    Notes: Summary A numerical method is treated for solving singular boundary value problems with solutions that can be represented as series expansions on a subinterval near the singularity. A regular boundary value problem is derived on the remaining interval, for which a difference method is used.     Notes: Abstract Treatment of metastatic malignant melanoma with interferon α (IFNα) results in objective remission in approximately 15% of patients. In a previous investigation, we found that about 50% of the patients achieved at least minor or short-lived remissions. In some tumours extensive areas of regressive tumour change occurred. However, even in these areas remnants of tumour cells were generally found. The short duration of the immune response in some patients and the incomplete eradication of the tumour can be due either to selection of non-immunogenic tumour cells or to down-regulation of the immune reactivity to the tumour. In the present paper, the expression of the ζ chain of the T cell receptor in CD3+ lymphocytes and the expression of CD28 in CD3+, CD4+ and CD8+ lymphocytes was studied in resectable melanoma metastases from 20 treated (IFNα or IFNα in combination with cisplatinum and dacarbazine) and 16 untreated patients. A double-staining technique was used, and the occurrence and distribution of lymphocytes showing down-regulation of the ζ chain or CD28 were separately registered in different areas of the metastases: close to the tumour cells in areas of unaffected tumour growth, in areas with regressive tumour changes, in areas with marked fibrosis and in stromal areas with densely packed lymphocytes. CD3+ζ− lymphocytes were found in all metastases, but their number and distribution varied considerably. Down-regulation of the ζ chain was most often found in areas of regressive changes. In contrast, T lymphocytes infiltrating close to the tumour cells had a stronger expression of the ζ chain (P = 0.016). Down-regulation was also found in stromal areas of densely packed lymphocytes and in areas of fibrosis. The pattern of down-regulation of CD28 in various subsets of lymphocytes was similar to that of ζ chain. The same pattern of down-regulation of CD28 and the ζ chain was found in both untreated and treated patients, indicating that the down-regulation is not due to treatment but to the release of immunosuppressor factors from areas with high tumour cell density or extensive destruction of tumour cells. These results concur well with the view that IFNα treatment can result in immune-mediated tumour cell destruction early in the treatment period and that this immune response to the tumour can be followed by immunosuppression within a few weeks.

    Notes: Summary Uptake, distribution and turnover of 5-Hydroxytryptamine (5-HT) was studied by cytofluorometric analysis of whole mast cells and individual granules. Injection of 5-HT as well as 5-Hydroxytryptophan (5-HTP) intraperitoneally or subcutaneously resulted in a parallel uptake of 5-HT in cells and granules. Intraperitoneal injections of 5-HT in such small quantities that may be available under physiological conditions resulted in an increase in fluorescence intensity of the mast cells, indicating a very efficient uptake mechanism for 5-HT in vivo. Much larger doses of 5-HTP were required to obtain a corresponding uptake of 5-HT in the mast cells. The 5-HT was rather rapidly taken up in the granules and eliminated very slowly, at the same rate both from granules and mast cells. The low elimination rate confirms our previous findings that the turnover of 5-HT is much lower in mast cells than in other amine containing cell systems. The combination of an extremely efficient, rapid uptake of 5-HT with a slow elimination suggests a specific function for mast cells in the regulation of free amine concentrations in tissues.

    Notes: Abstract High-order centered finite difference approximations of hyperbolic conservation laws are considered. Different ways of adding artificial viscosity to obtain sharp shock resolution are proposed. For the Riemann problem simple explicit formulas for obtaining stationary one- and two-point shocks are presented. This can be done for any order of accuracy. It is shown that the addition of artificial viscosity is equivalent to ensuring the Laxk-shock condition. Numerical experiments verify the theoretical results.

    Notes: Abstract We consider the steady state Stokes equations, describing low speed flow and derive estimates of the solution for various types of boundary conditions. We formulate the boundary conditions in a new way, such that the boundary value problem becomes non-singular. By using a difference approximation on a staggered grid we are able to derive a non-singular approximation in a direct way. Furthermore, we derive the same type of estimates as for the continuous case. Numerical experiments confirm the theoretical results.

    Notes: Abstract We present a fast high-order Poisson solver for implementation on parallel computers. The method uses deferred correction, such that high-order accuracy is obtained by solving a sequence of systems with a narrow stencil on the left-hand side. These systems are solved by a domain decomposition method. The method is direct in the sense that for any given order of accuracy, the number of arithmetic operations is fixed. Numerical experiments show that these high-order solvers easily outperform standard second-order ones. The very fast algorithm in combination with the coarser grid allowed for by the high-order method, also makes it quite possible to compete with adaptive methods and irregular grids for problems with solutions containing widely different scales.
