    Notes: Acid hydrolysis of dried fodder's yeast Candida utilis was studied by a central composite design taking into account three independent variables: temperature (105–165C), processing time (30–70 min) and HCl concentration (7.5–17.5%). Response variables were: color, dry matter solubilization and amino nitrogen. Temperature was the most significant variable followed by acid concentration and processing time. Response surfaces were generated from fitted polynomials. Higher temperatures to prevent amino nitrogen degradation were not investigated. As different effects were found for color development and amino nitrogen, a combined function was tested which yields the behavior of all responses simultaneously. The best conditions were: 150C, 15% HCl and 40 min. After neutralization with 6N NaOH and centrifugation, the raw sauce was supplemented with 12% NaCl, 0.1% monosodium glutamate, 2% vinegar and 0.1% sodium benzoate and evaluated by sensory analysis for eight distinct attributes. Imitation sauce from yeast was at least as good as commercial hydrolyzed soybean sauce.

    Notes: Abstract Two strains of Pleurotus spp., grown in solid state fermentation on sugar-cane straw, degraded the dry matter by 50% after 60 days. The rate of substrate consumption and the dry weight of fruiting bodies decreased in consecutive flushings. Both strains vigorously attacked hemicellulose (80% of total degradation) and lignin (70%). Fruiting bodies were rich in protein and lipids, and had a low content of carbohydrates and ash.

    Notes: Abstract Four mushroom strains ofPleurotus spp. were cultivated on sugar cane crop residues for 30 days at 26°C. Biochemical changes affected the substrate as a result of fungal growth, in terms of nitrogen, lignin, cellulose and hemicellulose contents. All strains showed a strong ligninolytic activity together with variable cellulolytic and xylanolytic action.Pleurotus sajor-caju attacked lignin and cellulose at the same rate, showing a degradation of 47% and 55%, respectively. A better balance was shown by theP. ostreatus-P. pulmonarius hybrid, which exhibited the poorest cellulolytic action (39%) and the highest ligninolytic activity (67%). The average composition of mushroom fruit bodies, in terms of nitrogen, carbohydrates, fats and amino acid profiles, was determined. Crude protein and total carbohydrate varied from 23% to 33% and 36% to 68% of dry matter, respectively. Fat ranged from 3.3% to 4.7% and amino acid content from 12.2% to 22.2%. Slight evidence for a nitrogen fixing capability was encountered in the substrate to fruit body balance.

    Notes: The feasibility of using ultrafiltration to concentrate RNA was investigated. Ultrafiltration flux and solute retentivity were affected by membrane cut-off and inlet pressure.The RNA can be concentrated by selecting a hydraulically permeable membrane (PM 30) and then operating the system at a sufficiently high pressure (2.07 · 105 Nm-2) and a high recirculation rate (150 cm-1). The use of this procedure was limited to a high-flux regime. However, a very high retentivity of macrosolute was achieved. The effect of transmembrane pressure on the apparent mass transfer coefficient in the “gel polarization model” was discussed and related to ultrafiltration flux.

    Notes: Magnesium and potassium ions are present in sugar cane molasses in a concentration of about 0.3% and 2.5%, respectively, which is high enough to support biomass production from Candida utilis. Culture broth with 40 g/l of total reducing substances supplemented by the addition of 1 ppm of Mg2+ leads to a higher yield (Y x/s) in batch fermentation experiments. The subsequent addition of Mg2+ up to 10 ppm decreases the yield coefficient from 0.53 to 0.42 without affecting the growth rate. Fermentation media supplemented with 1 to 10 ppm of K+ decreased both the yield coefficient and the specific growth rate. A Mg/K ratio of about 0.1 seems to be optimal for yeast biomass propagation.

    Notes: Growth inhibition of Candida utilis NRRL Y-660 took place in molasses stored at 60°C for 120 days. The specific growth rate (μmax) was reduced from 0.42 h-1 to 0.200 h-1 as a result of a lack of affinity from the microorganism to the substrate and the increasing maintenance necessities. The Ks values arose from 1.40 mg/ml to 4.28 mg/ml within the whole experiment. At the same time, the maintenance coefficient (m) increased from 0.250 to 3.80 mg/ml. In a continuous culture the “wash-out” conditions were reached at dilution rate values (D) close to 0.40 h-1. The process productivity decreased up to 15% from its original value in fresh molasses.

    Notes: The study of a heat-shock process for RNA reduction was carried out for different yeast strains. Different results were obtained from each of them. Candida utilis NRRL Y-660 shows its best performance after a 8-s. heat-shock in the presence of 3% NaCl. For commercial baker's yeast Saccharomyces cerevisiae and Kluyveromyces fragilis L-1930, similar results were obtained with only 1% of NaCl. The latter needed longer heat-shock periods. e.g. 15s. to give such an RNA reduction. Biomass recovery ranged from 60 to 75%, being higher for C. utilis and K. fragilis while excessive losses were observed in S. cerevisiae cells.No significant protein deterioration was obtained in the best performance samples. The aminoacid profile appears to be improved in comparison to the starting material in these strains after RNA reduction.
