abfall. Die Abnahme des Cytochroms nach reichlicher Versorgung beruht anscheinend auf einer verminderten Stimulierung seiner Synthese im Rahmen von Veränderungen der hormonellen Steuerung des Stoffwechsels, so daß nur ein indirekter Zusammenhang mit dem Mangel an Vitamin C vorliegt. Vermutlich sind die Folgen einer Vitamin-C-freien Fütterung, zumindest in der Leber, generell von der vorangegangenen Adaptation abhängig. Der starke Abfall des Cytochrom P-450 und die bereits beschriebene gleichzeitige Verringerung des mitochondrialen Kompartiments werfen die Frage auf, ob eine maximale Versorgung mit Vitamin C vorteilhaft bzw. physiologisch optimal ist.
