Summary The fine structure of agapornispox virus was studied in thin sections of infected chorioallantoic membranes and from pellets of chick embryo fibroblast culture supernatant after differential centrifugation. The architecture of agapornispox virus is comparable to vaccinia virus up to a certain point. It was possible to demonstrate more differentiated structural elements. The envelope of complete virus particles consists of three layers: an outer membrane-like coat, enveloping a central globular layer adhered to by an inner (“unit membrane”) membrane-like bordering structure. A lateral body-coat can be found between the inner membrane and the nucleoid limiting structure consisting of 5 layers. With neutral fixative the nucleoid-content proved to consist of fibrillar material of a higher order. A comparison of various stages of virus replication ranging from assembly of immature particles from areas of viroplasm to release of mature virus particles with complete envelope yielded some information on morphogenesis within virus particles. In immature virus particles peripherical bodies of fibrillar and membranelike architecture can be found, which are interpreted as initial stages of subsequent nucleoid content or nucleoid membrane respectively. Membrane-like transitional stages from particle coat to nucleoid coat seem to support the theory that the particle coat is involved in the formation of nucleoid coat.

Zusammenfassung Die Feinstruktur des Agapornidenpockenvirus wurde in Dünnschnitten infizierter Chorioallantoismembranen und in Pellets differentialzentrifugierter Kulturüberstände aus Hühnerembryo-Fibroblastenkulturen untersucht. Der Aufbau des Agapornidenpockenvirus ist weitgehend mit dem des Vacciniavirus vergleichbar, es können jedoch differenziertere Strukturen aufgezeigt werden: Die Hülle kompletter Partikeln bestellt aus drei Schichten. Eine äußere membranartige Grenze umschließt eine globuläre Mittelschicht, der nach innen wieder eine membranartige Grenzstruktur anliegt. Zwischen der inneren Grenzstruktur und der fünfschichtigen Nukleoidgrenze findet sich ein Lateralkörpermantel. Der Nukleoidinhalt erweist sich bei neutralem Fixierungsgemisch als höhergeordnetes fibrilläres Material. Der Vergleich verschiedener Stadien der Virusvermehrung von der Abschnürung unreif er Partikeln aus Viroplasmabezirken bis zur Ausschleusung reifer Virusformen mit kompletter Hülle ergibt Hinweise zur Morphogenese der Viruspartikeln. In unreifen Viruspartikeln finden sich Innenkörperchen mit fibrillärem und membranartigem Aufbau, die als Vorstufen des späteren Nukleoidinhaltes bzw. der Nukleoidgrenzstruktur interpretiert werden. Membranartige Übergänge von der Partikelgrenze zur Nukleoidgrenze sprechen für eine Beteiligung der Partikelgrenze an der Bildung der Nukleoidgrenze.
