    Notes: Summary In newborn infants of various ages, the influence of repeated administration of water and electrolytes on renal excretion of urine, sodium, potassium, free and bound hydrogen ions and on urine osmolarity was studied. In 14- and 21-day-old infants the repeated administration of water and electrolytes was followed by diminution of renal excretion of hydrogen ions and potassium, possibly caused by stimulation of hormonal regulation of ion exchange in the kidney. In animal studies performed with a larger water and electrolyte load, stimulation of renal excretion of sodium was caused by the pretreatment.

    Description / Table of Contents: Zusammenfassung Lithium ist bei jungen Ratten nahezu doppelt so toxisch wie bei 33–240 Tage alten Tieren. Jedoch kann 5 und 15 Tage alten Ratten im Vergleich zu erwachsenen eine subletale Lithiumdosis (75% der LD50) in einem deutlich kürzeren Zeitabstand erneut appliziert werden, um eine gleiche Letalitätsrate (50%) zu erzielen. Nach 2tägiger Vorbehandlung mit einmal täglich 0,15 mval Li+/100 g KG und Belastung mit 0,3 mval Li+/100 g KG am Tage des Diureseversuches konnten bei Ratten ausgeprägte Altersunterschiede in der renalen Lithiumausscheidung mit deutlich altersabhängig unterschiedlichen Auswirkungen auf die Ausscheidung von Natrium und Kalium beobachtet werden. Die Fähigkeit zur renalen Ausscheidung von Lithium entwickelt sich postnatal bei Ratten schneller als die Fähigkeit, zugeführtes Natrium auszuscheiden. Die Altersabhängigkeit der Toxizität von Lithium kann auf Altersunterschiede im pharmakokinetischen Verhalten von Lithium (Verteilung und renale Elimination) zurückgeführt werden.
    Notes: Abstract In 5- and 15-day-old rats, acute toxicity is much higher than in adults. However, in young rats two sublethal doses of lithium (75% of the LD50) can be injected in a shorter time interval than in adults, resulting in the same mortality (50%). After pretreatment of rats with 0.15 meq Li+/100 g b.wt. daily for 2 days and a load of 0.3 meq Li+/100 g b.wt. intraperitonially on the day on which diuresis experiments were performed, a distinct age-dependence in renal lithium elimination was observed, with age-dependent consequences for the elimination of sodium and potassium. In the postnatal period, the ability to excrete lithium matures earlier than the ability to eliminate sodium. Thus the age-dependence of lithium toxicity stems from the age-dependent pharmacokinetic parameters of lithium, such as distribution and renal elimination.

    Notes: Abstract The kinetics of vitamin E was followed in serum, liver and kidney of 10- and 55-day-old rats after the administration of a single i.m. dose of 100 mg α-tocopherol acetate/100 g body wt. The basal levels without vitamin E administration were significantly higher in serum and liver of 10- than 55-day-old rats. The effect of vitamin E on cisplatin (CP; 0.6 mg/100 g body wt., i.p.) nephrotoxicity was investigated by determining urinary volume and protein excretion, as well as the concentration of blood urea nitrogen (BUN) and lipid peroxides in renal tissue (LPO). Previously described age differences in CP nephrotoxicity were confirmed. The administration of vitamin E, 12 h prior to CP, diminished the toxic effect of CP in young and adult rats. This effect could not be enhanced by a second administration of vitamin E. The simultaneous administration of vitamin E and C 12 h prior to CP intensified the protective effect of a single administration of vitamin E in 10- and 55-day-old rats without influencing the concentration of platinum in renal tissue.
