    Notes: SUMMARY— The fatty acid composition of the lipids extracted from Georgia Red and Centennial varieties of sweet potatoes was studied to determine changes during storage at 15.5, 10, and 4.5°C. The two varieties did not differ initially in the relative proportions of fatty acids. However, changes in fatty acid composition were noted during storage and appeared to be mere pronounced at low storage temperatures. The most consistent changes noted were an increase in tetra-cosaenoic acid and a decrease in short chain saturated acids. The Centennial variety contained higher levels of total lipids, which were generally reflected in higher levels of the three fractions, (1) non-phospholipids, (2) cephalin and (3) lecithin. The increase in total lipids and the individual lipid fractions with storage is indicative of two processes that may have occurred in the stored roots. The lipids may have become more extractable as the respiring potato underwent compositional changes, or lipids were being synthesized from non-lipid components. Although changes observed in the relative proportions of fatty acids during storage at different temperatures were not always consistent, the possibility is suggested that changes in fatty acid composition may be related to changes in quality of the potato during storage.

    Notes: 9 Varieties of sweetpotatoes (lpomoea batatas) were harvested from con trolled experimental plots, graded, cured 5 to 7 days and stored for 5 months at 15.5°C and 80-85% r.h. Centennial, Canbake, Georgia Red, Copperskin Goldrush, Gem, Coastal Sweet, Julian, Nuggett and Puerto Rico were analyzed after curing and after storage for °Brix, acidity, solids, total lipids, levels of nonphospholipids, cephalin, lecithin and fatty acid composition of each lipid. Variations in mean measurements were °Brix 13.5–15.7, solids 24.6–31.8, pH 5.92–6.06; acidity 0.134–0.178 and lipids 1.21–2.50. Each of these was significantly influenced by variety of potato and by storage. Most of the fatty acids in total lipids and lipid fractions were influenced significantly by variety and storage of potatoes. Although changes in many of these measurements were significantly correlated, it was correlated that most of the changes during this “ideal” storage were not particularly related to variety since very few variety-by-storage interactions were statistically significant.

    Notes: Abstract The availability of high quality cDNA libraries is often crucial to the successful identification and characterization of genes. The concepts and potential pitfalls of constructing cDNA libraries are presented. Various applications requiring high quality cDNA libraries are outlined, including large-scale single pass sequencing of cDNA clones to generate expressed sequence tags (ESTs) and differential screening of cDNA libraries. The usefulness of combining such approaches for the discovery of novel disease-related and cardiovascular-based ESTs (CVBest) is discussed.

    Notes: Abstract The efficacy of adding nitrogen to a Co-Cr surgical implant alloy in order to improve tensile and fatigue properties has been investigated. Using the heat treatments described in Part 1 of this study, the tensile properties of specimens with nominally 0.14, 0.19, 0.21 and 0.33 wt% carbon were evaluated in air at room temperature. The fatigue testing consisted of a rotating beam fatigue test at room temperature in air, at a frequency of 10 Hz. The results of the mechanical tests indicated that interstitial nitrogen additions to low carbon alloys (nominally 0.14 wt % carbon) increased the yield strength while maintaining good ductility. However, this beneficial effect was not apparent for the material tested in fatigue, possibly because of the overriding influence of inherent flaws within the heat treated testpieces.

    Notes: Abstract The determination of the feasibility of adding nitrogen to a cobalt-chromium implant alloy was undertaken with the ultimate goal of the work being the improvement of the static and fatigue properties of the alloy. Nitrogen additions were made using high-temperature heat treatments in a nitrogen-containing gas atmosphere. The effects of the nitrogen additions were characterized in this study using several techniques. The maximum solid solubility of nitrogen in the alloy at 1200° C (the heat-treatment temperature) was found to be approximately 0.35 wt % N. X-ray diffraction using nitrogen heat-treated powder samples indicated that the addition of nitrogen in solution resulted in a lattice dilation lying in the range of 0.0021 to 0.0035 nm per wt % N. Above the solubility limit, Cr2N and Cr2(CN) were present at the nitrogen heat-treatment temperatures in the form of large second-phase particles. Ageing of the alloy containing approximately 0.35 wt % N at 400° C resulted in the precipitation of CrN. A study of the nitrogen distribution suggested that the diffusion of nitrogen ms affected by the carbon content of the cobalt-base alloy.
