    Notes: Antibodies binding the Ro (or SSA) and La (or SBB) proteins are commonly found in a high proportion of sera from patients with systemic lupus erythematosus or Sjögren's syndrome. The mechanism by which these autoantibodies arise is not known. Others and we have shown that immunization of nonautoimmune-prone mice with short peptides from the Ro ribonucleoprotein particle can induce autoimmunity to 60 kDa Ro and 52 kDa Ro as well as to the 48 kDa La protein after epitope spreading. We have explored the differences in the epitope spreading after 60 kDa Ro peptide immunization in several strains of mice. There is intra- and intermolecular diversification of the immune response after immunization of DBA/2J animals with a monomer peptide representing the residues 480–494 of the 60 kDa Ro protein, but this peptide does not induce epitope spreading when used as the immunogen in either C57Bl/6J or PL/J mice. Similar to previously studied BALB/c mice, DBA/2J mice have antibodies binding many epitopes of 60 kDa Ro, and some sera bind 52 kDa Ro as well as La. These mice have antinuclear antibody in their sera. These data demonstrate that Ro peptide immunization results in different outcomes depending upon the strain of mouse used. Furthermore, these data suggest that genetic variation is important with regard to responding towards short peptide immunization by epitope spreading.

    Notes: Intermolecular spreading of humoral autoimmunity to different components of the Ro (SS-A) and La (SS-B) ribonucleoprotein (RNP) complex has been reported following immunization with a single component of the complex. Although the immune response to the immunizing antigen is polyclonal and diversified, little is known about the specificity of the recruited autoimmune responses to the endogenous Ro and La antigens which drive B-cell spreading. To determine the specificity of intermolecular spreading to La, we examined sera from 52 kDa Ro (Ro52)- and 60 kDa Ro (Ro60)-immunized C3H/HeJ mice for reactivity with recombinant fragments spanning endogenous mouse (m)La by enzyme-linked immunosorbent assay (ELISA) and immunoblotting. Sera from mice primed and boosted with recombinant Ro52 and Ro60 showed reactivity restricted to the COOH-terminal fragment of mLa (aa361–415). The recruited anti-La response was species-specific, cross-reacting weakly with the corresponding region on the human La molecule, and was abrogated by the preabsorption of the Ro-immune sera with mLa 361–415. Analogous experiments using recombinant mRo60 fragments spanning the mRo60 molecule revealed a similar pattern of oligoclonality in the specificity of anti-Ro60 autoimmunity following active immunization with La and Ro52. These results suggest that intermolecular–intrastructural T–B help is limiting in this model, and reveal unsuspected immunodominance of selected Ro–La epitopes in the spreading of the autoantibody response to these structures. The focusing of the recruited autoantibody response to these COOH-terminal regions of the Ro and La polypeptides may also reflect the surface accessibility of these regions in La–Ro RNP.

    Notes: Abstract The architecture of an intelligent multistrategy assistant for knowledge discovery from facts, INLEN, is described and illustrated by an exploratory application. INLEN integrates a database, a knowledge base, and machine learning methods within a uniform user-oriented framework. A variety of machine learning programs are incorporated into the system to serve as high-levelknowledge generation operators (KGOs). These operators can generate diverse kinds of knowledge about the properties and regularities existing in the data. For example, they can hypothesize general rules from facts, optimize the rules according to problem-dependent criteria, determine differences and similarities among groups of facts, propose new variables, create conceptual classifications, determine equations governing numeric variables and the conditions under which the equations apply, deriving statistical properties and using them for qualitative evaluations, etc. The initial implementation of the system, INLEN 1b, is described, and its performance is illustrated by applying it to a database of scientific publications.

    Notes: Abstract The KT-1000 was used to measure anterior tibial displacement in three populations: normal subjects (n=120), patients with unilateral acute anterior cruciate ligament (ACL) disruptions (n=105), and patients with chronic unilateral ACL disruptions who were scheduled for ACL reconstructions (n=159). All patients with ACL disruptions were measured with and without anesthesia. Tibial displacement under three loading conditions was measured: 89 N anterior displacement force, manual maximum displacement force, and quadriceps contraction to lift the leg. The measurements of the normal knee in the injured populations were not significantly different from those of the knees in the normal population on any test. The injured knee tested with and without anesthesia was significantly different from the normal knee on all tests. The right-left difference in the normal population as less than 3 mm in 98% of patients in the 89-N test, 97% in the manual maximum test, and 99% in the quadriceps active test. The largest amount of displacement and the greatest difference in displacement between the injured and the normal knee was produced by the manual maximum test. The manual maximum injured-minus-normal knee displacement was 3mm or more in 99% of patients with chronic ACL disruptions and in 95% of patients with acute ACL disruptions.

    Notes: An inverse method is presented which accurately determines the pressure distribution for assigned wall shear in a two-dimensional, laminar, incompressible boundary layer. The method reformulates the mechul function scheme of Cebeci and Keller to produce a stable solution in the marching direction and to increase accuracy in the normal direction. In the reformulation a modified pressure gradient parameter variation in the normal direction is used in conjunction with three-point backward differences for streamwise derivatives and fourth-order accurate splines for normal derivatives. The resulting spline-finite difference equations are solved by Newton-Raphson iteration together with partial pivoting. Numerical solutions are presented for self-similar and non self-similar flows and compared with published results.

    Notes: A unified approach is presented for solving the two-dimensional incompressible boundary layer equations. Solutions are obtained for direct and inverse options using the same equation formulation by a simple interchange of boundary conditions. A modified form of the mechul function scheme obtains inverse solutions with specification of transformed wall shear, skin friction coefficient or displacement thickness distributions. Direct solutions may be obtained without altering the block tridiagonal structure of the system by simply requiring no corrections on the streamwise pressure gradient parameter. Fourth-order spline discretization approximates normal derivatives with two- and three-point backward differences approximating streamwise derivatives, yielding a fully implicit solution method. The resulting spline/finite difference equations are solved by Newton-Raphson iteration together with partial pivoting. The results of the study demonstrate the importance of proper linearization of all equations. The successful use of spline discretization is also tied to the use of strong two-point boundary conditions at the wall for cases involving reversed flow. Numerical solutions are presented for several non-similar flows and compared with published results.
