    Notes: Abstract The basidiomycete Coriolopsis gallica decolorizes alkaline paper effluents efficiently. In this work, we found that C. gallica produces laccase during this decolorization process. This enzymatic activity was produced in all media studied; however, the highest enzymatic activity was obtained in a medium containing paper effluent, where laccase was detected on the 2nd day of the experiment. The laccase activity of C. gallica was purified and characterized. The amino-terminal sequence of this protein showed the highest similarity with the laccase I of the basidiomycete PM1 and with Coriolus hirsutus laccase.

    Notes: Abstract A protocol was developed for Agrobacterium-mediated transformation of embryogenic suspension cultures of cassava. The bacterial strain ABI containing the binary vector pMON977 with the nptII gene as selectable marker and an intron-interrupted uidA gene (encoding β-glucuronidase) as visible marker was used for the experiments. Selection of transformed tissue with paromomycin resulted in the establishment of antibiotic-resistant, β-glucuronidase-expressing lines of friable embryogenic callus, from which embryos and subsequently plants were regenerated. Southern blot analysis demonstrated stable integration of the uidA gene into the cassava genome in five lines of transformed embryogenic suspension cultures and in two plant lines.

    Notes: Summary The in vitro decay of Aextoxicon punctatum and Fagus sylvatica wood by the fungi Trametes versicolor, Ganoderma australe, Phlebia chrysocrea and Lentinus cyathiformis was studied by the agar-block method, and then the decayed woods were analyzed by chemical and spectroscopic techniques. The results demonstrated the strong resistance of the A. punctatum wood to the brown-rot fungus L. cyathiformis; the resistance might be related to the low S/G lignin ratio in this Austral hardwood. Wood decay by the Austral white-rot fungi G. australe and P. chrysocrea was rather limited, and preferential degradation of lignin was not produced although all the fungi studied increased wood digestibility. The most characteristic white and brown-rot decay patterns were observed during the in vitro decay with T. versicolor and L. cyathiformis, respectively. Trametes versicolor caused high weight losses and reduced the lignin content of the wood, whereas L. cyathiformis produced a preferential removal of xylan. No important changes in the solid-state 13C NMR spectra were observed after wood degradation by T. versicolor, but this technique evidenced an increase in aromatic carbon by L. cyathiformis. This increase was higher than that found in the Klason lignin content, suggesting the presence of altered lignin fractions in the brown-rotted wood.

    Notes: Abstract The mushroom Flammulina velutipes and the white-rot fungus Trametes versicolor were cultivated separately on sugarcane bagasse for 40 days. Trametes versicolor produced laccase and manganese-peroxidase activities, showing a simultaneous degradation of lignin and holocellulose. However, only phenoloxidase activity was found with Flammulina velutipes. A preferential degradation of lignin was detected in F. velutipes, which exhibited a greater reduction in the ratio of weight loss to lignin loss than T. versicolor. A decrease in the syringyl/guaiacyl ratio observed with both fungi indicated the preferential degradation of non-condensed (syringyl-type) lignin units. An increase in the relative abundance of aromatic carboxylic acids suggested that the oxidative transformation of lignin unit side-chains was occurring. This was more noticeable with Flammulina velutipes than with T. versicolor.

    Notes: Summary We have conducted observations of possible microstructure on some zwitterionic polymers by means of Transmission Electron Microscopy. Two different solvents, with very different evaporation rates, were used to dessicate the samples onto the carbon covered microscope grids. While no sign of microstructure was apparent in the samples prepared from the more volatile solvent, a well defined grainy appearance was observed in the samples obtained from the less volatile solvent. This shows that sample preparation plays an important role on microstructure formation on zwitterionic polymers.

    Notes: Abstract In Chilean rain forest the presence and distribution of yeasts were studied during the fungal transformation of wood, producing a partially delignified material used as cattle feed (“huempe”). A total of 68 yeast species was reported, including a noticeable proportions of basidiomycetous and xylose-assimilating strains, the latter related to the increased availability of this sugar in the decayed wood. The results of the mathematic comparison of taxonomic data of 327 strains showed a delimitation of sharp clusters corresponding to different species, including several new species described from this substrate, and genera, as Rhodotorula and Cryptococcus. At the initial stages of wood decay, characterized by the presence of Candida species, yeast diversity was low. The highest values were found in the medium stage of decay, used as feed, with an important presence of the genera Apiotrichum, Rhodotorula, Cryptococcus and Schizoblastosporion. After establishing a non-linear ordination from Euclidean distances between samples, based on yeast abundances, a relationship was observed between the populations developing at the different stages of wood decay by Ganoderma applanatum. A succession of yeast species was demonstrated during wood degradation by this white-rot fungus.

    Notes: Abstract Four mushroom strains ofPleurotus spp. were cultivated on sugar cane crop residues for 30 days at 26°C. Biochemical changes affected the substrate as a result of fungal growth, in terms of nitrogen, lignin, cellulose and hemicellulose contents. All strains showed a strong ligninolytic activity together with variable cellulolytic and xylanolytic action.Pleurotus sajor-caju attacked lignin and cellulose at the same rate, showing a degradation of 47% and 55%, respectively. A better balance was shown by theP. ostreatus-P. pulmonarius hybrid, which exhibited the poorest cellulolytic action (39%) and the highest ligninolytic activity (67%). The average composition of mushroom fruit bodies, in terms of nitrogen, carbohydrates, fats and amino acid profiles, was determined. Crude protein and total carbohydrate varied from 23% to 33% and 36% to 68% of dry matter, respectively. Fat ranged from 3.3% to 4.7% and amino acid content from 12.2% to 22.2%. Slight evidence for a nitrogen fixing capability was encountered in the substrate to fruit body balance.
