    Notes: Summary Tumor marker identification in testicular cancer has contributed to early detection and monitoring of non-seminomatous disease. A placental alkaline phosphatase-like (PLAP-like) enzyme derived from seminomas has recently been focused upon as a possible marker for this disease. The biochemistry of the PLAP-like enzyme is reviewed, as well as its occurrence in tissue and sera from healthy persons and patients with testicular cancer.

    Notes: Abstract The purpose of this study was to investigate the effect of natural interferon (IFN) on the expression of placental alkaline phosphatase (PLAP) in a human seminoma xenograft in severe combined immunodeficient mice. Mice were injected intramuscularly with 3 × 105 U/mouse of IFN, twice a day, for five consecutive days. A significant increase in PLAP level of the xenografts followed IFN treatment. A radiolabeled anti-PLAP monoclonal antibody (MAb) was intravenously injected on the first day of IFN administration in order to determine if IFN has the potential to enhance the efficacy of an anti-PLAP MAb in the detection of seminoma. Enhanced retention of the anti-PLAP MAb was observed at 7 and 11 days after MAb administration. Thus, IFN treatment appears to have some effect on the efficacy of the anti-PLAP MAb in the detection of seminoma xenografts.

    Notes: Summary The tissue concentrations of a modified nucleoside, pseudouridine, and a normal nucleoside, uridine, were measured with high-performance liquid chromatography. Human kidneys were obtained from five patients with renal cell carcinoma and divided into a noncancerous part and a cancerous part. The pseudouridine concentration in the cancerous part of the kidneys ranged between〈2–2.8 nmoles/g and in the noncancerous part 4.3–19.4 nmoles/g (mean 10,9 nmoles/g). The uridine concentration in the cancerous and noncancerous parts of the kidney ranged between 19.6–179.1 nmoles/g (mean 110.7 nmoles/g) and 117.5–235.6 nmoles/g (mean 191.5 nmoles/g), respectively. The pseudouridine concentration appeared to be approximately seven times higher in the noncancerous part as compared to the cancerous part of the kidney. In the case of uridine, the difference was less pronounced.

    Notes: Summary Using a sensitive and specific method involving high-performance liquid chromatography, urinary levels of four modified nucleosides — pseudouridine (Ψ), 1-methylinosine (m1I), 1-methyladenosine (m1A), and 1-methylguanosine (m1G)-were investigated before and after treatment in 31 patients with cancer of the urinary organs or the female genital tract. Before treatment m1I was the most frequently elevated nucleoside (77%). Pretreatment urinary levels of Ψ, m1I, and m1A in patients with stage 2–4 cancer of the female genital tract were significantly elevated compared to human healthy volunteers (p〈0.005). Compared with the other nucleosides, Ψ appeared to correlate more closely with the clinical outcome (progression or regression) of patients with cancer of the female genital tract. In the case of patients with cancer of the urinary organs, m1I followed the clinical outcome better than the other nucleosides measured. Therefore Ψ and m1I seem to be useful for monitoring genito-urinary cancers.

    Notes: Abstract The potency of aluminum-chloro-tetrasulfonated phthalocyanine (AlS4Pc) as a photosensitizer in photodynamic therapy was evaluated in in vitro and in vivo studies. Compared with hematoporphyrin derivative (HpD), the following advantages of AlS4Pc were revealed: (1) AlS4Pc was less toxic than HpD in vitro without hight irradiation. (2) AlS4Pc showed more photodynamic-dependent cytotoxicity and anti-tumor effect in the red area of the spectrum (〉660 nm) at which tissue penetration is high. (3) AlS4Pc appeared to be removed more rapidly from normal tissues such as muscle and skin. (4) AlS4Pc showed less photodynamic-dependent cytotoxicity in vitro and milder cutaneous phototoxicity in vivo with UVA irradiation. On the basis of these observations, AlS4Pc shows considerable promise as a photosensitizer for PDT.

    Notes: Summary Serum PLAP/PLAP-like enzymes are elevated in testicular cancer patients and also in heavy smokers. Such elevations were measurable with anti-PLAP polyclonal and monoclonal antibodies, and an antigenic heterogeneity was demonstrated in the circulating enzymes. Thus polyclonal antibody revealed more of tumor PLAP-like antigen while a monoclonal antibody H7 detected PLAP of smokers better. PLAP of smokers and PLAP-like enzyme of cancer (seminoma) patients showed similar characteristics with respect to glycosylation pattern, charge and hydrophobicity. By these properties they were differentiated from PLAP of placenta.
