Zusammenfassung Zur Erzielung guter Fixationsergebnisse an der Stria vascularis, Prominentia spiralis und dem Ligamentum spirale des Meerschweinchens erwies sich Glutaraldehyd unter den von uns gewählten Fixationsbedingungen als ungeeignet, wobei starke Artefakte nach intravasaler und intracochleärer Perfusion der untersuchten Gewebe, geringe Artefakte nach Immersionsfixation auftraten. Mögliche Ursachen der Artefaktbildungen werden diskutiert. Keine Artefakte sahen wir nach Osmiumfixation, die wir in Form der intracochleären Perfusion und Immersionsfixation untersuchten.

Summary We have been examining the electron microscopical structure of the vascular stria, the spiral prominence and the spiral ligament in guinea-pigs. Fixation has been achieved by intravascular or intracochlear perfusion or immersion technique. Primary agents for fixation were glutaraldehyde or osmium tetroxide. Using glutaraldehyde, artefacts were produced in each instance. They consisted in the formation of intramitochondrial vacuoles with the immersion fixation, additional cellshrinking and enlargement of intercellular spaces could be seen with the perfusion fixation. After intravascular perfusion, those artefacts were located in the vascular stria, after intracochlear perfusion in the spiral ligament and in the stroma of the spiral prominence. Artefacts after intravascular perfusion with glutaraldehyde could not be prevented even after changing the fixation conditions. The reason for the artefact formation will be discussed further. No artefact formation could be seen using osmium tetroxide for intraeochlear perfusion or for the immersion technique.
