    Notes: A vesicle monoamine transporter was functionally identified, molecularly cloned, and characterized from a human substantia nigra cDNA library. The ATP-dependent transport of 5-[3H]hydroxytryptamine ([3H]5-HT) by digitonin-permeabilized fibroblasts expressing the vesicle monoamine/H± antiporter in culture exhibited a Km of 0.55 μM. Reserpine and tetrabenazine, inhibitors of two monoamine binding sites, effectively blocked [3H]5-HT accumulation with K1 values of 34 and 78 nM, respectively. Pretreatment of cells with as little as 10 nM reserpine in the presence of ATP abolished uptake. The rank order for substrate inhibition of [3H]5-HT uptake for both the previously reported rat vMAT1 and the human transporter clone followed the order 5-HT 〉 dopamine 〉 epinephrine 〉 norepinephrine 〉 1 -methyl-4-phen- ylpyridinium 〉 2-phenylethylamine 〉 histamine. The virtually identical transport characteristics of rvMATI and hvMAT1 confirm the relevance of neuropharmacological studies of rat brain biogenic amine uptake and storage to human brain neurochemistry.

    Notes: Abstract: A sensitive and selective radioimmunoassay for chromogranin A (Chrg A) has been developed to quantitate content, release, and biosynthesis of this secretory protein in neuroendocrine tissues. An antiserum raised against Chrg A from bovine adrenal medulla was found to detect predominantly only the Mr 70–75 kilodalton Chrg A in its native form, allowing the use of this antiserum as a quantitatively specific probe for Chrg A in cell-free extracts of the adrenal medulla and chromaffin cells. Chrg A comprises about 10% of the total protein of the chromaffin cell. It is released in parallel with Met-enkephalin and catechol-amines from the bovine chromaffin cell in primary culture in response to nicotine and nicotinic cholinergic agonists. From 14 to 22% of total Chrg A is released from the cell during a 15-min exposure to a maximally stimulatory dose of nicotine (10–100 μM). Chrg A release on nicotinic stimulation is blocked by D-600 and hexamethonium to the same extent as Met-enkephalin and catecholamine release. The parallel time course and percent release of Chrg A and Met-enkephalin indicate that these secretory polypeptides are contained in, and released from, functionally identical cellular compartments. Chrg A and Met-enkephalin pentapeptide sequences are present in the chromaffin cell at a ratio of about 2:1, although Chrg A is far more abundant on a mass basis. Chrg A and Met-enkephalin biosynthesis appear to be differentially regulated within the chromaffin cell, since chronic treatment of cells with nicotine and forskolin causes an elevation of Met-enkephalin pentapeptide without a concomitant elevation of intracellular levels of Chrg A.

    Notes: Abstract: Exposure of bovine chromaffin cells in primary culture to 0.01–1 μM reserpine caused a dose- and time-dependent increase in intracellular levels of the amidated enkephalin peptide metorphamide. Maximal levels (approximately 800% of control) were obtained at 0.1 μM reserpine and increased levels were apparent by 16 h of treatment. Metorphamide increases were at least fivefold more than that of either Met- or Leu-enkephalin, suggesting that reserpine stimulates both enkephalin processing and amidation in the secretory vesicle. Treatment with elevated potassium, which increases enkephalin levels by stimulating production of preproenkephalin messenger RNA, elicited an increase in metorphamide levels equivalent to, but not greater than, the increase in Met-enkephalin pentapeptide. The ratio of Met-enkephalin to metorphamide in untreated chromaffin cells is approximately 140:1, whereas the final Met-enkephalin: metorphamide ratio in reserpinized chromaffin cells is approximately 30:1, similar to the Met-enkephalin:metorphamide ratio in enkephalinergic neurons of the CNS.

