    Notes: Zusammenfassung Um das quantitative Verhalten von Kern- und Kernkörperchen bei verschiedener Wachstumsintensität von Gewebekulturen in vitro zu studieren, sind in 12650 Zellen die Kerne und in weiteren 1600 Zellen die Kerne und die Nucleolen planimetrisch ausgewertet und die erhaltenen Meßresultate variationsstatistisch aufgearbeitet worden. Zur experimentellen Beeinflussung der Wachstumsaktivität ist ungefähr die Hälfte der Hühnchenfibrocyten- und Osteoblasten-kulturen nach 24stündiger Inkubation im Brutschrank weitere 24 Std bei Raumtemperatur gehalten worden, während die andere Hälfte bis zur Fixierung bei Körpertemperatur weitergezüchtet wurde. Im Vergleich mit den Kontrollkulturen sind in den bei nur 20/22° C gehaltenen Kulturen die Kerne (Abb. 1-3) und insbesondere die Kernkörperchen (Abb. 3 und 4) kleiner, der nucleo-nucleoläre Quotient (Abb. 6 und 7) daher etwas größer (vgl. Tabelle 2). Sowohl die Kern-als auch die Nucleolengrößen-Frequenzkurven und die Variationskurven der nucleo-nucleolären Quotienten sind — bei logarithmischer Einteilung aller dieser Größen auf der Abszisse—Normalverteilungen, wobei die mittlere quadratische Abweichung der Variationskurven aus den langsamer wachsenden Geweben immer etwas kleiner und das Frequenzmaximum dementsprechend ein wenig höher gefunden wurde. Zwischen Kern- und Nucleolengrößen besteht eine gute lineare Korrelation (Abb. 5). Sowohl in den Fibrocyten- als auch in den Osteoblastenkulturen, die während 24 Std bei Zimmertemperatur gehalten wurden, war der Prozentsatz von Amitosen und zweikernigen Zellen höher als in den im Brutschrank gebliebenen Kontroll-kulturen (vgl. Tabelle 1).

Die Granula intramitochondrialia im distalen Abschnitt der Hauptstücke der Meerschweinchenniere sind je nach der Art der verwendeten Fixierung verschieden groß (Abb. 1–3). Die Fixierung nach Caulfield stellt sie am größten dar (Abb. 1). 2. Weist man die Natriumionen im Gewebe mit der Methode von Komnick nach, so sind die Granula größer und stark elektronendicht (Abb. 5 und 6). 3. Die Natriumverarmung von Meerschweinchen nach bilateraler Adrenalektomie führt zu einer Aufhellung der Granula. Dadurch wird in ihnen ein “Gerüst” sichtbar, das aus trilaminaren Strukturen von der Beschaffenheit der “unit membrane” besteht, welche in der Peripherie der kugeligen Granula intramitochondrialia angeordnet sind (Abb. 7).

The size of intramitochondrial granules in the distal portion of the proximal convoluted tubules in the kidney of guinea pigs depends considerably on the method of fixation (Figs. 1–3). Using Caulfield's technique, the granules are particularly large (Fig. 1). 2. The granules are larger and highly adielectronic after the presence of sodium ions has been demonstrated in the tissue according to Komnick's method (Figs. 5 and 6). 3. Following bilateral adrenalectomy in guinea pigs, the sodium loss leads to a clearing of the intramitochondrial granules. Thereby, a system of trilaminar structures of unit membrane type becomes visible (Fig. 7).

    Notes: Summary The repairin vitro of long bones after experimental injury was studied and influenced with bone extract (Ossopan). The explants were cultivated by the watch-glass method in a medium of chick-plasma and embryonic extract, varying the culture time from 1 to 12 days. The bones cultivated on the above medium with addition of bone extract, showed a greater number of cells in general and particularly in the region of the callus. Osteoblasts appeared as early as the fourth day. The early appearance and quantity of the periostal osteoid tissue was striking. By the eigth day, the repair in the treated bones became slower, while the callus formation in the uninfluenced bones progressed normally. Generally the bone-continuity was more or less restablished on the tenth day. These experiments prove the clear influence of bone extract on activation of bone healing during the first 8 days following explantation.

    Notes: Summary Renal cortical and plasma renin activities and the electron microscopic appearance of juxtaglomerular epitheloid cells were investigated in the unclamped renin-depleted kidney of rats with two-kidney-Goldblatt hypertension after removal of the clamped kidney. Within 24 h the renal renin content fell to still lower values, on the 2d day, it rose to one half its normal value, on the 7th day it reached its control value. Renin activity of plasma did not significantly differ from zero until the 3d day after removing the clamped kidney, reached 20% of normal on the 7th day and normal values between the 7th and the 21 st day. Sparse secretory granules appeared in the remaining renal cortex 12 h after removal of the clamped kidney. After 24 h secretory granules were formed by confluence of Golgi vesicles. After 2 days cisternae appeared in the rough-surfaced endoplasmic reticulum. After 4 days the electron-microscopic picture became normal. Renin appears to be elaborated in the rough-surfaced endoplasmic reticulum and concentrated into granules by the Golgi-apparatus. A fraction of the renal renin does not appear to be available for secretion.

    Notes: Zusammenfassung Durch in bestimmten Zeitabständen aufgenommene Mikrophotographien einzelner Zellkerne von in vitro gezüchteten Osteoblasten und karyometrische Auswertung der so erhaltenen Filme haben wir das intermitotische Kernwachstum untersucht. Besondere Aufmerksamkeit wurde auch dem Studium der möglichen Methodefehler geschenkt. In einer ersten, verhältnismäßig kurz dauernden postmitotischen Phase nimmt der Zellkern sehr rasch an Größe zu und zwar oft in einzelnen Wachstumsschritten, denen jeweilen eine Volumenvergrößerung um den Faktor √2 entspricht. In einer zweiten, stundenlang dauernden Phase, in der das Kernvolumen verdoppelt wird, erfolgt das Wachstum viel weniger schnell, besonders langsam in der Mitte des Verdoppelungsschrittes (also nach einer Volumenzunahme um √2). Die Größenzunahme von Kernen und Nukleolen erfolgt in einzelnen Schüben starken Wachstums, unterbrochen von Perioden verlangsamten oder eingestellten Wachstums („Ruheperioden“) und von vorübergehenden Wachstumsrückschlägen.
