    Notes: We have developed at LURE a multiwire proportional chamber with a spherical drift space. The wire chamber consists of two cathode planes, comprising 512 wires with a spacing of 1 mm, set on both sides of the anodic plane. The drift space, a gas filled region bounded by two spherically curved electrodes 144 mm apart, offers several advantages: high quantum efficiency, no parallax effect, equivalent spatial resolution in both directions, and smoothing of the pulsed structure of the synchrotron radiation. The gaseous mixture of argon-xenon (58%), ethane (40%), and ethyl alcohol (2%) is circulated in a closed circuit and is continuously purified. Ethyl alcohol, which avoids electrical discharges and sparks, is essential to operate the instrument at high counting rates (〉300 000 events/s). The signal processing, which makes use of one amplifier per cathode wire and of fast priority encoders, determines both coordinates with a resolution of 1 mm and a dead time of 240 ns. Each encoded event is stored into a 512×512 16 bit CAMAC histogramming memory. The experiment is controlled by a PDP11/34 linked to a VAX by a direct memory access channel. A complete set of programs, which performs the data collection and an off line data reduction, is operational. The adaptation of madnes, a general software package which performs an on line data reduction, is under way. Data, collected on a lysozyme crystal to 3.4-A(ring) resolution, give a reliability factor based on intensities of equivalent reflections of 4.7% without absorption corrections; the variation of the detector efficiency is 〈2.7%. A new version of the instrument is under realization. The position encoder, which uses flash ADCs and signal processors, has a resolution of 0.5 mm. Data are stored into a 1024×1024 16 bit VME histogramming memory linked to a micro VAX.

    Notes: In the solvent contrast variation technique, the structure factor is a linear function of contrast, and the intensity is therefore a parabolic function of contrast. A method has been developed which scales together data measured at different contrasts on different samples. Once the data are scaled, structure-factor amplitudes or intensities for any other contrast can be obtained by inter- (or extra-)polation. The magnitude of the relative phase change between contrast, an important piece of phasing information in single-crystal studies, can be determined.

    Notes: TAOS is a fully automatic system for protein crystallization. Based on a central five-axis robot, it performs vapour-diffusion experiments in commercially available plates with three types of drop geometry, i.e. hanging, sitting or sandwich drops. It can handle ten crystallization plates (up to 240 tests) and prepare precipitant solutions from up to ten different components with no operator intervention. Software help in preparing crystallization experiments and full archiving are provided. TAOS has been thoroughly tested for accuracy and reproducibility.

    Notes: A data reduction system for neutron crystallography using a two-dimensional planar multidetector is described. The method is based on an a priori calculation of the intensity distribution for each reflection for any crystal orientation. The orientation matrix of the crystal has to be known. From the calculated intensity distribution a mask is derived for each reflection. The detection elements of the detector which are inside the mask are summed to determine the intensity of that reflection and the cells outside are used to determine the background. The size of the mask is adapted to the relative height of the measured intensity of the reflector compared to the background. The theoretical intensity distribution for each reflection is calculated in reciprocal space from the primary-beam divergence and wavelength spread and the crystal mosaic spread, on the basis of the kinematic theory of diffraction. The derivation of the necessary equations, i.e. the determination of the resolution function of the instrument, is presented. Some typical results of data collection are also presented. The advantages of this data collection method are the safe determination of weak reflections, the easy discrimination of reflections which nearly overlap, the determination of crystal mosaic spread, and the discrimination of inelastic scattering in the proximity of intense reflections.
