Notes: 26 different compounds have been investigated experimentally for their sensitizing capacity in guinea pigs. 19 of these occur in propolis as well as in poplar bud exudates, and 14 of them are also found in balsam of Peru. 4 caffeates and benzyl isoferulate were found to be strong Sensitizers. 7 compounds were moderate, and 13 compounds showed only weak sensitizing potency. Methyl cinnamate was negative. Patch tests in 11 propolis-sensitive patients once more revealed 3-methyi-2-butenyl caffeate and phenylethyl caffeate as the major sensitizers. In addition to the 8 compounds already known to occur in propolis as well as in balsam of Peru, we detected 5 further substances that both materials have in common. Among these, benzyl isoferulate is considered a noteworthy sensitizer. Coniferyl benzoate, which was shown to be a moderate sensitizer, is present in fresh samples of balsam of Peru, while in propolis it has been detected only once so far. The flavonoid aglycones occurring in poplar bud exudates, and hence also in propolis, are weak sensitizers which play only a minor role m propolis hypersensitivity.

Notes: Nail polish that has completely dried on the fingernails contains water-soluble components that attain the skin during extensive but transient contact. This was proven by water extraction of thin layers of nail polish that had been painted onto glass plates and allowed to dry for 3 days. Comparing the isolated fractions and compounds with known nail polish ingredients revealed that the water-soluble substances are para- and ortho-toluenesulfonamide. Dihuty1 phthalate and 3 constituents of toluenesulfonamide-formaldehyde resin (TS-F-R), which is the basic material of almost all nail polishes sold worldwide. 12 female patients with proven nail polish allergy were patch tested with 21 nail polish components, including those isolated. Only 2 fractions were positive. These contained a monomer and a dimer created during condensation of TS-F-R. Their structures were elucidated. The 3rd compound, a trimer, remained negative, except in 1 case.

Notes: Primin (2-methoxy-6-pentyl-1,4-benzoquinone)is a naturally-occurring strong sensitizer from Primula obconica (Primulaceae), To determine the effect of side-chain length on senstizing potency 15 analogues with linear side chains from to from C1, to C15 and 4 C6- analogues with branched side chains were prepared synthetically and devoted to experimental sensitization in guinea pigs The results showed an increase of the sensitizing capacity with increasing length of the alkyl side chain from C1 to C10, reaching a maximum at C11 and C12. On further elongation the senstizing potency decreased beyond C13 reaching values which finally were as low as those of the C1 and C3 derivatives. The results mirror finding which finally were as low as those of the C1 and C3 derivatives. The results mirror findings which formerly have been obtained with other non-quinonoid compounds like catechols, phenols hydroquinones and gallates. In the plant kingdom. Compound approximating an “ideal allergen” consisting of a quinonoid ring with a 10 or 11 carbon-remarkably strong sensitizer found in Wigandia caracasana (Hydrophyllaceae)

Notes: 7 resin acids, 3 symbolically prepared derivatives and the neutral fraction of Chinese colophony were studied by experimental sensitization using a modified FCA method. 4 resin acids, laevopimaric. abietic. podocarpic and tetrahydroabietic. proved to be weak sensitizers Neoabietic, dehydroabietic, isopimarie acid and larixol remained negative. However, the derivative, methyl abietate and calcium abietate. were shown to be moderate sensitizers. and the maleic-modified adduct to be a moderate to strong sensitizer. As laevopimaric acid plays only a minor rote in colophony, abietic acid must he considered a major, even if weak, allergen in non-modified colophony. The neutral fraction of colophony is also partially involved in colophony allergy.

Notes: Irritancy of daffodil flowers and bulbs was assessed using various fresh plant preparations, solvent extracts and some of the known Amaryllidaceae alkaloids on guinea pigs. Sensitization was also carried out on guinea pigs using these plant preparations. solvent extracts and 7 fractions obtained after preparative chromatography of the bulb ether extract. Only 1 fraction, containing 2 alkaloids, was capable of inducing delayed hypersensitivity in the animals: the sensitivity achieved, however, was weak. The substances were identified as masonin and homolycorin, which acted as elicitors. but masonin may also be a sensitizer. While homolycorin is a known daffodil constituent, masonin has not been found previously in Narcissus pseudonarcissus. 3 other alkaloids as well as chelidonic acid and isorhamnetin were non-elicitors in the sensitized guinea pigs.

Notes: A 15-year-old girl developed perioral dermatitis and swelling of the lips, having played a flute made of cocus wood for 2 years. Patch tests with wood shavings, as well as with subsequently isolated constituents named focus 1 and focus II. were Strongly positive. Alter complete purification, boltContact allergens were identified as isoflavan derivatives, While Cocus 1 was shown to be 7,8-dihydroxy-2,4,5trmethoxyisoflava, the structure of focus II could not be elucidated exactly. It could have been Either 7,8,3′-trihydroxy 2,4 dimethoxyisoflavan or 7,8,2-trihydroxy-3′,4-dime-thoxyoflavan The exact position of the hydroxy and methoxy group in C-2 and C-3 can be determined only by complete synthesis Of both compounds or by isolation of greater amounts of focus II for further oxidation experiments. Due t0 the stronger patch lest reaction and the higher yield cocus I in the wood, this hydroxyisoflavan must be recognized as the main contact allergen.

Notes: Experimental and chemical investigations revealed that common ivy (Hedera helix susp. Helix) contains 3 compounds which are powerful irritants and moderate sensitizers. Only 2 of these constituents, falcarinol and didehydrofalcarinol, are present in the plant during the whole year. Besides Panax ginseng and Schefflera arboricala, this is the third species of the Araliaceae in which these polyacetylenic sensitizers have been found. Falcarinol and didehydrofalcarinol also occur in Hedera helix subsp. canariensis. 4 patients have been patch tested. Even in low concentrations (0.03%). the main allergen falcarinol elicited strong reactions in all of them. One of the authors became sensitized during the investigations.
