    Notizen: Aims : NANOG is a key regulator of embryonic stem cell (ESC) self-renewal and pluripotency. Our recent genome-wide gene expression profiling study of the precursor of testicular germ cell tumours, carcinoma in situ testis (CIS), showed close similarity between ESC and CIS, including high NANOG expression. In the present study we analysed the protein expression of NANOG during normal development of human testis and in a large series of neoplastic/dysgenetic specimens.Methods and results : We detected abundant expression of NANOG in CIS and in CIS-derived testicular tumours with marked differences; seminoma and embryonal carcinoma were strongly positive, differentiated somatic elements of teratoma were negative. We provide evidence for the fetal origin of testicular cancer as we detected strong expression of NANOG in fetal gonocytes up to gestational week 20, with subsequent down-regulation occurring earlier than for OCT-4. We detected no expression at the protein level in normal testis.Conclusions : NANOG is a new marker for testicular CIS and germ cell tumours and the high level of NANOG along with OCT-4 are determinants of the stem cell-like pluripotency of the preinvasive CIS cell. Timing of NANOG down-regulation in fetal gonocytes suggests that NANOG may act as a regulatory factor up-stream to OCT-4.

    Notizen: The identity of cytokinins extracted from leaves of Begonia×cheimantha Everett cv. Nova has been studied by purification and fractionation on Dowex 50 H’ and Sephade× L.H 20 columns followed by reverse-phase HPLC separation and determination by enzyme immunoassay. The following four cytokinins were identified: trans-zeatin, trans-zeatin riboside, isopentenyladenin and isopentenyladenosin. At least three other compounds, possibly glucosides, were detected by activity in the tobacco callus bioassay but not yet identified. The quantity of the identified cytokinins was several times higher at 15 than at 24°C, the former temperature being optimal for regeneration of adventitious buds. At 15°C there was no clear effect of photoperiod whereas short days at 24°C increased the activity several-fold compared with long day conditions.

    Notizen: Summary Plasma levels of nortriptyline and perphenazine were measured in six patients on continuous nortriptyline treatment before, during and after oral administration of perphenazine 4 mg t.i.d. In four patients the plasma levels of the conjugated and unconjugated principal metabolite 10-hydroxy-nortriptyline were also measured. Urinary excretion of conjugated and unconjugated 10-hydroxy-nortriptyline and plasma levels of perphenazine were determined in all six patients. During treatment with perphenazine two patients showed a slight increase in the plasma level of nortriptyline. The changes in metabolite excretion rate were inconclusive. Thus, there did not appear to be any important pharmacokinetic interaction between the two drugs at the doses used, which were normal therapeutic doses. The previously reported inhibitory effect of perphenazine on the metabolism of nortriptyline probably depended therefore, either on administration of a higher dose of perphenazine, or on treatment in the reverse sequence — a single dose of nortriptyline was given to patients already receiving perphenazine.

    Notizen: Abstract Auxin (11 μM α-naphthaleneacetic acid) and cytokinin (1.4 μM kinetin) regulate cytokinin accumulation by cytokinin-requiring (C-) and cytokinin-autotrophic (C+) lines of Havana 425 tobacco (Nicotiana tabacum L.) tissues. No trans-zeatin riboside (ZR) (〈0.5 pmol·g-1 fresh weight) was detected in six C- and nine C+ lines grown for 14 d on auxin + cytokinin and auxin medium, respectively. C+ lines, but not C- lines accumulated ZR (1.9–5.1 pmol·g-1 fresh weight) when incubated on hormone-free medium but both lines accumulated ZR when incubated on kinetin medium. Therefore, it appears that kinetin treatment can induce ZR accumulation and that this accumulation is blocked by auxin treatment. Similar effects were obtained with some lines of cells autotrophic for both auxin and cytokinin. Tobacco plants carrying the dominant Habituated leaf-1 allele (Hl-1) differ from wild-type plants in that leaf-derived tissues in culture exhibit a C+ phenotype. No differences in ZR content were found in C+ leaf tissues from Hl-1/Hl-1 plants and C+ tissues that arise epigenetically in wild-type plants. This indicates that the H-1 allele does not act to induce overproduction of ZR. The Hl-1 allele is known to have oncogenic functions similar to the isopentenyl transferase (ipt) locus of the Ti plasmid. Although Hl-1/Hl-1 cells transformed with Ti plasmids defective at the ipt locus are tumorigenic and hormone-autotrophic in culture, they contain low levels of ZR typical of non-transformed Hl-1/Hl-1 cells. Therefore, the high levels of ZR characteristics of cells transformed with wild-type Ti plasmids are not necessary for expression of the tumor phenotype.
