Summary A simple colorimetric screening procedure using the tetrazolium salt, MTT, has been adapted for use in determining the cytotoxic effects of agents to nonproliferating cells from murine-derived, primary lymphocyte cultures. Moreover, cytotoxicity from short-term exposure to test compounds is assessed rapidly by this technique. The formazan crystals produced from each culture, a byproduct of the enzymatic conversation of MTT by the mitochondria of living cells, are solubilized and read on a spectrophotometer. This assay is used to differentiate the possible immediate cytotoxic effect of a test compound from a noncytotoxic, immunosuppressive effect as determined by other means. The general application for this procedure is for a rapid assessment of the viability of resting cell cultures by nonvisual, nonradioisotopic means.
