Summary 1. By incubations of rat epididymal fat pads, the glucose uptake, the 14CO2 production of added 14C-1-glucose, the incorporation of 14C-1-glucose into lipids and glycogen, and the incorporation of 14C-1-leucine into fatfree tissue residue (protein) was determined under the addition of 6-aminonicotinamide in the presence and absence of insulin. 2. Addition of 6-aminonicotinamide decreases the insulin stimulated glucose uptake (halfmax. cone. 0,3 10−5 M). The effect of 6-aminonicotinamide is seen about 2 hours after addition of the substance. Concomitant addition of NAD or nicotinamide prevents the inhibitory effect of 6-aminonicotinamide (halfmax. conc. 0.6 resp. 1.1 10−5 M). 6-ANAD (6-aminonicotinamide adenine dinucleotide) also inhibits insulin stimulated glucose uptake (halfmax. conc. 10−5). From these effects it is concluded that 6-ANAD is the really effectory substance of 6-aminonicotinamide. 3. To the same extent as the glucose uptake into adipose tissue, the production of 14CO2 from 14C-1-glucose, the incorporation of 14C-1-glucose into lipids, and the incorporation of 14C-1-leucine into proteins are inhibited. 4. This means that several insulin sensitive reactions are inhibited by the addition of 6-aminonicotinamide. The most probable mechanism of its action seems therefore insulin being excluded from its site of action. 5. Independently thereof 6-aminonicotinamide increases the incorporation of 14C-1-glucose into glycogen of the adipose tissue. This effect could be explained by the earlier described inhibitory effect of 6-ANAD on the phosphorylase-b-kinase (in muscle).

Zusammenfassung 1. Am epididymalen Fettgewebe der Ratte wird die Glucoseaufnahme während der Inkubation, die 14CO2-Bildung aus 14C-1-Glucose, die Inkorporation von 14C-1-Glucose in Lipide und Glycogen, sowie die Inkorporation von 14C-1-Leucin in den fettfreien Gewebsrückstand (Protein) unter Zugabe von 6-Aminonicotinamid mit und ohne Insulin-Anwesenheit untersucht. 2. 6-Aminonicotinamid-Zusatz setzt die Glucoseaufnahme herab (halbmax. Hemmkonz. 0,3 10−5 M), soweit sie durch Insulin stimulierbar ist. Die Auswirkung des 6-Aminonicotinamids tritt erst etwa 2 Std nach Zugabe auf. Gleichzeitiger Zusatz von NAD oder Nicotinamid hebt die Hemmwirkung des 6-Aminonicotinamids auf (halbmax. Effekte mit 0,6 resp. 1,1 10−5 M). 6-ANAD (6-Aminonicotinamid-Adenindinucleotid) hemmt die insulinstimulierbare Glucoseaufnahme ebenfalls (halbmax. Hemmkonz. 10−5 M). Aus diesen Befunden wird gefolgert, daß 6-ANAD die eigentliche Wirkform des 6-Aminonicotinamids darstellt. 3. Im gleichen Ausmaß wie die Glucoseaufnahme ins Fettgewebe wird die 14CO2-Bildung aus 14C-1-Glucose, sowie die Inkorporation von 14C-1-Glucose in Lipide und von 14C-1-Leucin in Proteine gehemmt. 4. Damit werden mehrere insulinempfindliche Reaktionen durch 6-Aminonicotinamidzusatz gehemmt. Die Ausschaltung des Insulins vom Wirkort wird als wahrscheinlicher Mechanismus diskutiert. 5. Unabhängig davon erhöht 6-Aminonicotinamid im Fettgewebe den Einbau von 14C-1-Glucose in Glycogen. Dafür könnte die schon früher (für den Muskel) beschriebene Hemmwirkung von 6-ANAD auf die Phosphorylase-b-Kinase verantwortlich sein.
