    Description / Table of Contents: Abstract Background: Presentation of current data and knowledge in the field of experimental and clinical neuroendocrinology. Aim: Overview of current data on the pathogenesis and therapeutic strategies of pituitary disorders. 1. Hypothalamic and pituitary regulation: characterization of influence of hormone secretion and cell proliferation of anterior pituitary cells by an intrapituitary network of cytokines and growth factors. Special importance on the role of the interleukin-6-family. Characterization of the role of gaseous transmitters in neuroendocrinology. 2. Molecular pathogenesis of pituitary adenomas: presentation of the current knowledge status with regard to familial syndromes. 3. Therapy of pituitary adenomas: presentation of new developments of therapeutic aspects, in particular medical treatment, surgery and radiation. Outlook on the future role of gene therapy. 4. Growth hormone replacement in adults: short overview of diagnosis and therapy of growth hormone deficiency in the adult.
    Notes: Zusammenfassung Hintergrund: Darstellung der aktuellen Daten und Erkenntnisse im Bereich der experimentellen und klinischen Neuroendokrinologie. Ziel:Überblick über aktuelle Daten zu Pathogenese und therapeutischen Strategien von Hypophysenerkrankungen. 1. Hypothalamisch-hypophysäre Regulation: Charakterisierung der Beeinflussung von Hormonsekretion und Zellproliferation von Adenohypophysenzellen durch ein intrahypophysäres Netzwerk von Zytokinen und Wachstumsfaktoren. Besondere Darstellung der Bedeutung der Interleukin-6-Familie. Charakterisierung auch der Bedeutung gasförmiger Transmitter in der Neuroendokrinologie. 2. Molekulare Pathogenese von Hypophysenadenomen: Darstellung des aktuellen Kenntnisstandes, insbesondere auch unter Berücksichtigung der familiären Syndrome. 3. Therapie der Hypophysenadenome: Darstellung neuer Entwicklungen auf den therapeutischen Gebieten, insbesondere medikamentöse Therapie, operative Maßnahmen und Strahlentherapie. Ausblick auf die zukünftige Bedeutung der Gentherapie. 4. Wachstumshormonsubstitution bei Erwachsenen: kurze Übersicht zur Diagnose und Therapie des Wachstumshormonmangels beim Erwachsenen.

    Notes: Abstract • Background: Experimental evidence suggests a role of endothelin-1 (ET) in the regulation of intraocular pressure (IOP). • Method: Therefore, inpatients undergoing cataract surgery, ET-like immunoreactivity (STIR) was measured by radioimmunoassay in pooled samples of aqueous humor of eyes with primary open-angle glaucoma (POAG) and normotensive eyes with cataract only. • Results: ETIR was significantly (P 〈 0.05) higher in patients with cataract and POAG (20.5 ± 1.8 pg/ml,n = 12; preoperative IOP 21.4 ± I.1 mmHg,n = 33) than in patients with cataract only (15.8 ± 1.6 pg/m1,n = 15; preoperative IOP 16.0 ± 0.6 mmHg,n = 77). • Conclusion: This finding may indicate a role of ET in POAG or ocular antihypertensive treatment, and its relevance should be further investigated.
