    Notes: Dendritic arbor development of subventricular zone-derived interneurons is a critical step in their integration into functional circuits of the postnatal olfactory bulb. However, the mechanism and molecular control of this process remain unknown. In this study, we have developed a culture model where dendritic development of purified subventricular zone cells proceeds under serum-free conditions in the absence of added growth factors and non-neural cells. We demonstrate that the large majority of these cells in culture express GABA and elaborate dendritic arbors with spine-like protrusions but they do not possess axons. These neurons expressed receptors for neurotrophins including p75, TrkB and TrkC but not TrkA. Application of exogenous neurotrophins, including brain-derived neurotrophic factor (BDNF), neurotrophin-3 (NT3) and nerve growth factor (NGF), to cultures stimulated dendritic growth and led to more complex dendritic arbors during the initial 3 days in culture. Our results suggest that these effects are independent of Trk receptors and mediated by the p75/ceramide signaling pathway. We also show that brain-derived neurotrophic factor is the only neurotrophin that is able to influence late-phase dendritic development via TrkB receptor activation. These results suggest that dendritic arbor development of subventricular zone-derived cells may be regulated by neurotrophins through the activation of p75 and the TrkB receptor signaling pathways in a sequentially defined temporal pattern.

    Notes: Abstract To correlate presence or absence of a 7q11 microdeletion with the clinical picture of the Williams-Beuren syndrome (WBS), we investigated 29 patients with a clinical diagnosis of WBS or WBS-like features, aged 1–30 years, using molecular analysis and/or fluorescent in situ hybridization (FISH). Deletions at 7q11 were found in 75% of the patients (22 out of 29). Nine deletions occurred on a paternal, and ten on a maternal chromosome; three deletions were demonstrated by FISH only, and parental origin could thus not be determined. All deletion patients aged between 2 years and puberty displayed a distinct pattern of facial features (including periorbital fullness, short nose with flat bridge, wide mouth, and full lips and cheeks), the characteristic outgoing social behaviour, as well as moderate growth and mental retardation. Two-thirds (15 out of 22) had a cardiovascular malformation, but only one third (7 of 22) had supravalvular aortic stenosis (SVAS). A stellate iris pattern was also present in one-third of the patients only. In the four adult patients with 7q11 deletions, there was prominence of the lower lip whereas fullness of cheeks and periorbital tissue was not seen. Conclusion This study confirms that WBS has a unique clinical picture which can be diagnosed clinically, but also shows that the relative frequency of individual features may have been overemphasized in the past, and that a minority of patients may exist who are clinically indistinguishable from WBS but who appear to have no deletion at 7q11.

    Notes: Summary Various benzoic acids 14C-labelled in para and meta methoxyl groups as well as (O-methyl-14C) p-methoxy cinnamic acid were tested for O-demethylation in cell suspension cultures of Phaseolus aureus Roxb. and Glycine max Merr. On the basis of 14CO2-formation and product analyses the O-demethylation reactions were shown to be specific for para methoxyl groups. A vanillate-O-demethylase known from microbial sources seemed to be absent in the plant cell cultures. In this and in an earlier publication (Berlin et al., 1971) some twenty 14C-labelled aromatic compounds were tested for catabolic reactions in the cell cultures, and here we report on the product analyses and the general pattern of distribution of radioactivity. Finally, some indications for compartmentalisation in connection with catabolic studies of aromatic compounds in plant cell cultures are discussed. Decarboxylation of substituted benzoic acids in the cell cultures is restricted to aromatic acids possessing a hydroxyl group in the para position. Only trace amounts of labelled CO2 were released from (carboxyl-14C)-anisic acid. This acid, however, was nearly quantitatively demethylated to p-hydroxybenzoic acid, which itself was decarboxylated to a considerable extent after being fed to cell suspension cultures. Similar differences in respect to decarboxylation were observed with syringic acid produced by demethylation of trimethoxybenzoic acid and syringic acid applied directly to the cell cultures.
