    Notes: Summary The preparation of an antiserum to a purified suspension of adenovirus type 5 hexons is described. The antiserum reacts by complement fixation tests with type 5 and with many other adenoviruses. The anti-hexon serum can be used for the immunofluorescent detection of the site of hexon synthesis within tissue culture cells infected with many different adenoviruses, and therefore allows diagnostic detection of the adenovirus group antigen. In experimentally infected cultures, “group” identification could be made within 24 hours of infection when many different adenoviruses were tested.

    Notes: Summary A method is described for the removal of contaminating (endogenous) protein from high titre virus suspension. The method depends upon adsorption of the virus to barium sulphate suspension at 37° C and its subsequent elution at 4° C into sodium citrate solution. The efficiency of protein removal varied from 90 to 98.9%, corresponding to a fall from 1400 to 150 μg/ml, and from 9500 to 110 μg/ml with Coxsackie B 4 and mumps viruses, respectively. Other viruses, for which intermediate figures were obtained included several myxoviruses, vaccinia, herpes simplex and respiratory syncytial virus. Subsequent immunization of guinea-pigs with ‘protein-reduced’ virus suspensions confirmed the retention of viral immunogenicity after treatment in terms of complement-fixing, neutralizing and immunofluorescent antibody titres induced. All the treated viruses except respiratory syncytial virus produced an antiserum in guinea-pigs completely free from non-specific fluorescent when used without dilution in a homologous immunofluorescent test system.

    Notes: Summary An investigation of some of the characteristics of the VERO cell line (Cercopithecus aethiops kidney) is reported, in which the suitability of the cells for use in routine diagnostic virology was examined. VERO cells will: 1. grow to monolayers as rapidly as other heteroploid cell lines, but will maintain as usable monolayers in conventional maintenance medium for a significantly longer time; 2. grow a selection of ten representative viruses to a higher titre than will two other heteroploid lines or two cell strains; 3. support the growth of a very wide range of viruses with development in most instances of a characteristic cytopathic effect. VERO cells are recommended as possible substitutes for primary monkey kidney cell cultures, and are superior in overall performance to primary human amnion, HEp-2 or HeLa cells for diagnostic use in virology.

    Notes: Summary A method is described for the construction of five intersecting pools of guineapig antiserum to be used in the immunofluorescent identification of eight common respiratory viruses. The component antisera are highly purified and produce no non-specific fluorescence at working titre before or after incorporation into pools. The antiserum pools are fully stable at 4°C for up to 2 weeks, and for at least 3 months when stored at −20° C. Due to their intersecting nature, a direct “read-out” of the type of virus present is possible. The pool system also offers the advantage of built-in negative controls for each specimen examined.