    Notes: Abstract: An upstream enhancer element [tissue specifier element (TSE)] located between 4.66 and 4.02 kb from the transcription start site is important for cell type-specific expression and phorbol ester induction of the vasoactive intestinal peptide (VIP) gene. An element located within 100 bases of the VIP promoter [the VIP cyclic AMP-responsive element (VIP-CRE)] confers cyclic AMP and phorbol ester responsiveness to heterologous promoters. The possibility that these two regions of the VIP gene function cooperatively to determine tissue-specific and second messenger-dependent expression of the VIP gene was addressed by assaying transcription from a VIP-luciferase reporter gene with progressive deletions from the 5′ flanking sequence of the gene, with or without inactivation of the proximal VIP-CRE. Basal expression of the reporter gene in both SH-EP and SK-N-SH human neuroblastoma cells, which express endogenous VIP mRNA, was absolutely dependent on the presence of the upstream TSE. Full constitutive expression was also dependent on the intact VIP-CRE. Forskolin-mediated induction of the reporter gene in SH-EP and SK-N-SH cells was completely abolished by mutations in the VIP-CRE but not by deletion of the upstream sequence, indicating that the VIP-CRE alone determines cyclic AMP responsiveness. In contrast to reports that the VIP-CRE imparts 12-O-tetradecanoylphorbol 13-acetate (phorbol 12-myristate 13-acetate; PMA) responsiveness to heterologous promoters, PMA stimulation in SK-N-SH cells was independent of an intact VIP-CRE but dependent on a region between −2.5 kb and the VIP-CRE. Sequencing of the entire 5.2-kb VIP 5′ flank revealed a consensus PMA-responsive element (TGACTCA) 2.25 kb upstream of the transcription start site that may represent the site imparting PMA responsiveness to the VIP gene.

    Notes: High Tc superconducting materials with nominal compositions of Tl1-xBix(Sr, Ba)2Ca2Cu3O9 (0≤x≤0.5) have been investigated using x-ray powder diffractometry and electron microscopy. It was found that Tl and Bi cations were in an ordered arrangement in the (Tl, Bi)–O atomic layer. Although the proposed supercell is 4a×4b×4c derived from the basic unit cell of the undoped TlBa2Ca2Cu3O9 (Tl-1223), the basic two-dimensional superlattice in the (Tl, Bi)–O layer is 2a×4b or 4a×2b with Bi cations at the center and the corner of the rectangle, giving an ideal and preferred composition with the Tl:Bi ratio of 3:1. Accordingly, the maximum Bi doping for Tl in the solid solution is 25%. The proposed model of the superstructure was supported by computer simulation of electron diffraction patterns. The mechanism of formation of the superstructure is discussed. © 1996 American Institute of Physics.

    Notes: The BrO radical, prepared by the Br+O3 reaction, has been investigated by ultraviolet photoelectron spectroscopy. Two vibrationally resolved bands were observed corresponding to the ionizations BrO+(X 3Σ−)←BrO(X 2Π) and BrO+(a 1Δ)←BrO(X 2Π). These assignments are supported by the results of complete active space self-consistent field/multireference configuration interaction (CASSCF/MRCI) calculations performed as part of this work. The adiabatic ionization energies of these bands were measured as (10.46±0.02) and (11.21±0.02)eV, respectively. Measurement of the vibrational separations in these bands led to estimates of the vibrational constants in the ionic states of (840±30) cm−1 and (880±30) cm−1, and Franck–Condon simulations of the vibrational envelopes gave values of the ionic state bond lengths of (1.635±0.005) and (1.641±0.005) Å for the X 3Σ− and a 1Δ states of BrO+, respectively. The O+Br2 reaction was found to give a band at (10.26±0.02) eV associated with a reaction product. Comparison of the results obtained for the Br+O3 reaction showed that it could not be assigned to ionization of BrO. Calculations of the first adiabatic ionization energies and Franck–Condon simulations of the vibrational envelopes of the first photoelectron bands of BrO2 and Br2O and their isomers demonstrated that this band corresponds to the first ionization of OBrO, the BrO2+(X 1A1)←BrO2(X 2B1) ionization. Franck–Condon simulations were performed with the experimental geometry of BrO2(X˜ 2B1) but with different cationic state geometries. The simulated envelope which most closely matched the experimental envelope gave geometrical parameters of re=1.6135 Å and (angle)OBrO=117.5° for the ionic state. © 2000 American Institute of Physics.

    Notes: Summary. In a series of 368 incontinent women who presented to our urodynamic clinic for assessment, 232 (63%) were diagnosed as having genuine stress incontinence, and 136 (27%) as having detrusor instability. Obesity (〉20% more than average weight for height and age) was significantly more common in women with genuine stress incontinence and detrusor instability than in the normal population. In those with detrusor instability the body mass index was found to increase with age and parity. In women with genuine stress incontinence the body mass index increased with age and the number of previous incontinence operations; it was higher in nulliparous than in parous women. There was no significant difference between obese and nonobese women in any of the urodynamic variables measured in the two incontinence groups.
